A fast and sensitive chemiluminescent enzyme-linked immunosorbent assay method to measure pathogenic bacteria, Escherichia coli O157:H7, on immuno-chromatographic membrane was studied. Non-specific binding of proteins on membrane strip was controlled to attain the best performance of immunosensor by optimising the composition of a running buffer. The specificity of the proposed immunostrip was confirmed by conducting experiments for four different micro-organisms. A chemiluminescent signal could be successfully generated from a proposed immunostrip sensing system, and a significant change in the chemiluminescent light intensity with the concentration of target microbes was obtained. E. coli O157:H7 could be quantitatively measured in the range of 1.1?×?103?–1.1?×?107 CFU (colony forming units) mL?1 within 16?min by using the developed chemiluminescent immunostrip. 相似文献

An efficient and convenient synthesis of novel chromeno[3′,4′:5,6]pyrano[2,3-d]pyrimidines is described, using the electrogenerated anion of acetonitrile as the base in the presence of tetrabutylammonium fluoride as an effective supporting electrolyte in a one-pot, three-component condensation of barbituric acid, an aromatic aldehyde or isatin, and 4-hydroxycoumarin. The generation of free radicals is a key process in the formation and the collapse of the bubbles in water, however, the direct and dynamic observation of the radicals in this process at single bubbles has never been achieved. Here, the hydroxyl (OH.) and oxygen (O2.−) radicals at single oxygen bubbles are continuously traced using chemiluminescence (CL), in which these radicals at the bubble react with the surrounding luminol in the solution emitting the light. Varied increase trends of luminescence are observed in the generation of a bubble, floating, short parking at the water/air interface and the final explosion, revealing the complexity in the distribution of radicals at the bubble unprecedentedly. Despite more radicals are observed at the bubble generated at a deep position under the water for the stabilization, almost the same amount of radicals are included in the bubbles that is independent on the water pressure during the production of the bubble. This rich information collected from the dynamic study of bubbles illustrates the complicated generation and distribution process of radicals at the bubbles, and will facilitate the understanding of the function about the bubbles. 相似文献

该文基于酶辅助靶标循环信号放大策略构建了用于黄曲霉毒素B1(AFB1)高灵敏检测的化学发光适体传感器。以G-四链体/氯化血红素DNA酶为信号分子设计了免标记的适体探针H1-S1和发夹探针H2。适体探针结合目标AFB1,在核酸外切酶I辅助下,触发靶标循环反应产生发夹H1。发夹H1与H2杂交,释放出完整的G-四链体序列,并进一步与氯化血红素结合形成G-四链体/氯化血红素DNA酶。DNA酶通过催化氧化鲁米诺-H2O2化学发光体系产生化学发光信号,实现AFB1的放大检测。在最优实验条件下,化学发光强度与AFB1质量浓度的对数在0.001~100 ng/mL范围内呈良好的线性关系,相关系数(r2)为0.9955,检出限为0.93 pg/mL,回收率为93.7%~107%。该适体传感器操作简单、灵敏度高、特异性好,在黄曲霉毒素污染检测方面具有良好的应用前景。 相似文献

We provide evidence of single attoliter oil droplet collisions at the surface of an ultra‐microelectrode (UME) by the observation of simultaneous electrochemical current transients (i–t curves) and electrogenerated chemiluminescent (ECL) transients in an oil/water emulsion. An emulsion system based on droplets of toluene and tri‐n‐propylamine (2:1 v/v) emulsified with an ionic liquid and suspended in an aqueous continuous phase was formed by ultrasonification. When an ECL luminophore, such as rubrene, is added to the emulsion droplet, stochastic events can be tracked by observing both the current blips from oxidation at the electrode surface and the ECL blips from the follow‐up ECL reaction, which produces light. This report provides a means of studying fundamental aspects of electrochemistry using the attoliter oil droplet and offers complementary analytical techniques for analyzing discrete collision events, size distribution of emulsion systems, and individual droplet electroactivity. 相似文献

基于鲁米诺(luminol) 化学发光体系,采用自主研发的在线臭氧浓度检测仪,建立了一种实时在线检测臭氧浓度的方法,用于分析测定痕量浓度水平的臭氧气体。 考察了鲁米诺、氢氧化钾、部分醇类化合物和表面活性剂等因素对化学发光强度的影响。 结果表明,在鲁米诺(0.005 mol/L)、氢氧化钾(0.05 mol/L)体系中加入乙二醇(体积分数1.5%)、甲醇(体积分数1.5%)、乙醇(体积分数1.0%)、丙三醇(体积分数3.0%)能显著增强鲁米诺体系检测O3的化学发光信号,而甲醛溶液 (体积分数3.0%)能有效抑制NO2信号的干扰。 同时,测得检测臭氧的检出限为1.26 μg/m3、相对标准偏差为0.32%,相对误差为0.75%。 利用该体系测定臭氧,具有信号稳定、精密度好、准确度高、检出限低等优点,适用于大气中微量O3的在线连续检测。 相似文献
