A method for the determination of human immunoglobulin G(IgG) based on a colloidal gold label by fluorospectrophotometry was developed. The sandwich immunoreaction among goat-anti-human IgG, human IgG and goat-anti-human IgG labeled with colloidal gold nanoparticles was applied in this experiment. First, a sandwich immunocomplex was formed on the surface of 96 well clear polystyrene high bind stripwellTM microplate. After the formation of the sandwich immunocomplex, a solution was added to dissociate the immunocomplex at room temperature. Then the solution of each well was transferred into the corresponding test tube. Thirdly, the rhodamine B solution was injected into each test tube. The rhodamine B chloraurate was extracted into ether that was measured with a spectrofluorometer. The experimental results indicate that the fluorescence intensity increased with the increase of human IgG concentration. The fluorescence intensity of rhodamine B chloraurate at 570 nm was proportional to the logarithm of human IgG concentration in a range from 10 to 5×105 ng/mL. It was shown that the determination of human IgG was easily made with the proposed fluorospectrophotometry.  相似文献

A sensitive competitive flow injection chemiluminescence (CL-FIA) immunoassay for immunoglobulin G (IgG) was developed using gold nanoparticle as CL label. In the configuration, anti-IgG antibody was immobilized on a glass capillary column surface by 3-(aminopropyl)-triethoxysilane and glutaraldehyde to form immunoaffinity column. Analyte IgG and gold nanoparticle labeled IgG were passed through the immunoaffinity column mounted in a flow system and competed for the surface-confined anti-IgG antibody. CL emission was generated from the reaction between luminol and hydrogen peroxide in the presence of Au (III), generated from chemically oxidative dissolution of gold nanoparticle by an injection of 0.10 mol L−1 HCl–0.10 mol L−1 NaCl solution containing 0.10 mmol L−1 Br2. The concentration of analyte IgG was inversely related to the amount of bound gold nanoparticle labeled IgG and the CL intensity was linear with the concentration of analyte IgG from 1.0 ng mL−1 to 40 ng mL−1 with a detection limit of 5.2 × 10−10 g mL−1. The whole assay time including the injections and washing steps was only 30 min for one sample, which was competitive with CL immunoassays based on a gold nanoparticle label and magnetic separation. This work demonstrates that the CL immunoassay incorporation of nanoparticle label and flow injection is promising for clinical assay with sensitivity and high-speed.  相似文献

Both discrete and distributed delays are considered in a two‐neuron system. We analyze the influence of interaction coefficient and time delay on the Hopf‐pitchfork bifurcation. First, we obtain the codimension‐2 unfolding with original parameters for Hopf‐pitchfork bifurcation by using the center manifold reduction and the normal form method. Next, through analyzing the unfolding structure, we give complete bifurcation diagrams and phase portraits, in which multistability and other dynamical behaviors of the original system are found, such as a stable periodic orbit, the coexistence of two stable nontrivial equilibria, and the coexistence of a stable periodic orbit and two stable equilibria. In addition, the obtained theoretical results are verified by numerical simulations. Finally, we perform the comparisons of the obtained results of Hopf‐pitchfork bifurcation with other Hopf‐fold bifurcation results in some biological neural systems and give the obtained mathematical results corresponding to the physical states of neurons. This paper is concerned with the optimal distributed control of the viscous weakly dispersive Degasperis–Procesi equation in nonlinear shallow water dynamics. It is well known that the Pontryagin maximum principle, which unifies calculus of variations and control theory of ordinary differential equations, sets up the theoretical basis of the modern optimal control theory along with the Bellman dynamic programming principle. In this paper, we commit ourselves to infinite dimensional generalizations of the maximum principle and aim at the optimal control theory of partial differential equations. In contrast to the finite dimensional setting, the maximum principle for the infinite dimensional system does not generally hold as a necessary condition for optimal control. By the Dubovitskii and Milyutin functional analytical approach, we prove the Pontryagin maximum principle of the controlled viscous weakly dispersive Degasperis–Procesi equation. The necessary optimality condition is established for the problem in fixed final horizon case. Finally, a remark on how to utilize the obtained results is also made. This paper is dedicated to the Oldroyd‐B model with fractional dissipation (?Δ)ατ for any α > 0. We establish the global smooth solutions to the Oldroyd‐B model in the corotational case with arbitrarily small fractional powers of the Laplacian in two spatial dimensions. Moreover, in the Appendix, we provide some a priori estimates to the Oldroyd‐B model in the critical case, which may be useful and of interest for future improvement. We give some integrability conditions for the coefficients of a sequence of elliptic systems with varying coefficients in order to obtain the stability for homogenization. In the case of equations, it is well known that equi‐integrability and bound in L1 are enough for this purpose; however, this is based on the maximum principle, and then, it does not work for systems. Here, we use an extension of the Murat–Tartar div‐curl lemma because of M. Briane, J. Casado‐Díaz, and F. Murat in order to obtain the stability by homogenization for systems uniformly elliptic, with bounded coefficients in , with N the dimension of the space. The aim of this paper is to propose optimal strategies for dengue reduction and prevention in Cali, Colombia. For this purpose, we consider two variants of a simple dengue transmission model, epidemic and endemic, each of which is amended with two control variables. These variables express feasible control actions to be taken by an external decision‐maker. First control variable stands for the insecticide spraying and thus targets to suppress the vector population. The second one expresses the protective measures (such as use of repellents, mosquito nets, and insecticide‐treated clothes) that are destined to reduce the number of contacts (bites) between female mosquitoes (principal dengue transmitters) and human individuals. We use the Pontryagin's maximum principle in order to derive the optimal strategies for dengue control and then perform the cost‐effectiveness analysis of these strategies in order to choose the most sustainable one in terms of cost–benefit relationship.
