The chiral separation of simendan enantiomers using capillary electrophoresis was studied with beta-cyclodextrin (beta-CD) as chiral selector. The influences of the concentration and pH of borate buffer solution, beta-CD concentration and methanol content in the background electrolyte were investigated. These factors were compared with those in an HPLC with beta-CD as chiral mobile phase additive (CMPA-HPLC). The quantification properties of the developed CE method were examined. A baseline separation of simendan enantiomers was achieved in the background electrolyte of 20 mmol/L borate buffer (pH 11.0) containing 12 mmol/L beta-CD-methanol (50:50 in volume ratio). The CE method is comparable with CMPA-HPLC in chiral resolution, although the optimal pH in CE (11.0) is much higher than that (6.0) in CMPA-HPLC. This chiral CE method is applicable to the quantitative ananlysis and enantiomeric excess value determination of L-simendan. 相似文献

A resonance light scattering (RLS) imaging method was proposed based on imaging and measuring the RLS features of single suprahelical species of DNA, and its appfication to DNA assay was also investigated. In acidic medium, porphine-5,10,15,20-tetrakis(p-phenyltlimethylaminium) (PTPTMA), could stack along the molecular surface of DNA with the mode of long-range assembly to induce the formation of suprahelical species of DNA, resulting in strong RLS signals in the range of 450-510 nm. Under the excitation of 488 nm fight beam of argon ion laser source, single suprahelical species could be observed with the aid of a common microscope due to the strong scattered fight emitted by the suprahelical species. By capturing the RLS images of the single suprahelical species with a cooled charge coupled device (CCD) camera, and analyzing the RLS data, herein an RLS imaging method of DNA was proposed based on the linear relationship between the counts of suprahelical species in the detection focus plane and the concentration of DNA in nanograms. When 1.8 μmol/L PTPTMA was employed, both calf thymus DNA (ct DNA) and fish sperm DNA (fs DNA) in the range of 25-1100 ng/mL could be detected with the limits of detection lower than 25 ng/mL (3a). Sulfur diisocyanate is generated from a heterogeneous reaction of gaseous sulfur dichloride with silver cyanate and studied for the first time in the gas phase. Combined with quantum chemical calculations, the electronic structure is characterized by photoelectron spectroscopy (PES). Simultaneously, an investigation of the possible ionization and dissociation processes for the molecular cation is presented based on experimental soft ionization mass spectrometry. From the calculated bond-dissociation energies, the dissociation pathway is determined. S(NCO)2+ undergoes 1,3-sigmatropic rearrangement with a smaller barrier height (9.9 kcal mol(-1)) than the neutral counterpart. Thus, the 1,3-sigmatropic rearrangement is preferred for the molecular cation, and OCNCO+ and NS+ is produced by subsequent dissociation of the rearrangement product. A class of extended 2,5‐disubstituted‐1,3,4‐oxadiazoles R1‐C6H4‐{OC2N2}‐C6H4‐R2 (R1=R2=C10H21O 1 a , p‐C10H21O‐C6H4‐C?C 3 a , p‐CH3O‐C6H4‐C?C 3 b ; R1=C10H21O, R2=CH3O 1 b , (CH3)2N 1 c ; F 1 d ; R1=C10H21O‐C6H4‐C?C, R2=C10H21O 2 a , CH3O 2 b , (CH3)2N 2 c , F 2 d ) were prepared, and their liquid‐crystalline properties were examined. In CH2Cl2 solution, these compounds displayed a room‐temperature emission with λmax at 340–471 nm and quantum yields of 0.73–0.97. Compounds 1 d , 2 a – 2 d , and 3 a exhibited various thermotropic mesophases (monotropic, enantiotropic nematic/smectic), which were examined by polarized‐light optical microscopy and differential scanning calorimetry. Structure determination by a direct‐space approach using simulated annealing or parallel tempering of the powder X‐ray diffraction data revealed distinctive crystal‐packing arrangements for mesogenic molecules 2 b and 3 a , leading to different nematic mesophase behavior, with 2 b being monotropic and 3 a enantiotropic in the narrow temperature range of 200–210 °C. The structural transitions associated with these crystalline solids and their mesophases were studied by variable‐temperature X‐ray diffractometry. Nondestructive phase transitions (crystal‐to‐crystal, crystal‐to‐mesophase, mesophase‐to‐liquid) were observed in the diffractograms of 1 b, 1 d , 2 b, 2 d , and 3 a measured at 25–200 °C. Powder X‐ray diffraction and small‐angle X‐ray scattering data revealed that the structure of the annealed solid residue 2 b reverted to its original crystal/molecular packing when the isotropic liquid was cooled to room temperature. Structure–property relationships within these mesomorphic solids are discussed in the context of their molecular structures and intermolecular interactions. 相似文献

A novel capillary flow device has been developed and applied to study the orientation of worm‐like micelles, among other systems. Small‐angle X‐ray scattering (SAXS) data from micelles formed by a Pluronic block copolymer in aqueous salt solution provides evidence for the formation of worm‐like micelles, which align under flow. A transition from a rod‐like form factor to a less persistent conformation is observed under flow. Flow alignment of worm‐like micelles formed by the low molar mass amphiphile system cetyl pyridinium chloride+sodium salicylate is studied for comparative purposes. In completely stereospecific [4+2] cycloadditions, the perfluorinated selenocarbonyls 1 and 2 react both with trans‐trans‐2, 4‐hexadiene and cis‐trans‐2, 4‐hexadiene to yield 3, 6‐dihydro‐cis‐3, 6‐dimethyl‐2H‐selenapyrans 3 , 4a and 4b . The observed stereoselectivity leads to the conclusion, that the [4+2] cycloaddition of perfluorinated selenocarbonyls follows a concerted pathway. An identical mixture of isomers was isolated when using the precursor for 2 , trimethylstannyl (pentafluoroethyl)selane, which reacts with both 1, 3‐dienes over several weeks to form a mixture of syn‐2‐fluoro‐3, 6‐dihydro‐cis‐3, 6‐dimethyl‐2‐trifluoromethyl‐2H‐selenapyran ( 4a ) and anti‐2‐fluoro‐3, 6‐dihydro‐cis‐3, 6‐dimethyl‐2‐trifluoromethyl‐2H‐selenapyran ( 4b ) in the same ratio as found for 2 , thus proving the intermediate formation of Se=C(F)CF3 ( 2 ). Complex 2D NMR experiments were used to distinguish the isomers 4a and 4b and to assign the 1H, 13C and 19F NMR data of the selenaheterocycles. 相似文献

The formation of oxides at the surface of Fe–1.5%Mn and Fe–0.6%Mn binary alloys was investigated as a function of the conditions of the heat treatments. Both the influence of temperature and the atmosphere under which the experiments were performed were studied. The range of annealing temperatures was adjusted to 800°C. The atmosphere consisted of a mixture of N2–5%H2 and traces of water vapour, with different fixed dew points ranging from −10°C to −30°C. The state of the annealed surfaces was determined using in situ analytical devices attached to the annealing reactor in order to avoid surface contamination or the formation of native oxides after the experiments due to contact with air. The structure and composition of the surfaces were determined by reflection high-energy electron diffraction (RHEED) and electron spectroscopy (XPS, AES). Four novel organotin complexes of two types—[R2Sn(o‐SC6H4CO2)]6 (R=Me, 1 ?H2O; nBu, 2 ) and {[R2Sn(m‐CO2C6H4S)R2Sn(m‐SC6H4CO2)SnR2]O}2 (R=Me, 3 ; nBu, 4 )—have been prepared by treatment of o‐ or m‐mercaptobenzoic acid and the corresponding R2SnCl2 (R=Me, nBu) with sodium ethoxide in ethanol (95 %). All the complexes were characterized by elemental analysis, FT‐IR and NMR (1H, 13C, 119Sn) spectroscopy, TGA, and X‐ray crystallography diffraction analysis. The molecular structure analyses reveal that both 1 and 2 are hexanuclear macrocycles with hydrophobic “pseudo‐cage” structures, while 3 and 4 are hexanuclear macrocycles with double‐cavity structures. Furthermore, the supramolecular structure analyses show that looser and more intriguing supramolecular infrastructures were also found in complexes 1 – 4 , which exist either as one‐dimensional chains of rings or as two‐dimensional networks assembled from the organometallic subunits through intermolecular C? H???S weak hydrogen bonds (WHBs) and π–π interactions. 相似文献

Two-dimensional (2-D) Blue Native/SDS gel electrophoresis combines a first-dimensional separation of monomeric and multimeric proteins in their native state with a second denaturing dimension. These high-resolution 2-D gels aim at identifying multiprotein complexes with respect to their subunit composition. We applied this method for the first time to analyze two human platelet subproteomes: the cytosolic and the microsomal membrane protein fraction. Solubilization of platelet membrane proteins was achieved with the nondenaturing detergent n-dodecyl-beta-D-maltoside. To validate native solubilization conditions, we demonstrated the correct assembly of the Na,K-ATPase, a functional multimeric transmembrane protein, when expressed in Xenopus oocytes. We identified 63 platelet proteins after in-gel tryptic digestion of 58 selected protein spots and liquid chromatography-coupled tandem mass spectrometry. Nine proteins were detected for the first time in platelets by a proteomic approach. We also show that this technology efficiently resolves several known membrane and cytosolic multiprotein complexes. Blue Native/SDS gel electrophoresis is thus a valuable procedure to analyze specific platelet subproteomes, like the membrane(-bound) protein fraction, by mass spectrometry and immunoblotting and could be relevant for the study of protein-protein interactions generated following platelet activation. 相似文献
