反相高效液相色谱法半制备分离[T(4-MOP)PS4]卟啉的研究

本文研究了新水溶性５，１０，１５，２０－四（４－甲氧基－３－磺酸苯基）卟啉［Ｔ（４－ＭＯＰ）ＰＳ４］的反相高效液相色谱（ＨＰＬＣ）分离条件。采用ＳｈｉｍｐａｃｋＰＲＥＰ－ＯＤＳ半制备色谱柱，用含有１０ｍｍｏｌ／Ｌ四乙基碘化铵的乙腈－水（体积比２５：７５）为流动相，流速１８ｍＬ／ｍｉｎ，于４１８ｎｍ波长下检测。［Ｔ（４－ＭＯＰ）ＰＳ４］与合成中生成的杂质组分完全分离。经此制备的卟啉纯度高，已成功地应用于自来水样中微量钴、锌、铜离子的ＨＰＬＣ测定中。     

磷钨钼钒杂多蓝光度法测定钼的探索

本文研究了磷钨钼钒四元杂多酸及其还原产物杂多蓝的形成条件、光度性质、组成和在测定高速钢中高钼的应用。试验表明,磷钨钼钒杂多蓝光度法测定钼具有新颖性、高选择性、较高准确性和简便性,适于高速钢中>1％Mo的测定。     

Fluorescence detection and quantitation of recombinant proteins containing oligohistidine tag sequences directly in sodium dodecyl sulfate-polyacrylamide gels

Two fluorophore-nitrilotriacetic acid conjugates, Pro-Q Sapphire 365 and Pro-Q Sapphire 488 oligohistidine gel stains, have been developed for the fluorescence detection of fusion proteins containing oligohistidine tags directly in sodium dodecyl sulfate polyacrylamide gels, without the requirement for electroblotting, reporter enzymes or secondary detection reagents. Pro-Q Sapphire 365 oligohistidine gel stain exhibits bright-blue fluorescence (emission maximum = 450 nm) when illuminated with UV-A or UV-B light from a standard ultraviolet transilluminator. Pro-Q Sapphire 488 oligohistidine gel stain exhibits bright-green fluorescence (emission maximum = 515 nm) when illuminated with visible light from a laser-based gel scanner equipped with a 470 nm second-harmonic generation (SHG) or 488 nm argon-ion laser source. Typically, 25-65 ng of oligohistidine-tagged fusion protein in whole cell lysates is detectable using either stain. After documenting the fluorescence signal from the Pro-Q Sapphire dyes, gels may be post-stained with the red-fluorescent SYPRO Ruby protein gel stain in order to reveal the total protein pattern.     

Hydride generation atomic fluorescence spectrometric determination of ultratraces of selenium and tellurium in cow milk

A sensitive procedure has been developed for selenium and tellurium determination in milk by hydride generation atomic fluorescence spectrometry (HG-AFS) after microwave-assisted sample digestion. The method provides sensitivity values of 1591 and 997 fluorescence units ng⁻¹ ml⁻¹ with detection limits of 0.005 and 0.015 ng ml⁻¹ for Se and Te, respectively. The application of the developed methodology to the analysis of cow milk samples of the Spanish market evidenced the presence of concentration ranges from 11.1 to 26.0 ng ml⁻¹ for Se, and from 1.04 to 9.7 ng ml⁻¹ for Te having found a good comparability with data obtained after dry-ashing of samples.     

Enhancement of electronic excitation energy transfer in the colloidal crystals of colloidal silica suspensions doped with fluorescent dyes

The efficiency of electronic excitation energy transfer from photo-excited rhodamine 110 (Rh110, energy donor) to rhodamine B (RhB, energy acceptor) in an exhaustively deionized colloidal silica suspension has been studied. This colloidal suspension shows Bragg reflection due to the formation of colloidal crystals and the Bragg-peak wavelength is controllable by the volume fraction of the silica spheres. When the Bragg-peak wavelength matches with the fluorescence band of Rh110, a depletion was observed in the Rh110 fluorescence spectrum. This means the fluorescence of Rh110 is partially trapped due to the Bragg reflection inside the crystal lattice. In the coexistence of RhB, the enhancement of RhB fluorescence intensity was observed. These facts clearly indicate the trapped photon energy of Rh110 is efficiently transferred to RhB within the colloidal crystals. The quantitative measurements showed that the enhancement of the transfer efficiency is 20% (or slightly more) in the present experimental conditions.     

醌亚胺类碱性染料在极谱分析中的应用——用天青Ⅰ测定痕量亚硝酸根

本文提出一个基于在酸性介质中,是青Ⅰ与NO_2~-发生亚硝化反应单扫描示波极谱法测定痕量NO_2~-的新方法。在0.06mol/L盐酸介质中。天青Ⅰ的亚硝化产物于-0.65V(vs.SCE)产生一灵敏的二次导数峰。峰高与NO_2~-浓度在0.002～0.24μg/mL范围内成直线关系。方法应用于水样、土壤中NO_2~-的测定,结果满意。
