Synthesis of New Pyrimidine,Quinazoline and Diazatricyclo Derivatives by Multicomponent Reaction and Evaluation of Their Biological Activity

Abstract

A series of new pyrimidine and quinazoline derivatives was synthesized by a Biginelli-like reaction of urea/thiourea, aldehyde, and ketone in the presence of hydrochloric acid as a catalyst. In a similar way, some novel diazatricyclo derivatives were obtained via a Biginelli-like reaction followed by an intramolecular Michael-type addition. The yields of products were reasonable after recrystallization from ethanol. All newly synthesized compounds were characterized using IR and NMR (¹H and ¹³C) spectroscopy and elemental analysis. The antibacterial activity of these compounds was investigated against Staphylococcus aureus (RTCC, 1885), and Escherichia Coli (ATCC, 35922).

Substituted Quinolinones. Part 19. New and Unexpected Results from Oxidation of 3-Acetyl-1-Alkyl- 4-Hydroxyquinolin-2(1H)-Ones Using Selenium Dioxide

Abstract

Oxidation of 3-acetyl-1-alkyl-4-hydroxyquinolin-2(1H)-ones using selenium dioxide under Riley conditions was described. The oxidation reaction produced a mixture of 2 unexpected α-keto acid and its dehydrated dimer derivatives. The oxidation reaction was studied under different reaction conditions in order to maximize the yields and optimize reaction conditions. Also, 1-alkyl-4-hydroxy-3-(2-nitroacetyl)quinolin-2(1H)-one and/or 3-acetyl-1-alkyl-4-diflouro-boryloxyquinolin-2(1H)-one derivatives were subjected to the same oxidation reaction giving rise improved reaction yields and selectivity in case of the boron-complex. Alkaline degradation of the dehydrated dimers led to formation of the 4-hydroxy-2-oxoquinoline-3-carboxylic acids while under the same conditions the α-keto acids underwent deoxalylation.

Total synthesis of pleurolactone

The stereoselective total synthesis of pleurolactone has been accomplished in 7 steps. The key synthetic features were the construction of four contiguous stereocenters using an endo-selective Diels-Alder reaction and high diastereoselective dihydroxylation.     

Silica iodide catalyzed,ultrasound-promoted,one-pot four-component synthesis of novel 1,4,5,6-tetrahydropyridine-3-carboxylate derivatives

Silica iodide catalyzed, competent synthesis of 15 novel substituted-1,4,5,6-tetrahydropyridine-3-carboxylate derivatives was achieved by a one-pot four-component cyclocondensation of Meldrum's acid, aryl aldehydes, aromatic amines and ethyl acetoacetate under ultrasonic irradiation. In comparison to the conventional methods, our protocol is convenient, eco-friendly and offers several benefits such as: mild conditions, high yields, shorter reaction time and ease in product isolation.     

Synthesis of the Novel Chiral Catalysts by Click Chemistry and Their Application

Novel cinchonine ammonium salt derivatives have been prepared by 1,3‐dipolar cycloaddition. Their chiral catalytic efficacy was investigated in the asymmetric alkylation of N‐diphenylmethyleneglycine t‐butyl ester in the water phase. As the special structure of the catalyst, its asymmetric alkylation catalytic behavior both in organic solvents and in water is satisfactory, which is environmentally friendly.     

Novel Synthesis of some Spiro Heterocycles Derived from 3-Hydroxy-3(2-Oxocyclohexyl)-Indolin-2-One

The reaction of 3-hydroxy-3(2-oxocyclohexyl)indolin-2-one 1 with some active methylene, bidentates, phenylisocyanate, phenylisothiocyanate, carbon disulfide, benzaldehyde, ketoketene S,S-acetal, cyanoketene S,S-acetal, active halo compounds or benzylidenemalononitrile affords a series of new spiro heterocyclic systems.
