在pH 4 .80NaAc HAc缓冲介质中 ,盐酸小檗碱 (BH)与十二烷基苯磺酸钠 (DBS)由于静电引力和疏水作用力形成缔合微粒 ,在 4 70nm处有一共振散射峰。随着DBS浓度增大 ,该峰急剧增强 ,即存在缔合微粒的散射光增强效应 ;345nm处的吸光度减弱 ,即存在缔合微粒的减色效应。研究了共振散射光谱测定BH的影响因素 ,提出了测定 (0 .35~ 4 .4 )× 10 -5mol/L盐酸小檗碱的共振散射光谱新方法 ,用于复方黄连素片剂和针剂样品的测定 ,结果满意  相似文献

The interaction of anticancer herbal drug berberine with double-strand DNA (dsDNA) and single-strand DNA (ssDNA) in solution, dsDNA immobilized on the glassy carbon electrode prepared by Langmuir-Blodgett technique, were investigated by electrochemical techniques (cyclic voltammetry, differential pulse voltammetry) and UV spectroscopy. The presence of DNA results in a decrease of the currents and a negative shift of the electrode potentials from the DPV curves of berberine, indicating the dominance of electrostatic interactions. The spectroscopy data confirmed that the predominant interaction between berberine and DNA is electrostatic. The binding of berberine with DNA, when analyzed in terms of the cooperative Hill model, yields the binding constant K(a)=2.2(+/-0.2)x10(4) M(-1), corresponding to the dissociation equilibrium constant K(d)=4.6(+/-0.3)x10(-5) M, which in the range of the applied concentrations of DNA (bp) and berberine, and a Hill coefficient m=1.82(+/-0.08) in Britton-Robinson buffer solution (0.05 M, pH 5.72) at T=298 K (25 degrees C). Apparently, at least two molecules of berberine have to bind as a couple to cause, e.g., the "elementary event" of current change. The results are suggestive for further fruitful applications of this anticancer herbal drug and DNA-modified electrodes.  相似文献

Using 266 nm laser flash photolysis it has been demonstrated that Berberine (BBR) in aqueous solution is ionized via a mono-photonic process giving a hydrated electron, anion radical that formed by hydrated electron react with steady state of BBR, and neutral radical that formed from rapid deprotonation of the radical cation of BBR. The quantum yield of photoionization is determined to be 0.03 at room temperature with KI solution used as a reference. Furthermore utilizing pH changing method and the SO4 radical oxidation method, the assignment of radical cation of BBR was further confirmed, the pKa value of it was calculated, and the related set up rate constant was also determined.  相似文献

采用超声波法提取关黄柏中有效成分盐酸小檗碱,应用正交试验法筛选盐酸小檗碱最佳提取工艺。采用高分离度快速液相色谱(RRLC)测定关黄柏中有效成分盐酸小檗碱的含量。色谱条件为:色谱柱:XDB-C18(4.6mm×50mm,1.8μm);流动相:乙腈-0.3%磷酸缓冲溶液(29∶71);检测波长:266nm;流速:0.7mL/min;柱温:25℃;进样量:2μL。盐酸小檗碱在0.075—0.3μg范围内线性关系良好,r=0.9998,其平均回收率为99.2%,RSD为1.48%。本法操作快速简便、准确可靠,适用于关黄柏中盐酸小檗碱的含量测定。  相似文献

In this work, a novel potassium ion (K+) sensor is presented using berberine–G-quadruplex complex as a fluorescent probe. This sensor is based on the K+that can induce the G-rich DNA to form G-quadruplex conformation. The G-quadruplex can bind berberine to form berberine–G-quadruplex complex, resulting in remarkable enhancement of fluorescence emission of the berberine–G-quadruplex system. In the presence of 800 mM sodium ion (Na+), the fluorescence of the berberine–G-quadruplex complex increased linearly with increasing K+ concentration in the range of 0.005–1.0 mM. The turn-on fluorescent assay is simple, inexpensive, and highly sensitive. We observed that Na+ in 10,000-fold molar excess does not interfere. The molecular mechanisms which produce enhanced fluorescence of berberine were discussed.  相似文献

In this paper, bovine serum albumin (BSA) was selected as a target molecule, and the sonodynamic damage to proteins in the presence of berberine (BER) and its mechanism were studied by means of absorption and fluorescence spectra. The results of hyperchromic effect of absorption spectra, and quenching of intrinsic fluorescence spectra indicated that the ultrasound-induced BSA molecules damage was enhanced by BER. The damage degree of BSA molecules increased with the increase in ultrasonic irradiation time and BER concentration. The results of synchronous fluorescence and three-dimensional fluorescence spectra confirmed that the synergistic effects of ultrasound and BER induced the damage of BSA molecules. The results of oxidation-extraction photometry with several reactive oxygen species (ROS) scavengers indicated that the damage of BSA molecules could be mainly due to the generation of ROS, and 1O2 was the major mediator of the ultrasound-induced BSA molecules damage in the presence of BER.  相似文献

Hyperglycemia is frequently accompanying with hyperlipidemia. To explore the potent drugs with dual-activity and dual-site effects that could reduce blood glucose and blood lipid at the same time, fibrate group with lipid-lowering effect on the 9th position of berberine(BBR) was introduced using the drug design combination principle and the multi-target collaborative treatment method. Moreover, the molecular structure of BBR was modified, and six 9-substituted derivatives of BBR were designed and synthesized, among which, five compounds have never been reported before. In addition, the molecular structures of these derivatives were identified using liquid chromatography-mass spectrometry(LC-MS), 1H nuclear magnetic resonance(1H NMR) and 13C NMR, respectively. Furthermore, the microwave irradiation experimental technique was applied in the synthesis reaction using the novel microwave synthesizer, which accelerated the reaction rate, enhanced the reaction yield, reduced the reaction by-products, and simplified the post-processing steps. In the meantime, the 9-position regioselective demethylation of BBR was explored through quantum chemical calculation during the synthesis of berberrubine. The computations were consistent with the experimental results, which contributed to deducing the mechanism of its selective methylation.  相似文献
