X=Y=ZH Systems as potential 1,3-dipoles. Part 58: Cycloaddition route to chiral conformationally constrained (R)-pro-(S)-pro peptidomimetics

Imines of (1S,9S)-t-butyl-9-amino-octahydro-6,10-dioxo-6H-pyridazino[1,2-a][1,2]diazepine-1-carboxylate undergo thermal (toluene, 110°C) or LiBr-DBU catalysed (MeCN, room temperature) regio- and stereo-specific cycloaddition to a range of chiral dipolarophiles giving enantiopure spiro-cycloadducts in excellent yield. The reactions proceed via intermediate NH azomethine ylides and litho azomethine ylides, respectively and results in the multiplication of chiral centres from 2 (one of which is lost in the process) to 5.     

Synthesis of cis bis-β-lactams via Staudinger cycloaddition reaction using C2-symmetric 1,2-diamines

An efficient stereoselective synthesis of bis-β-lactams via cycloaddition reaction (Staudinger reaction) of ketenes with bisimines derived from C₂-symmetric 1, 2-diamines is described. The reaction provided diastereomeric mixture of meso and C₂-symmetric cis-bis-β-lactams with higher selectivity for meso-bis-β-lactams.     

Preparation and stereoselectivity of 1,3-dipolar cycloaddition ofD-glucose-derived nitrones to N-arylmaleimides

Summary Nitrones2 derived fromD-glucose oxime and benzaldehydes without employing any protection of hydroxyl group were isolated in pure state. The 1,3-dipolar cycloaddition of2 to N-arylmaleimides gave predominantly theanti isoxazolidines3 and was rationalized byZ/E isomerization of N-glycosylnitrones2. The structure and steric configuration of the products have been assigned on the basis of¹H- and¹³C-NMR spectroscopy. AM1 calculations of the nitrones and MM2 calculations of the adducts were performed.

Darstellung und Stereoselektivität der 1,3-dipolaren Cycloaddition vonD-Glucose-abgeleiteten Nitronen an N-Arylmaleimiden

Zusammenfassung Die Nitrone2 wurden ausD-Glucoseoxim und Benzaldehyden ohne Schutz von Hydroxylgruppen in reinem Zustand erhalten. Die 1,3-dipolare Cycloaddition von2 an N-Arylmaleimiden ergab bevorzugt dieanti-Isoxazolidine3; dies wurde über eineZ/E-Isomerisierung der N-Glycosylnitrone2 rationalisiert. Struktur und Stereochemie wurden auf Basis von¹H- und¹³C-NMR-Spektroskopie ermittelt. Außerdem wurden AM1-Berechnungen an den Nitronen und MM2-Rechnungen an den Addukten ausgeführt.

     

Photochemistry of β-(4-sydnonyl)-o-divinylbenzene: competitive cis-trans isomerization and photolysis

New cis- and trans-3-aryl-4-[2-(2-vinylphenyl)ethenyl]sydnones 2, aryl = phenyl, p-tolyl were prepared and transformed on irradiation in the presence of acrolein to a pyrazoline derivative that aromatized during isolation to trans-1-tolyl-3-[2-(2-vinylphenyl)ethenyl]pyrazole 7 and trans-5-formyl-1-tolyl-3-[2-(2-vinylphenyl)ethenyl]pyrazole 8.     

Stereocontrolled synthesis of (+)- and (−)-iridomyrmecin from citronellene enantiomers

Natural (+)-iridomyrmecin and its unnatural enantiomer (−)-iridomyrmecin were synthesized by intramolecular [3+2] dipolar cycloaddition of silyl nitronates that had been generated from the nitro derivatives of (+)- and (−)-citronellenes, respectively. 新型螺环六氢山酮素-氧化吲哚-苯并呋喃酮类拼接物的合成及其抗白血病细胞增殖活性

以色酮-氧化吲哚合成子与3-烯基苯并呋喃酮为原料,在催化剂1,8-二氮杂二环十一碳-7-烯作用下,在二氯甲烷中发生Michael/Michael加成关环反应,合成了6个未见文献报道的螺环六氢山酮素-氧化吲哚-苯并呋喃酮类拼接物(3a^3f),产率75%~87%,dr值3/1~5/1,其结构经1H NMR,13C NMR和HR-MS(ESI-TOF)表征。采用MTT法研究了3a^3f对人白血病细胞(K562)的体外抗增殖活性。结果表明:化合物3c,3e和3f对K562增殖有一定的抑制活性。

Synthesis and photochemistry of beta,beta'-di(2-furyl)-substituted o-divinylbenzenes: intra- and/or intermolecular cycloaddition as an effect of annelation

New heteroaryl-substituted o-divinylbenzenes, 2,2'-(1,2-phenylenedivinylene)difuran (9), 2,2'-(1,2-phenylenedivinylene)bisbenzo[b]furan (10), and 2,2'-(1,2-phenylenedivinylene)bisnaphtho[2,1-b]furan (11), were prepared and irradiated at various concentrations; intramolecular photocycloaddition and intermolecular [2+2] twofold photoaddition reactions took place to give bicyclo[3.2.1]octadiene derivatives 12-14 and cyclophane derivatives 15-17, respectively. Compound 11 was the most selective of these o-divinylbenzenes, which, owing to pi-pi intra- or intermolecular complexation, gave only the exo-bicyclo[3.2.1]octadiene derivative 14 at low concentrations, and only the cyclophane derivative 17 at high concentrations.     

Synthesis of dihydropyrazolo[3′,4′:3,4]pyrrolo[1,2-a]indoles and spiro-[3H-indole-3,3′-[Δ-1,2,4]-triazolin]-2-ones via intra and intermolecular 1,3-dipolar cycloadditions

A new class of dihydropyrazolo[3′,4′:3,4]pyrrolo[1,2-a]indoles and spiro[3H-indole-3,3′-[Δ²-1,2,4]-triazoline]-2-ones were synthesised via intra and intermolecular 1,3-dipolar cycloaddition reactions in good yields.     

1,3-Dipolar cycloaddition of azides with electron-deficient alkynes under mild condition in water

We report a simple synthetic protocol for the 1,3-dipolar cycloaddition of azides with electron-deficient alkynes. Alkyne with at least one neighboring electron-withdrawing group proceeds with the cycloaddition successfully without any catalysts at room temperature in water. Under this simple condition, we evaluated a series of small molecule model reactions and then coupled an azido-DNA molecule with electron-deficient alkynes for the formation of [1,2,3]-triazole heterocycle, providing a potential method for introducing functional groups to DNA under biological conditions.
