The human milk secretory immune system is known to be the first line of protection for the newborn infant against various
pathogens. Secretory IgA (sIgA), the typical immunoglobulin found in secretions, can fight infections through many mechanisms.
Using different methods, we have shown that sIgA from the milk of healthy women possesses DNAse and RNAse activities. The
catalytic center is localized in the light chain of catalytic sIgA, while the DNA-binding center is predominantly formed by
its heavy chain. The enzymic properties and substrate specificity of catalytic sIgA distinguish it from other known DNases
and RNases. It is reasonable to assume, that the milk DNA- and RNA-hydrolyzing antibodies are capable not only of neutralizing
viral and bacterial nucleic acids by binding these antigens as well as by hydrolyzing them. The DNA-hydrolyzing activity of
Abs raises the possibility that these catalytic Abs may provide protective functions for the newborn through the hydrolysis

Wavelength selection framework for classifying food and pharmaceutical samples into multiple classes

Near infrared (NIR) spectroscopy is an efficient, low‐cost analytical technique widely applied to identify the origin of food and pharmaceutical products. NIR spectra‐based classification strategies typically use thousands of equally spaced wavelengths as input information, some of which may not carry relevant information for product classification. When that is the case, the performance of predictive and exploratory multivariate techniques may be undermined by such noisy information. In this paper, we propose an iterative framework for selecting subsets of NIR wavelengths aimed at classifying samples into categories. For that matter, we integrate Principal Components Analysis (PCA) and three classification techniques: k‐Nearest Neighbor (KNN), Probabilistic Neural Network (PNN) and Linear Discriminant Analysis (LDA). PCA is first applied to NIR data, and a wavelength importance index is derived based on the PCA loadings. Samples are then categorized using the wavelength with the highest index and the classification accuracy is calculated; next, the wavelength with the second highest index is inserted into the dataset and a new classification is performed. This forward‐based iterative procedure is carried out until all original wavelengths are inserted into the dataset used for classification. The subset of wavelengths leading to the maximum accuracy is chosen as the recommended subset. A sensitive and rapid magnetic nanoparticle-based fluorescent immunoassay for the determination of aflatoxin M1 in raw milk was developed. Aflatoxin M1 was converted to aflatoxin M1-o-carboxymethyl oxime. The aflatoxin M1-oxime was used for the preparation of aflatoxin M1-oxime-fluoresceinamine conjugate through the carbodiimide reaction. The aflatoxin M1-oxime-fluoresceinamine conjugate was characterized by ultraviolet–visible and infrared spectroscopy. Magnetic nanoparticles (Fe3O4) were synthesized and modified by 3-(aminopropyl)triethoxysilane. The size of initial (139?nm) and functionalized magnetic nanoparticles (147?nm) was determined by particle analysis. The optimal mass of immobilized antibody (25?µg) and optimal concentration of aflatoxin M1-oxime-fluoresceinamine conjugate (15?µg?mL?1) for magnetic nanoparticle-based fluorescent immunoassay were determined. The developed immunoassay provided a linear aflatoxin M1 concentration range from 3.0 to 100?pg?mL?1 in bovine milk. The detection limit was 2.9?pg?mL?1. The results of aflatoxin M1 magnetic nanoparticle-based fluorescent immunoassay in heat-treated milk and phosphate-buffered saline at pH 6.6 were compared. The influence of the somatic cell count, pH, and fat concentration in bovine milk on the aflatoxin M1 immunoassay was investigated. The influence of the milk species on the immunoassay was also characterized. The high fat concentration ovine milk depressed the sensitivity of the aflatoxin M1 immunoassay. 相似文献

A simple, rapid, field-portable colorimetric method for the detection of melamine based on melamine-induced color change of label-free gold nanoparticles (Au NPs) was developed in this study. Melamine can induced the aggregation of Au NPs and results in the color change from wine-red to purple, which provided a platform for rapid and field-portable colorimetric detection of melamine. The proposed method can be used to detect melamine in liquid milk and infant formula with a detection limit of 1.0 and 4.2 ppm, respectively, within 30 min by naked eyes observation without the aid of any advanced instrument and the need of any complex pretreatment, and detect as low as 0.15 ppm of melamine in liquid milk and 2.5 ppm of melamine in infant formula with UV-vis-spectroscopy. The McMurry coupling of (tetraphenylcyclobutadiene)cobalt(cyclopentadienyl) ketones, (C4Ph4)Co[C5H4C(O)R], where R = Me, 3a, or Et, 3b, with a range of substituted benzophenones furnished a series of cobaltifens, organometallic analogues of tamoxifen whereby a phenyl ring has been replaced by an organo-cobalt sandwich moiety. These systems of the general formula (η4-C4Ph4)Co[η5-C5H4C(R)C(Ar)Ar′], where R = Me or Et, and Ar = Ar′ = p-C6H4X where X is OH, 2a and 2b, OMe, 2c and 2d, OBn, 2e and 2f, or O(CH2)2NMe2, 12a and 12b, and where Ar = C6H4OH and Ar′ = C6H4O(CH2)2NMe2, 2g and 2h, have been characterised by NMR spectroscopy and/or X-ray crystallography. The effect of 2a and 2b, 2g and 2h, and 12a and 12b on the growth of MCF-7 (hormone-dependent) and MDA-MB-231 (hormone-independent breast cancer cells) was studied. The dihydroxycobaltifens 2a and 2b exhibit a strong estrogenic effect on MCF-7 cells while the aminoalkyl-hydroxycobaltifens, 2g and 2h, were found to be only slightly cytotoxic on MDA-MB-231 cells (IC50 = 27.5 and 17 μM); surprisingly, however, the bis-(dimethylaminoethoxy)cobaltifens, 12a and 12b were shown to be highly cytotoxic towards both cell lines (IC50 = 3.8 and 2.5 μM). 相似文献

Promising recent investigations have shown that breast malignancies exhibit restricted diffusion on diffusion-weighted imaging (DWI) and may be distinguished from normal tissue and benign lesions in the breast based on differences in apparent diffusion coefficient (ADC) values. In this study, we assessed the influence of intravoxel fat signal on breast diffusion measures by comparing ADC values obtained using a diffusion-weighted single shot fast spin-echo sequence with and without fat suppression. The influence of breast density on ADC measures was also evaluated. ADC values were calculated for both tumor and normal fibroglandular tissue in a group of 21 women with diagnosed breast cancer. There were systematic underestimations of ADC for both tumor and normal breast tissue due to intravoxel contribution from fat signal on non–fat-suppressed DWI. This ADC underestimation was more pronounced for normal tissue values (mean difference=40%) than for tumors (mean difference=27%, P<.001) and was worse in women with low breast tissue density vs. those with extremely dense breasts (P<.05 for both tumor and normal tissue). Tumor conspicuity measured by contrast-to-noise ratio was significantly higher on ADC maps created with fat suppression and was not significantly associated with breast density. In summary, robust fat suppression is important for accurate breast ADC measures and optimal lesion conspicuity on DWI. 相似文献

To address people's concerns of exogenous oxytocin (OT) administration to lactating bovines, a study was undertaken to (a) establish an enzyme immunoassay (EIA) for OT determination in milk, (b) quantify OT in milk of cows administered OT, and (c) study influence of pasteurization on OT stability in milk. A sensitive EIA validated according to the criteria of European Union—Decision 2002/657/EC was developed for OT in skim milk in an analytical range of 10-250 pg mL−1 with a decision limit (CCα) of 30 pg mL−1 and detection capability (CCβ) of 41.5 pg mL−1. Milk samples collected from cows (n = 38) administered either 25 or 50 IU OT prior to milking were investigated for the presence of OT. There was no significant difference among both groups with the mean concentrations of OT being 15.8 and 14.9 pg mL−1 for cows subjected to 25 and 50 IU OT administration, respectively. The OT levels in skim milk of control cows (n = 30; untreated) were basal (around 10 pg mL−1). All the analyzed milk samples were below the CCα value of 30 pg mL−1. Pasteurization of OT spiked milk samples at different temperature and sample holding conditions reduced the immunological activity of OT to 43% at 110 °C. However, no further decline occurred in the immunological activity with increased pasteurization temperature and time. It was concluded that the milk OT concentrations after OT administrations were minimal and below the assay decision limit. However, OT was quite stable to pasteurization in OT spiked milk. 相似文献
