地黄滋阴补血填髓,广泛用于贫血和脑疾病患者,但其功效机制尚未阐明.本文探讨地黄水提物(Rehmannia glutinosa’s water extracts,RGWE)改善血虚大鼠记忆及其滋阴补血填髓的可能机制.采用断尾放血结合注射环磷酰胺制备贫血大鼠模型,随机分溶媒组和地黄水提物3,6,10g/kg治疗组,灌服等体积自来水或不同剂量地黄水提物10天,采用Morris水迷宫观测大鼠空间记忆能力,采用酶联免疫吸附测定方法(Enzyme linked immunosorbent assay,ELISA)检测血浆红细胞生成素(EPO)水平,免疫组化和免疫蛋白印迹技术(Western blotting)检测分析脑EPO及其受体表达水平.成功制备血虚模型,表现为大鼠红细胞数和血红蛋白显著下降,与造模前比较差异显著(P<0.05),溶媒组空间记忆能力显著下降,地黄治疗组空间记忆能力明显提高,逃避潜伏期明显缩短(P<0.05),一定时间内穿越平台次数显著增多(P<0.05);免疫组化和Western blotting结果显示脑组织EPO及其受体表达、血浆EPO水平均较溶媒组明显升高(P<0.05).地黄水提物显著提高贫血... 相似文献

In view of existing contradictory assignments of the symmetrical stretching vibrations associated with the formal C-C and C-F bonds of trans/cis oxalyl fluoride, an additional theoretical analysis of the corresponding calculated wavenumbers was preformed on trans-C2O2F2 and cis-C2O2F2 based on previously calculated ab initio scaled force fields at the HF/6-31G computational level and new force fields calculated at the MP2/aug-cc-pVTZ level. This novel analysis included computational data from the isotopic shifts brought about by incorporating 13C and 14C atoms into the structure. A detailed examination of the calculated wavenumbers made it possible to validate the assignments of the ν2 and ν3 wavenumbers in the trans-C2O2F2 and cis-C2O2F2 molecules as the formal C-C bond stretching and the formal C-F bond symmetrical stretching vibrations, respectively. 相似文献

The membrane‐bound tumor‐associated glycoprotein MUC1 is aberrantly glycosylated in cancer cells compared with normal cells, and is therefore considered an attractive target for cancer immunotherapy. However, tumor‐associated glycopeptides from MUC1 do not elicit a sufficiently robust immune response. Therefore, antitumor vaccines were developed, which consist of MUC1 glycopeptides as the B epitopes and immune‐stimulating toll‐like receptor 2 (TLR 2) lipopeptide ligands. These fully synthetic vaccine candidates were prepared by solid‐phase synthesis of the MUC1 glycopeptides. The Pam3Cys lipopeptide, also synthesized on solid‐phase, was C‐terminally coupled to oligovalent lysine cores, which N‐terminally incorporate O‐propargyl oligoethylene glycol acyl side chains. The MUC1 glycopeptides and lipopeptide lysine constructs were then conjugated by click chemistry to give oligovalent synthetic vaccines. An enzyme-linked immunosorbent assay (ELISA) is reported for human immunoglobulin G based on synthesized hemin–MnO2 nanocomposite as the label. Enhanced sensitivity was obtained due to the increased catalytic activity of the hemin–MnO2 nanocomposite toward 3,3′,5,5′-tetramethylbenzidine compared to hemin and MnO2 alone. The synthesized hemin–MnO2 nanocomposite was characterized by transmission electron microscopy and its catalytic activity to 3,3′,5,5′-tetramethylbenzidine was investigated by ultraviolet–visible absorption spectroscopy. After assembly of the sandwich-type immunoassay in the 96 wells of the plate the hemin–MnO2-based label catalyzed 3,3′,5,5′-tetramethylbenzidine into blue compounds that were monitored by a plate reader. The absorbance increased with the concentration of human immunoglobulin G. The immunoassay displayed high sensitivity, a long linear dynamic range, and good selectivity for human immunoglobulin G. The immunoassay was also used for the determination of human immunoglobulin G in serum with favorable results. The developed assay combines the high throughput and low cost of ELISA with the simplicity of nanocomposite labeling and is suitable for application in clinical diagnosis. 相似文献

The aep-PNA is a chiral and cyclic PNA analogue, which has a stronger and base dependent binding affinity with complementary DNA. To understand the base dependent properties at monomer level, the structural studies of aep-PNA-(T/C/A) monomers have been carried out focussing on the conformational analysis of pyrrolidine ring pucker in aep-PNA by 1H NMR and the coupling constant data fitted into PSEUROT software. The results indicate that the type of pyrrolidine pucker depends on the electronic nature of substituent, implying the effect of pyrimidine or purine substituents in determining the ring pucker in monomers. This may consequently influence the aep-PNA oligomer conformation. Since pyrrolidine nucleic acids have emerged as an important class of PNA analogues, present results may have importance for their future development. 相似文献

Streptococcus pneumoniae LTA is a highly complex glycophospholipid that consists of nine carbohydrate residues: three glucose, two galactosamine and two 2‐acetamino‐4‐amino‐2,4,6‐trideoxygalactose (AATDgal) residues that are each differently linked, one ribitol and one diacylated glycerol (DAG) residue. Suitable building blocks for the glucose and the AATDgal residues were designed and their synthesis is described in this paper. These building blocks permitted the successful synthesis of the core structure Glcβ(1‐3)AATDgalβ(1‐3)Glcα(1‐O)DAG in a suitably protected form for further chain extension ( 1 b , 1 c ) and as unprotected glycolipid ( 1 a ) that was employed in biological studies. These studies revealed that 1 a as well as 1 lead to interleukin‐8 release, however not via TLR2 or TLR4 as receptor. 相似文献

Modified rhodamine 6G molecules (Rh-Al or Rh-Aln) with polymerizable double bonds had been copolymerized with 1,8-naphthalimide derivatives. In copolymers PRNAM, naphthalimide moieties are connected to rhodamine moieties at nitrogen atom of rhodamine moiety, and in copolymers PRNM naphthalimide moieties are connected to rhodamine moieties at ester group of rhodamine moiety. We report on their photostability in liquid solution and in solid film. The photodegradation kinetics of novel copolymers functionalized with laser dyes based on modified rhodamine 6G and 1,8-naphthalimide has been studied by UV-VIS absorption spectroscopy. The results show that the rates of the photofading reactions of these novel copolymers follow quasi-first-order. The photostability of the copolymers functionalized with laser dyes in solid films is better than that in solutions. The different type of the copolymer shows the different photostability. 相似文献
