N,N′-diBoc-dityrosine (DBDY), which was synthesized by the oxidative C–C coupling of 2 N-Boc-l-tyrosine molecules, was conjugated with two isoniazid (INH) molecules. Due to the quenching effect of INH, DBDY–(INH)2 lacks the fluorescence of DBDY. As such, it was tested for use in the detection of proteases by measuring fluorescence recovery. In this study, serine proteases (chymotrypsin, trypsin, subtilisin, and proteinase K), metalloproteases (thermolysin and carboxypeptidase A, dispase, and collagenase), aspartic proteases (pepsin and aspergillopepsin) and cysteine proteases (papain and chymopapain) were chosen. Reported optimum assay conditions were chosen for each enzyme. Only papain and chymopapain catalyzed the hydrolysis of DBDY–(INH)2 and led to fluorescence recovery, possibly due to their extensive binding sites and the INH-mediated inhibition of metalloproteases and aspartic proteases.  相似文献

Abstract

Based on the selected monoclonal antibody, the effect of three reaction formats on sensitivity, specificity, and determination ability for real samples were investigated. The results showed the antibody-coated format was the optimal assay for parathion determination. The sensitivity of the assay was 3.20 ng/ml, with a detection limit of 0.40 ng/ml, and the assay time was 1.5 h. The average recoveries of parathion in river water, rice, cucumber, green soy bean, and cabbage were 98.56%, 89.46%, 99.25%, 118.57%, and 101.39%, respectively. In addition, when rice and cabbage extracts were analyzed by the assay and HPLC, the correlation was greater than 0.9.  相似文献

Despite efforts to control fungal contamination, some mycotoxins, including deoxynivalenol, zearalenone, aflatoxin, and ochratoxin, are ubiquitous in the rural China. The purpose of this study was to establish a simple method for noninstrumental detection of these residues by a multianalyte dot enzyme-linked immunosorbent assay. The multianalyte dot enzyme-linked immunosorbent assay was simple, inexpensive, and fast and is expected to have broad applications in the food processing industry in rural China.  相似文献

Abstract

Mangrove plants are endowed with various biologically active compounds which have potent antibacterial and antioxidant properties. In present study, a bioactivity-guided fractionation for antibacterial and antioxidant active metabolites from the twigs of Avicennia officinalis collected from Kuala Selangor Nature Park, Selangor, Malaysia gave 13 major fractions. The antibacterial activity of A. officinalis fractions using well-diffusion showed strong selectivity on the Gram-positive bacteria (Staphylococcus epidermidis, S. aureus and Bacillus subtilis) with minimum inhibition concentration (MIC) values of 0.156-5.00?mg/mL. However, no antibacterial activities were observed on the Gram-negative bacteria (Vibrio cholera, Enterobacter cloacae and Escherichia coli). The active antibacterial fractions were further isolated using several chromatographic techniques to give two naphthofuranquinones, namely, avicenol C (1) and stenocarpoquinone B (2). Meanwhile, the antioxidant activity of A. officinalis fractions were evaluated using DPPH radical scavenging assay exhibited low antioxidant activities. Molecular structure of the naphthofuranquinones was elucidated using 1?D and 2?D NMR spectroscopy.  相似文献

Analytical chemistry is playing an ever-increasingly important role in the global wine industry. Chemical analysis of wine is essential in ensuring product safety and conformity to regulatory laws governing the international market, as well as understanding the fundamental aspects of grape and wine production to improve manufacturing processes. Within this field, advanced instrumental analysis methods have been exploited more extensively in recent years. Important advances in instrumental analytical techniques have also found application in the wine industry. This review aims to highlight the most important developments in the field of instrumental wine and grape analysis in the African context. The focus of this overview is specifically on the application of advanced instrumental techniques, including spectroscopic and chromatographic methods. Recent developments in wine and grape analysis and their application in the African context are highlighted, and future trends are discussed in terms of their potential contribution to the industry.  相似文献

A new bioessential Knoevenagel condensate Schiff base ligand (L) was synthesized by the reaction of 3‐(4‐hydroxy‐3‐methoxybenzyl)pentane‐2,4‐dione and 4‐aminoantipyrine. The ligand forms monomeric divalent transition metal complexes ( 1 – 4 ) which were characterized using spectral and analytical data. All these complexes have the general formula [ML]Cl2, where M = Co(II), Ni(II), Cu(II) and Zn(II). They are electrolytic in nature and adopt square planar geometry. The binding propensity of these complexes with calf thymus DNA was investigated using absorption spectrophotometric titration, cyclic voltammetry and viscosity measurements. The binding constant values imply that the complexes bind with DNA via intercalation mode. The in vitro antibacterial and antifungal activities reveal that the complexes have good antimicrobial efficacy against a set of pathogens. The nucleolytic cleavage activity of these complexes on pUC18 DNA was investigated using agarose gel electrophoresis. Also, the in vitro cytotoxicity of the synthesized complexes against a panel of human tumour cell lines (MCF‐7 and HeLa) and normal cell lines (NHDF and HEK) was assayed using the MTT method. Interestingly, complex 1 exhibits more potent anticancer activity than cisplatin and other complexes.  相似文献

This study was designed to isolate, characterise and explore in vitro antioxidant potential of compounds from the chloroform extract of Cedrus deodara wood belonging to Pinacae family. We have investigated the in vitro free radical scavenging activity of isolated compounds by measuring total antioxidant capacity (TAC) and reducing power ability. From the chloroform extract five sesquiterpenes namely atlantone, himaphenolone, atlantolone, deodardione and atlantone-2,3-diol were identified. Himaphenolone and deodardione from the chloroform extract of this plant have been isolated for the first time. A marked dose-dependent reducing power and strong TAC were found in favour of himaphenolone and atlantone-2,3-diol, respectively. Moreover, deodardione and atlantolone also exhibit good antioxidant potentials. However, atlantone has showed least antioxidant activity. In conclusion, the chloroform extract of C. deodara exhibited significant antioxidant potential mainly due to the presence of sesquiterpenes which may be responsible for various pharmacological activities of this plant.  相似文献

A sensitive, accurate and highly stereoselective assay for the simultaneous determination of venlafaxine (VEN) and its equipotent metabolite, O‐desmethyl venlafaxine (ODV), in human plasma was developed and validated. Analytes were simultaneously extracted from plasma using solid‐phase extraction and detected by tandem mass spectrometry in positive ion mode with a turbo ion spray interface. Deuterium‐labeled VEN and ODV were used as internal standards. Chromatographic separation was performed on a Chiral AGP column, using a time programmed gradient flow with a total run time of 16 min. The method has a lower limit of quantitation of 0.60 ng/mL. The assay was linear over a range 0.60–300.00 ng/mL for both the enantiomers of VEN and ODV, respectively, with coefficient of correlation > 0.99. The extraction recoveries were >77.0% on an average for all the four analytes. The analytes were found stable in plasma through three freeze (?15 °C) and thaw cycles and under storage at room temperature for 8 h, and also in mobile phase at 10 °C for 54 h. The method has shown good reproducibility, with intra‐ and inter‐day variation coefficients < 9%, for all the analytes, and has proved to be very reliable for analysis of VEN and its metabolite in clinical study samples. Two reversed-phase liquid chromatographic (RP-LC) methods have been developed for the determination of sitagliptin phosphate monohydrate (STG). The first method comprised the determination of STG alone in bulk and plasma; and in its pharmaceutical preparation. This method was based on isocratic elution of STG using a mobile phase consisting of potassium dihydrogen phosphate buffer pH (7.8)-acetonitrile (70:30, v/v) at a flow rate of 1 mL min−1 with flourometric detection. The flourometric detector was operated at 267 nm for excitation and 575 nm for emission. In the second method, the simultaneous determination of STG and metformin (MET) in the presence of sitagliptin alkaline degradation product (SDP) has been developed. In this method, the ternary mixture of STG, MET and SDP was separated using a mobile phase consisting of potassium dihydrogen phosphate buffer pH (4.6)-acetonitrile-methanol (30:50:20, v/v/v) at a flow rate of 1 mL min−1 with UV detection at 220 nm. Chromatographic separation in the two methods was achieved on a Symmetry® Waters C18 column (150 mm × 4.6 mm, 5 μm). Linearity, accuracy and precision were found to be acceptable over the concentration ranges of 0.25-200 μg mL−1 for STG with the first method and 5-160 μg mL−1, 25-800 μg mL−1 for STG and MET, respectively with the second method. The optimized methods were validated and proved to be specific, robust and accurate for the quality control of the cited drugs in pharmaceutical preparations.  相似文献

This review discusses the application of quantum dots (QDs) to chemical and biological detection, for which they have excellent features, particularly size-dependent optical properties.We can summarize the main areas discussed in this review as follows:(1) QDs associated with enzyme-linked immunosorbent assay (ELISA), chip detection and capillary electrophoresis (CE) enhance the sensitivity and the speed of detection of residues;(2) QDs are applied with other techniques, including polymerase chain reaction (PCR), fluorescence resonance-energy transfer (FRET) analysis, fluorescence in-situ hybridization (FISH) and western blot analysis; and,(3) QDs combined with the above techniques can successfully detect DNA and protein.We also cover perspectives and challenges in analytical applications of QDs.  相似文献
