Stereoselective Synthesis of the Hexasaccharide Repeating Unit of the Cell Wall Polysaccharide from Kineosporia aurantiaca VKM Ac-720T Employing the Direct Glycosylation with Anomeric Hydroxy Sugars Involving Glycosyl Phthalate Intermediates

Synthesis of a suitably protected form of the hexasaccharide repeating unit of the cell wall polymer from Kineosporia aurantiaca VKM Ac-720 ^T has been achieved by the stereoselective direct glycosylation of a trisaccharide acceptor with a trisaccharide donor having an anomeric hydroxy group involving a glycosyl phthalate intermediate. Both the trisaccharide acceptor and the trisaccharide donor were obtained from a common trisaccharide, of which two β-mannopyranosyl linkages were constructed stereoselectively by employing the direct glycosylation method with the anomeric hydroxy sugar involving a glycosyl phthalate intermediate and the 2′-carboxybenzyl glycoside method, respectively.     

NaBH3CN and other systems as substitutes of tin and silicon hydrides in the light or heat-initiated reduction of halosugars: a tunable access to either 2-deoxy sugars or 1,5-anhydro-itols

UV light-promoted reduction of acetobromoglucose by NaBH₃CN in t-BuOH afforded 1,3,4,6-tetra-O-acetyl-2-deoxy-α-d-arabino-hexopyranose in high yield and purity, via a Surzur–Tanner rearrangement, while, with 10 mol % thiophenol added, acetylated 1,5-anhydro-d-glucitol was cleanly obtained. Such tin-free and mild reductions, presumed to proceed via radical pathways, were more efficient with NaBH₃CN compared to NaBH₄ or NaBD₄, and do not occur with acetochloroglucose. Similar reductions to 1,3,4,6-tetra-O-acetyl-2-deoxy-α-d-arabino-hexopyranose were achieved upon heating to 80 °C t-BuOH or CH₃CN solutions of NaBH₃CN and AIBN, but with a lower selectivity due to competing ionic reactions. With other pyranosyl bromides, reductions by NaBH₃CN could be tuned similarly (d-galacto), but some (d-manno, 5-thio-d-xylo) gave mainly or exclusively 1,5-anhydro-itols. Other conditions, or reagents promoting SET process, afforded also reduced products, but with lower rates or selectivities. Primary iodides were reduced readily with NaBH₃CN under UV light.

Glycosyl azides have been prepared from a range of readily available glycosyl halides by a solvent‐free mechanochemical procedure employing a planetary ball mill in good to excellent yields.     

Efficient Synthesis of Flaccidoside II,a Bioactive Component of Chinese Folk Medicine Di Wu

A bidesmosidic triterpene saponin, flaccidoside II, which was isolated from Di Wu, a Chinese folk medicine from dry rhizome of Anemone flaccida Fr. Schmidt, was efficiently synthesized in a convergent approach. We employed two glycosyl trichloroacetimidate donors in a one-pot reaction as a key step.

Glycosylations with a septanosyl fluoride donor lacking a C2 protecting group

Septanosyl fluorides, prepared from protected pyranoses, were used as donors in glycosylation reactions. The fluorides were synthesized by the addition of vinyl Grignard to the pyranoses followed by ozonolysis and then DAST-mediated fluorination. Activation in the presence of nucleophiles then provided the product glycosides. High α-stereoselectivity was observed for glycosylations using a donor that had a free C2 hydroxyl group; a model where the hydroxyl group participates to guide the stereochemical outcome is proposed.     

固相萃取-气相色谱/质谱法同时测定化妆品中的邻苯二甲酸酯和对羟基苯甲酸酯

采用超声协助甲醇提取、固相萃取净化、气相色谱/选择离子质谱联用法,同时测定化妆品中8种邻苯二甲酸酯和4种对羟基苯甲酸酯。该方法线性范围广、重现性好、快速简便、干扰小。样品的加标回收率为80%～100%;含量检测的相对标准偏差小于10%;方法的检出限为0.1~5.0 μg/kg。用该方法对15种实际样品中的12种残留物进行定量检测,结果表明除了一种样品中不含待测物外,其余样品均检测到3~7种待测物。其中以对羟基苯甲酸甲酯、对羟基苯甲酸丙酯、邻苯二甲酸丙酯、邻苯二甲酸环己酯和邻苯二甲酸乙基庚基酯为主。     

基质固相分散-液相色谱-质谱法测定蔬菜中的邻苯二甲酸酯

采用基质固相分散-液相色谱-质谱法测定保护地蔬菜中邻苯二甲酸酯(PAEs)含量,并分析了蔬菜中PAEs污染状况,研究了水和洗涤液浸泡对蔬菜中PAEs的消除效果。蔬菜样品经弗罗里硅土和石墨化碳黑研磨均匀后,用乙酸乙酯淋洗净化,再用液相色谱/电喷雾质谱法测定。PAEs的添加回收率为82.7%~105.7%;RSD为1.7%~6.1%;检出限为邻苯二甲酸二甲酯(DMP):0.99ng;邻苯二甲酸二乙酯(DEP):0.75ng;邻苯二甲酸二丁酯(DBP):0.70ng;邻苯二甲酸二异辛酯(DEHP):1.9ng。本方法前处理简单,具有较高的准确度和灵敏度。黄瓜、番茄、西葫芦中4种PAEs总量分别为2.02、1.26、0.91mg/kg;洗涤实验结果表明,水和洗涤液浸泡可显著降低蔬菜中的PAEs含量。     

Hetero Diels-Alder reactions of acyl phosphonates: synthesis of glycosyl type phosphonates

We have prepared glycosyl type phosphonates via hetero Diels-Alder (HDA) reactions of acyl phosphonates with electron rich dienes. HDA reactions of acyl phosphonates with Danishefsky’s diene required thermal activation to yield the desired dihydropyranones in good yield (70-91%). The reactions with Brassard’s diene involved Lewis acid promotion to yield the corresponding lactones, though in moderate yield (33-69%).     

Stereoselective synthesis of β-glycosyl esters of cis-cinnamic acid and its derivatives using unprotected glycosyl donors

The β-glycosyl esters of cis-cinnamic acid were synthesized directly using Hannesian’s unprotected glycosyl donor and the carboxylic acid in toluene. This protocol does not require protecting groups on the glycosyl donors, and high stereoselectivity was achieved. The first synthesis of a potent allelochemical, 1-O-cis-cinnamoyl-β-d-glucopyranose, is also described.
