近红外反射光谱法(NIRS)测定棉仁粉中蛋白质和棉酚含量的研究 总被引:2,自引:0,他引:2
选用49份不同蛋白质和棉酚含量的陆地棉种质资源和188份陆地棉重组近交系为材料,以多年份、多地点种植收获的种子材料组成原始样品集,分别对棉仁粉中蛋白质含量和棉酚含量进行化学测定,采用改进的偏最小二乘法(Modified PLS)和(2,4,4,1)的数学转换方法建立近红外反射光谱(NIRS)定标模型,以寻找棉籽蛋白质含量和棉酚含量的快速测定方法。结果表明,蛋白质含量的定标决定系数(RSQ=0.933)和交叉检验决定系数(1-VR=0.929)较高,定标标准误差(SEC=0.623)和交互校验标准误差(SECV=0.638)较小,预测模型的建模效果较好,可替代化学分析。棉酚含量预测模型的RSQ,1-VR,SEC和SECV分别为0.836,0.811,0.074和0.079,模型预测效果略差于蛋白质模型,但仍可用于棉仁粉中棉酚含量的测定。 相似文献

Gossypol was obtained as an yellow platelike crystal with m.p. 210-214 . In CDCl3 there were three tautomers of gossypol: Ⅰ aldehyde, Ⅱ lactol, Ⅲ ketal, in equilibrium .Their total 1H NMR spectra were assigned by means of 1D and 2D NMR techniques including 1H-1H cosy ,DEPT, HMQC (1H Detected Heteronuclear Multiple Quantum Coherence) and HMBC (1H Detected Heteronuclear Multiple Bond Connectivity) experiments.This paper first reported that we took use of the 2D NMR techniques to assign all of 1H NMR chemical shifts of each tautomer , through the assignments of each peaks we investigated the tautomerism of gossypol . We concluded that when gossypol ( Ⅰ ) was put into CDCl3 , it would tautomerized three tautomers, they stable existed and attained tautomeric equilibrium in a molar ratio of 6:2:1 according to peaks intensity ratios in CDCl3. Conditions are reported for the reverse-phase HPLC analysis of gossypol and of some of its derivatives, including anhydrogossypol, gossypolone, gossypol hexaacetate, and Schiff's base derivatives. It is shown that field desorption (FD) and chemical ionization (CI) mass spectrometry have advantages over electron impact (EI) mass spectrometry for the characterization of these compounds and that combined high-performance liquid chromatography/mass spectrometry (LC/MS), especially when used in the CI mode, is particularly effective. 相似文献

Gossypol and its 1: 1 complexes with copper acetate in acetonitrile were studied by FTIR and UV-visible spectroscopy. With the complexation with Cu cation the tautomeric equilibrium of gossypol is completely shifted from the aldehyde-aldehyde to the lactol-lactol tautomer. The acidic character of O11H group in the gossypol-Cu complex led to the selective esterification of the lactol form of gossypol by the acetate group. 相似文献

Summary An ion-pair, liquid chromatographic method for the determination of total gossypol in chicken liver has been developed. It involves hydrolysis of bound gossypol in the presence of ascorbic acid, partitioning into chloroform and isocratic separation on Spherisorb ODS2, 10 m, column with a methanol-water mobile phase containing octane-1-sulfonate. Precision data based on within-day and between-days variation suggested an overall relative standard deviation of 8.1%, while overall recovery was 81.1%±3. 相似文献

This study was completed to determine effects of electron beam (EB) irradiation at doses of 15, 30 and 45 kGy on anti-nutritional factors, ruminal degradation and in vitro crude protein (CP) digestibility of whole cottonseed (WCS), soybean (SB) and canola seeds (CS). EB-irradiation eliminated completely (P<0.001) phytic acid of WCS, SB and CS at a dose of 30 kGy. EB-irradiation decreased linearly (P<0.001) the total glucosinolate content of CS. Trypsin inhibitor activity of 15, 30 and 45 kGy EB-irradiated SB was decreased by 19, 73 and 88%, respectively. Free gossypol content of WCS was reduced linearly (P<0.001) by irradiation. EB-irradiation increased linearly (P<0.001) CP digestibility of feeds. In conclusion, EB-irradiation was an effective processing method for improving the nutritive value of WCS, SB and CS. 相似文献

Crystals of the Schiff base derivative of gossypol with 3,6,9-trioxa-decylamine were examined using X-ray diffraction, FT-IR and CPMAS spectroscopy. The Schiff base crystallizes as a racemate in the space group C2/c with a=24.390(5), b=12.026(2), c=14.810(3) Å, β=102.78(3)°, and Z=4. The results of the FT-IR, and CPMAS study of the crystals are in agreement with the X-ray data. The FT-IR spectrum of the crystals shows that the OH groups at position 1,1′ and 6,6′ as well as the N16-H proton are involved in weak intermolecular and intramolecular hydrogen bonds, respectively. The FTIR and CP-MAS spectral behaviour is in agreement with the crystallographic results demonstrating the existence of the enamine-enamine tautomeric form of the Schiff base studied. 相似文献

Identification and quantification of gossypol in cotton by using packed micro-tips columns in combination with HPLC 总被引:1,自引:0,他引:1
Self-packed micro-tip columns containing a C18-bonded silica stationary phase, based on the same principles as solid-phase extraction methods, were used to obtain gossypol and related sesquiterpenoid aldehyde-enriched fractions. The enriched metabolite fractions were then analyzed by optimized high-performance liquid chromatography (HPLC) with a C18 column (4.6 mm×25 cm) eluted with the binary mobile phase acetonitrile–0.1% aqueous TFA solution (80:20). This method has proven to be highly reproducible. The precision and accuracy, as %RSD and %RME values, were determined to be less than 15% for the method. The minimum detection limit of gossypol was determined to be 10 ng (absolute gossypol). Absolute recovery was greater than 94% with a standard deviation of ±3.68%. This is a simple, fast, and cost-effective method for isolation, identification, and quantification of gossypol and related secondary metabolites. Comparative analysis of gossypol content was performed on different parts of the cotton plant (seeds, stems and leaves) of two different cultivars of Gossypium hirsutum L. (Acala1517–70 and OR19). The results indicate that the OR19 cv naturally contains higher gossypol levels than the Acala cv. It was also found that treatment of leaves with a Verticillium dahliae-derived elicitor induced production of deoxyhemigossypol rather than gossypol. 相似文献
