Summary Molecular surface areas accessible to a 4 ? diameter spherical probe were calculated from crystallographic data for five proteins: α-chymotrypsinogen A, lysozyme, trypsinogen, ribonuclease A and ribonuclease S. The retention factors of various proteins were measured on stationary phases having polyether- and phenylligates and with aqueous eluents containing (NH4)2SO4, Na2SO4 or NaCl at pH 7.0. The logarithmic retention factors were plotted against the salt molality and the hydrophobic interaction parameters evaluated from the limiting slopes of the plots at high salt concentrations for the proteins in the chromatographic systems investigated. The hydrophobic interaction parameters thus obtained were linear in both the molecular surface areas of the proteins and the molal surface tension increments of the salts. The experimental results obtained with these relatively simple proteins of known molecular structure, which were available in high purity, support earlier theoretical predictions for the dependence of the hydrophobic interaction parameter on the surface area of the protein and the surface tension raising effect of the salt.  相似文献

Summary A sequence for the fractionation of the amylasic components from a malted barley extract is proposed using two salt-promoted, adsorption processes: thiophilic interaction chromatography (TIC) and hydrophobic interaction chromatography (HIC).Two fractions containing -amylase activity were recovered during the thiophilic chromatography; the first was resolved in to -amylase I and -amylase I by HIC on a phenyl-sepharose column; an enrichment factor of 32 was achieved for -amylase I. The other amylasic component eluted from the thiophilic gel was characterized as -amylase II. Although the adsorption of malt amylases on phenylsepharose and the thiophilic adsorbent is salt promoted, the interactions involved in each case are clearly distinguished by the different behaviour and disparate salt effects.  相似文献

In order to clarify some aspects of the hydrophobic interactions, the enthalpies of dilution of monoethylurea, 1,3-dimethylurea, and 1,3-diethylurea have been determined calorimetrically at 25°C. The calorimetric data, expressed in terms of excess enthalpy, permit the evaluation of the pair and triplet interaction coefficients. The analyses of these and of the analogous coefficientsg xx andg xxx, derived from osmotic data, indicate a driving force favorable to the interactions among the hydrated solute molecules. Nevertheless, the positive values of theh xx andh xxx coefficients seem to suggest that the source of the effect is a rearrangement of the water molecules rather than a direct association of the solute molecules. There are evidences of a strict correlation between the enthalpic and the entropic effects. Preliminary data were presented at the International Conferences on Chemical Thermodynamics at Baden (1973) and Montpellier (1975). 从TGEV 3CL蛋白酶二聚体结构出发,研究了TGEV 3CL蛋白酶二聚体单体之间的静电和疏水相互作用.蛋白质的静电相互作用通过有限差分方法求解Poisson-Boltzmann方程得到,疏水相互作用通过分析溶剂可及性表面模型得到.考察了不同pH值对TGEV 3CL蛋白酶二聚体静电和疏水相互作用的影响,在pH值为5.5~8.5时,二聚体静电相互作用能、静电去溶剂化能和疏水自由能都较小,表明在该条件下静电和疏水相互作用有利于二聚体的稳定存在,这符合实验结晶所需条件.pH值对静电去溶剂化能的影响大于疏水自由能,表明静电作用是造成强酸或强碱条件下二聚体不能稳定存在的主要原因.  相似文献

A new family of hydrophobically modified polyacrylamides was synthesized via copolymerization of acrylamide (AM) and anionic surface-active monomer of acrylamide-type, sodium 2-acrylamido-tetradecane sulfonate (NaAMC14S), in aqueous solution. In the copolymerization, by varying various factors, such as the feed ratio of NaAMC14S to AM and the amount of added electrolyte NaCl and initiator, we prepared copolymers NaAMC14S/AM with different block structures. The relationship between structures and hydrophobic association properties of copolymer chains was studied by using fluorescence probe and viscosimetry. Effects of the content and length of the hydrophobic blocks and the total molecular weight on hydrophobic association of the copolymers in pure water and in brine solution were examined, respectively. The results show that in pure water, hydrophobic association of the copolymers was enhanced as the content and length of the hydrophobic block increase. On the other hands, for a given content and length of the hydrophobic block, the hydrophobic association of the copolymers was enhanced as the total molecular weight increases. For all the copolymers studied, the apparent viscosity of their solutions in pure water has a limited value, but the apparent viscosities of the copolymer brine solutions are much higher than that of their corresponding water solutions, and show strong positive salinity sensitivity. Similarly, the hydrophobic association of the copolymer in brine solutions was enhanced as the content and length of the hydrophobic block increase.  相似文献

Monodisperse, cationic polymer particles bearing quaternary ammonium groups effectively self-organized on hydrophobic solid substrates such as alkylated glass plates and polymer films to form particle monolayers. With an increase of the particle surface charge density, the surface coverage decreased and the morphology of particle monolayers changed from aggregated type to dispersed type. The dispersed type of particle monolayers having a relatively regular particle distance was formed at higher temperature. The self-organization behaviors on alkylated glass plates were different from those on unmodified glass plates through electrostatic interaction. The formation of particle monolayers on alkylated glass plates occurred only over a certain latex concentration range in contrast with that on unmodified glass plate. The adhesive strength of particle monolayers was enhanced by annealing at temperatures above the glass transition temperature (T g) of the particles. Lens-shaped particle monolayers were fabricated by annealing the dispersed type of particle monolayers.  相似文献

With the aim of optimizing the chromatographic process by avoiding any preliminary derivatizing step, we examined the chromatographic behaviour of a selected set of unconjugated bile acids looking at the dominant factors that affect the performances of three different stationary phases: RP-8, RP-18 and RP-18 Base Deactivated (RP-18-BD). Accordingly to its structural peculiarity, the RP-18-BD column combined with a specific mobile phase has proved to be the most suitable one, in enhancing both separation factor α and resolution R S within the selected set of analytes. To avoid the limitation of the widely used prediction methods of soil organic carbon partition coefficients (KOC) from hydrophobic parameters, e.g., the n-octanol/water partition coefficients (KOW) and the reversed phase high performance liquid chromatographic (RP-HPLC) retention factors, the soil column liquid chromatographic (SCLC) method was developed for KOC prediction. The real soils were used as the packing materials of RP-HPLC columns, and the correlations between the retention factors of organic compounds on soil columns (ksoil) and KOC measured by batch equilibrium method were studied. Good correlations were achieved between ksoil and KOC for three types of soils with different properties. All the square of the correlation coefficients (R2) of the linear regression between log ksoil and log KOC were higher than 0.89 with standard deviations of less than 0.21. In addition, the prediction of KOC from KOW and the RP-HPLC retention factors on cyanopropyl (CN) stationary phase (kCN) was comparatively evaluated for the three types of soils. The results show that the prediction of KOC from kCN and KOW is only applicable to some specific types of soils. The results obtained in the present study proved that the SCLC method is appropriate for the KOC prediction for different types of soils, however the applicability of using hydrophobic parameters to predict KOC largely depends on the properties of soil concerned.  相似文献

The effect of pH on the interfacial adsorption activity of pulmonary surfactant was examined. Measurements of the surface tension were made in a Wilhelmy-like surface microbalance specially designed to assay small volumes of hypophase in thermostatically controlled conditions. Alkaline pH caused a significant decrease of the surface activity of both pulmonary surfactant and a lipid extract from surfactant (LES) (containing all of the lipids and surfactant protein-B (SP-B) and surfactant protein-C (SP-C) hydrophobic surfactant proteins, but lacking surfactant protein-A). The pK calculated from the change of surface activity versus pH was 9.18±0.26 and 9.27±0.31 for pulmonary surfactant and LES, respectively. The results from this study support the idea that electrostatic interactions between basic residues of SP-B and SP-C and negatively charged surfactant phospholipids could be important for the interfacial adsorption activity of pulmonary surfactant.  相似文献
