A flow system for substrate recycling of NAD+/NADH was set up with an enzyme reactor containing coimmobilized glycerol dehydrogenase (GDH) and diaphorase. The product from the diaphorase catalysis, hexacyanoferrate(II), aws detected amperometrically at a glassy carbon electrode. The amplification factor was 150 for a reactor volume of 100 μ l at a flow-rate of 0.5 ml/min. With a stopped flow of four minutes, the signal increased another 88 times, resulting in a signal amplification of 13 300 times. Equations are derived for the amplification factor and used for a discussion of the optimization of amplification systems. The Km for GDH with glycerol as a substrate was found to be 5 × 10−3 M at pH 8.0. GDH from Cellulomonas sp. was purified on a gel filtration column and the purified enzyme showed a specificity toward NAD+, compared to NADP+, that was higher than 99.9%. Due to the NAD+ specificity of the purified GDH, the enzyme amplification system reported here could be used in detection systems for enzyme immunoassays when using alkaline phosphatase as a label and NADP+ as a substrate. The stability of immobilized GDH and diaphorase is several orders of magnitude better than that of alcohol dehydrogenase, which is the enzyme commonly used for NAD+-specific detection in these applications. 相似文献

Since hollow-fiber flow field-flow fractionation (HF FIFFF) utilizes a cylindrical channel made of a hollow-fiber membrane, which is inexpensive and simple in channel assembly and thus disposable, interests are increasing as a potential separation device in cells, proteins, and macromolecules. In this study, performance of HF FIFFF of proteins is described by examining the influence of flow rate conditions and length of fiber (polyacrylonitrile or PAN in this work) on sample recovery as well as experimental plate heights. The interfiber reproducibility in terms of separation time and recovery was also studied. Experiments showed that sample recovery was consistent regardless of the length of fiber when the effective field strength (equivalent to the mean flow velocity at the fiber wall) and the channel void time were adjusted to be equivalent for channels of various fiber lengths. This supported that the majority of sample loss in HF FIFFF separation of apoferritin and their aggregates may occur before the migration process. It is finally demonstrated that HF FIFFF can be applied for characterizing the reduction in Stokes' size of low density lipoproteins from blood plasma samples obtained from patients having coronary artery disease and from healthy donors. 相似文献

Flow field-flow fractionation (flow FFF), a separation technique for particles and macromolecules, has been used to separate carbon nanotubes (CNT). The carbon nanotube ropes that were purified from a raw carbon nanotube mixture by acidic reflux followed by cross-flow filtration using a hollow fiber module were cut into shorter lengths by sonication under a concentrated acid mixture. The cut carbon nanotubes were separated by using a modified flow FFF channel system, frit inlet asymmetrical flow FFF (FI AFIFFF) channel, which was useful in the continuous flow operation during injection and separation. Carbon nanotubes, before and after the cutting process, were clearly distinguished by their retention profiles. The narrow volume fractions of CNT collected during flow FFF runs were confirmed by field emission scanning electron microscopy and Raman spectroscopy. Experimentally, it was found that retention of carbon nanotubes in flow FFF was dependent on the use of surfactant for CNT dispersion and for the carrier solution in flow FFF. Langmuir-Blodgett (LB) films are constructed by successively transferring monomolecular layers formed at the air-water interface onto solid substrates. One of the advantages of the LB technique in fabricating molecular aggregates lies in the fact that it can employ various kinds of molecules by mixing them at the air-water interface. The mixed system may exhibit new properties that are not observed for individual components. This method would be useful, for example, in the studies of the formation and control of the J-aggregates of functional dyes that attract attention both in science and technology. In this paper, I review this subject mainly based on our recent results in merocyanines. LB films of merocyanine dyes, mixed with arachidic acid (C(20)), exhibit J-aggregate formation and have been serving as typical systems in revealing the physical and structural aspects of nanosized molecular aggregates constructed as monolayers. In the case of LB films of a merocyanine dye having benzothiazole as donor nucleus (abbreviated as DS), electron spin resonance (ESR) spectroscopy has been successful in determining the characteristic in-plane orientation of dye molecules with respect to the dipping direction, which led to the discovery of the flow orientation effect during the dipping process of LB films as the origin of optical dichroism often observed in LB films. In this article, after an introduction of ESR spectroscopy, three major topics on the merocyanine J-aggregation and its characterization in mixed films are discussed. The first topic is the observation and control of the size of J-aggregates in the dilution limit of dyes in arachidic acid matrix for a methyl-substituted DS (6-Me-DS). Dependence of atomic force microscopy (AFM) patterns on the molar ratio allows the identification of dye domains. J-band optical peak analysis based on the Kuhn's extended dipole model, supplemented by a novel application of femtosecond pump-probe optical spectroscopy, yields the size of the J-aggregates of 10(3). The second topic is the control of the J-band peak wavelengths by mixing two different kinds of dye molecules. The first case is the mixture of a J-forming 6-Me-DS and non-J-forming merocyanine analog, DO with benzo-oxazole instead of benzothiazole of DS. The second case is the mixture of both J-forming dyes but with different J-band peak positions, 6-Me-DS and another analog of 5-Cl-DS. The optical peak shifts depending on the molar mixing ratio will be presented. The last topic is related to the elucidation of electronic states of dye molecules in the J-aggregates. Light-induced ESR (LESR) of DS films with stable isotope ((15)N or (13)C)-substituted dyes provide clear evidence for the photoinduced charge transfer by the detection of hyperfine structures. Moreover, infrared (IR) spectroscopy of (13)C-enriched dye identifies the IR absorption peak of the relevant carbon in the chromophore. The results give evidence for the enhanced intramolecular charge transfer of dyes in the J-aggregates compared with an isolated merocyanine composed of donor and acceptor moiety. Lastly, the Cl attachment in 5-Cl-DS leads to a significant enhancement of the nitrogen hyperfine coupling in the LESR spectra. These examples and others demonstrate the potential of LB films of merocyanines in the studies of the nanosized molecular aggregates in monolayer assemblies. 相似文献

An automatic flow procedure for the determination of glycerol in wines by employing a flow system based on multicommutation and enzymatic reaction is described. Glycerol dehydrogenase was immobilized on aminopropyl glass beads and packed into a column that was coupled to the flow system. The NADH produced by the enzymatic reaction was monitored by spectrophotometry at 340 nm and its radiation absorption presented a relationship with glycerol concentration. The system manifold comprised a set of three-way solenoid valves controlled by a microcomputer, which was furnished with electronic interfaces and runs a software that was designed to carry out on-line sample dilution, reagent addition, and data acquisition. The procedure allows the determination of glycerol in wine samples without any prior pretreatment. The procedure presented as profitable features a linear response range between 2.0 and 10.0 g l−1 glycerol (R=0.998), a detection limit of 0.006 g l−1 glycerol, a relative standard deviation of 1.8% (n=14) for a typical wine sample presenting 5.3 g l−1 glycerol, a sampling throughput of 33 determinations per hour, and a NAD+ consumption of 0.8 mg per determination. Bioassay directed-fractionation led to the identification of litseaverticillols A-H (1-8) from the leaves and twigs of Litsea verticillata Hance. These new sesquiterpenes possess a unique skeleton that was recently designated as ‘litseane’. The structures of these compounds were determined by spectroscopic means including 1D and 2D NMR data. Structural configurations were determined by ROESY experiments. Mosher ester reactions and optical rotation measurements established the sesquiterpenes 1-8 as racemates. Isolates 1-8 inhibited HIV-1 replication in HOG.R5 cells with IC50 values ranging from 2 to 15 μg/ml (8-58 μM) while affecting the growth of HOG.R5 at concentrations 2-3-fold higher. Based on this data, structure-activity relationships can be discerned, suggesting compounds of this class are good candidates for analog production. 相似文献

The application of on-line photochemistry with flow injection (FI) and liquid chromatography (LC) in conjunction with atmospheric pressure electrospray mass spectrometry (LC-APESI-MS) for the identification of similar indole derivatives is reported here. The photo-transformation of the indole compounds is strongly affected by the substituent groups on the aromatic and heterocyclic rings. Upon photolysis for 2.5 min, the mass spectrum of tryptamine (Try) which has no OH substituent on the aromatic ring does not differ greatly from that obtained without photolysis. However, after photolysis of serotonin (Ser) which has one OH group on C5 of the aromatic ring, the mass spectrum indicates the formation of dimers and higher molecular weight ions. The fragmentation pattern of 5-hydroxytryptophol (Phol) without photolysis resembles that of Ser with a base peak of m/z 160. Upon photolysis using MeOH-H2O (10/90), Phol is found to form a base peak at m/z 375 (100%) and a major peak at m/z 214 (66%) in addition to other ions with lower abundance. Melatonin (Mel) and tryptophan (Phan) upon photolysis are found to form high molecular weight ions with a relative low abundance. The mass spectrum of indole-3-acetic acid (Inaa) with on-line photolysis also shows different ions that are not formed without photolysis. 相似文献

We address a novel truck scheduling problem arising in crossdocking logistics, in which inbound trucks carry items (pallets) which must be sorted and loaded onto outbound trucks. We minimize the utilisation of the warehouse by focusing on the synchronisation between the different related trucks. The problem is to assign the trucks to the doors of the warehouse and sequence them, in order to minimize the total time spent in the system by the pallets. We discuss the complexity of the problem, showing that even with a single door the problem is NP-hard in general, and discuss some special cases. 相似文献

The concentration of potassium was determined by a combination of flow injection analysis (FIA) with an all-solid-state potassium sensor detection. The all-solid-state potassium-selective electrode possessing long-term potential stability was fabricated by coating an electroactive polypyrrole/poly(4-styrenesulfonate) film electrode with a plasticized poly(vinyl chloride) membrane containing valinomycin. The simple FIA system developed in this laboratory demonstrated sensitivity identical to that in the batch system and achieved considerably rapid assay (150 samples h−1). Analyses of soy sauce and control serum samples by this FIA system yielded results in good agreement with those obtained by conventional measurements. 相似文献
