Preliminary theoretical and empirical work suggest that increased thyroarytenoid muscle activity may either increase or decrease fundamental frequency, depending on cricothyroid muscle activity and a new cross-sectional area parameter. This parameter is defined as the ratio of muscular tissue in vibration to total tissue in vibration. Canine laryngeal nerves were stimulated to measure vocal-fold length changes. These data, combined with previously reported tissue density, passive stress, and passive frequency data, were used to construct a set of curves predicting canine fundamental frequency from thyroarytenoid and cricothyroid muscle activity and the area ratio. The results suggest that high cricothyroid muscle activity and small area ratios tend to cause fundamental frequency lowering with increased thyroarytenoid muscle activity.  相似文献

Proteomic analysis of biological samples in disease models or therapeutic intervention studies requires the ability to detect and identify biologically relevant proteins present in relatively low concentrations. The detection and analysis of these low-level proteins is hindered by the presence of a few proteins that are expressed in relatively high concentrations. In the case of muscle tissue, highly abundant structural proteins, such as actin, myosin, and tropomyosin, compromise the detection and analysis of more biologically relevant proteins. We have developed a practical protocol which exploits high-pH extraction to reduce or remove abundant structural proteins from skeletal muscle crude membrane preparations in a manner suitable for two dimensional gel electrophoresis. An initial whole-cell muscle lysate is generated by homogenization of powdered tissue in Tris-base. This lysate is subsequently partitioned into a supernatant and pellet containing the majority of structural proteins. Treatment of the pellet with high-pH conditions effectively releases structural proteins from membrane compartments which are then removed through ultracentrifugation. Mass spectrometric identification shows that the majority of protein spots reduced or removed by high-pH treatment were contractile proteins or contractile-related proteins. Removal of these proteins enabled successful detection and identification of minor proteins. Structural protein removal also results in significant improvement of gel quality and the ability to load higher amounts of total protein for the detection of lower abundant protein classes.  相似文献

Pig muscle aldolase was insolubilized by covalent attachment to a polyacrylamide matrix containing carboxylic functional groups. The catalytic activity of the Akrilex C-aldolase was 2014 units/g solid, i.e., an activity loss of only about 5% relative to the initial activity. The pH optimum for catalytic activity shifted form 7.25 to 7.5 and the apparent temperature optimum from 313 to 318 K. The Michaelis constant of the insolubilized enzyme was significantly higher than that of the soluble aldolase. Heat- and urea-inactivation experiments revealed that the immobilization increased the stability of the enzyme.  相似文献

在pH=5.2的B-R缓冲溶液中,对槲皮素与牛血清白蛋白(BSA)相互作用的吸收光谱进行了初步研究。结果表明:槲皮素在290nm处有一强的吸收峰,当加入BSA作用后该吸收峰的强度升高,峰位基本不变,且没有新的吸收峰出现。在此波长下测定其复合物的吸光度,其吸光度的增加值(ΔA)与BSA的浓度在18—220μg/mL范围内呈良好的线性关系(r=0.9991),检出限为8μg/mL。该方法具有简便、快速、选择性好、灵敏度高等特点,应用于鲜奶粉和液态纯牛奶样品中总蛋白的测定,回收率分别为89.5%,93.2%,结果与考马斯亮蓝G250法基本一致。  相似文献

利用连续双波长近红外光技术测量肌肉中血红蛋白含氧量的变化情况,同时应用运动心率无线实时采集设备,设计了检测实时肌肉组织血氧含量和瞬时心率实验方案,对6名健康男性进行了100 m跑同步实测实验。实验表明,肌肉组织氧合血红蛋白浓度在100 m跑结束后继续降低,达到最低值的时刻滞后于运动结束时刻的时间为(6.65±1.10)s;心率则在运动结束后继续上升,达到最高值的时刻滞后于运动结束时刻的时间为(8.00±1.57)s。这些结果显示了双波长近红外光组织氧测试技术和运动心率无线实时采集系统能够正确检测极量强度运动时的血氧参数和心率,揭示了极量强度运动过程中肌肉中氧运输与消耗的过程及其与心率之间的动力学特征。  相似文献

Naringenin, a flavanone compound highly enriched in grapefruits, has been identified as a possible inhibitor of cell proliferation; and thus has the potential to act as an antitumorigenic agent. In this study, the binding of naringenin to bovine serum albumin (BSA) was studied at the physiological conditions (pH=7.40) by fluorescence and UV-vis spectroscopy. Naringenin strongly quenches the intrinsic fluorescence of BSA, and a decrease in the fluorescence quenching constant was observed together with an increase in temperature, which indicates that the fluorescence quenching of BSA by naringenin is a result of the formation of naringenin-BSA complex. Binding parameters calculating from Stern-Volmer method and Scatchard method showed that naringenin bind to BSA with the binding affinities of the order 104 L mol−1. Thermodynamic parameters such as ΔG, ΔH and ΔS, were calculated at different temperatures, showing that electrostatic interactions were mostly responsible for the binding of naringenin to BSA. Site marker competitive displacement experiments demonstrating that naringenin bind with high affinity to site I (subdomain IIA) of BSA. Furthermore, the effect of metal ions to naringenin-BSA system was studied, and the specific binding distance r (3.30 nm) between donor (Trp-212) and acceptor (naringenin) was obtained according to fluorescence resonance energy transfer (FRET).  相似文献

A novel protein imprinted polymer was prepared using acryloyl-β-cyclodextrin (β-CD) and acrylamide as monomers on the surface of silica gel. The bovine hemoglobin was used as template and β-CD was allowed to self-assemble with the template protein through hydrogen bonding and hydrophobic interaction. Polymerization was carried out in the presence of acrylamide as an assistant monomer, which resulted in a novel protein imprinted polymer. After removing the template, imprinted cavities with the shape and spatial distribution of functional groups were formed. Bovine serum albumin (BSA) cytochrome c (Cyt) and lysozyme (Lyz) were employed as non-template proteins to test the imprinting effect and the specific binding of bovine hemoglobin to the polymer. The results of the adsorption experiments indicated that such protein imprinted polymer, which was synthesized with β-CD and acrylamide as monomers, could selectively recognize the template protein.  相似文献

用光谱法研究头孢噻肟钠(CS)和牛血清蛋白(BSA)在模拟生命体生理条件下相互作用的特征.结果表明CS主要以静态猝灭方式使BSA的荧光强度显著降低;利用同步荧光法研究了CS和BSA作用后牛血清蛋白的构象发生了变化;求得不同温度下二者的结合常数和结合位点数,探讨了微量金属离子对实验体系结合常数的影响,并根据热力学参数确定了CS和BSA之间主要依靠静电力结合.根据Froester非辐射能量转移机理,测定了CS与BSA相互结合时的作用距离2.56nm,表明BSA与CS之间发生了非辐射能量转移.  相似文献
