The activity of enzyme I (EI), the first protein in the bacterial PEP:sugar phosphotransferase system, is regulated by a monomer–dimer equilibrium where a Mg2+-dependent autophosphorylation by PEP requires the homodimer. Using inactive EI(H189A), in which alanine is substituted for the active-site His189, substrate binding effects can be separated from those of phosphorylation. Whereas 1 mM PEP (with 2 mM Mg2+) strongly promotes dimerization of EI(H189A) at pH 7.5 and 20 °C, 5 mM pyruvate (with 2 mM Mg2+) has the opposite effect. A correlation between the coupling of N- and C-terminal domain unfolding, measured by differential scanning calorimetry, and the dimerization constant for EI, determined by sedimentation equilibrium, is observed. That is, when the coupling between N- and C-terminal domain unfolding produced by 0.2 or 1.0 mM PEP and 2 mM Mg2+ is inhibited by 5 mM pyruvate, the dimerization constant for EI(H189A) decreases from >108 to <5 × 105 or 3 × 107 M−1, respectively. With 2 mM Mg2+ at 15–25 °C and pH 7.5, PEP has been found to bind to one site/monomer of EI(H189A) with KA′106 M−1 (ΔG′=−33.7±0.2 kJ mol−1 and ΔH=+16.3 kJ mol−1 at 20 °C with ΔCp=−1.4 kJ K−1 mol−1). The binding of PEP to EI(H189A) is synergistic with that of Mg2+. Thus, physiological concentrations of PEP and Mg2+ increase, whereas pyruvate and Mg2+ decrease the amount of dimeric, active, dephospho-enzyme I. 相似文献

Ginseng (root of Panax ginseng C. A. Meyer) cells were cultivated on medium supplemented with various carbohydrates including sucrose, glucose, and fructose,
at initial concentrations ranging from 10 to 110 g/L. Sucrose was shown to be the superior carbon source to the monosaccharides
for ginseng cell growth and the optimal concentration was between 30 and 50 g/L. An increase in the initial concentration
within this range increased the maximum cell density and growth index significantly, whereas much higher concentrations inhibited
cell growth. Feeding of sucrose and some other medium components during the growth (fed-batch mode) was more effective in
enhancing the cell growth and biomass productivity, increasing the growth index by more than 60–70% and biomass productivity

Conformational analysis of vasoactive intestinal peptide (VIP) receptor binding inhibitor Leu1-Met2-Tyr3-Pro4-Thr5-Tyr6-Leu7-Lys81 by various NMR techniques and constrained molecular dynamics (MD) simulation studies revealed that the molecule had a turn structure involving its Tyr3-Pro4-Thr5-Tyr6 moiety with intramolecular hydrogen bond between Tyr6NH→Tyr3CO. In order to mimic the structure of 1, peptidomimetic analogs 2-4 were synthesized using conformationally constrained scaffolds of 3,4-dideoxy furanoid sugar amino acids (2S,5R)-ddSaa1 5 and its enantiomer (2R,5S)-ddSaa2 6. All these analogs displayed well defined three-dimensional structures akin to that found in 1. Peptides 2 and 3, which differed only in the sugar amino acid stereochemistry, show propensity of structures with identical intramolecular hydrogen bonds between ThrNH→MetCO. Dielectric-relaxation studies in the frequency range 200 kHz to 35 GHz are reported for a range of sugars (from mono- to trisaccharides) in aqueous solution. The complex dielectric spectra were analyzed using a weighted least-squares minimization method to resolve the various component relaxations, and the implications of the analyses in terms of the molecular dynamics of solute and solvent and the interactions between solute and solvent are discussed. For the highest concentration studied (ca. 2M), it was found that the most significant analysis required three discrete relaxation processes, whereas lower concentration samples could usually be satisfactorily fitted with two. Irrespective of any uncertainty in model selection, a number of conclusions regarding the solute-solvent interactions can be made, and it is shown how final quantification of the extents of hydration can be made using the input of information from other techniques. 相似文献
