A novel method for the determination of Pb by on-line cloud point extraction coupled to inductively coupled plasma optical emission spectrometry with ultrasonic nebulization (USN–ICP OES) is presented. The cloud point system was formed in the presence of non-ionic micelles of polyethyleneglycolmono-p-nonylphenylether (PONPE 7.5) and was retained in a minicolumn filled with particles of PTFE. Since the micelles of PONPE were able to extract Pb from the solutions, the use of a complexing reagent was not necessary. Afterwards, the surfactant-rich phase containing the analyte was removed from the minicolumn with nitric acid and introduced into the ultrasonic nebulizer. A total enhancement factor of 150 was obtained for a preconcentration time of 3.3 min, with respect to the direct determination of Pb by conventional ICP OES. The values of the detection (3σ) and quantification (10σ) limits for the preconcentration of 10 mL of sample solution were 0.09 µg L− 1 and 0.2 µg L− 1 respectively. The precision, expressed as the relative standard deviation (RSD), for 10 replicate determinations at 5.0 µg L− 1 Pb level was 6.0%. Verification of the accuracy was carried out by analysis of two certified reference materials (NIST SRM 1640e and VKI QC Metal LL1). The method was successfully applied to the determination of Pb in drinking water samples.  相似文献

In this study, octadecyltrimethylammonium surfactant was immobilized onto a cation exchange membrane for the application in solid-phase extraction of phenolic compounds. The results indicate that an HCl prewashing step and the use of hydroxide (or methoxide) counter ion could greatly improve the immobilized surfactant capacity. Through elemental and thermogravimetric analyses, the resulted immobilization percentage on the membrane (compared to membrane ion exchange capacity) was about 50, 100, and 150%, respectively, for the feed surfactant amount of 150, 2000, and 5000 μmol (volume = 20 mL). Phenol, 4-nitrophenol, 2,4-dimethylphenol, 2,4-dichlorophenol, 4-chloro-3-methylphenol, and bisphenol A were the tested compounds in a breakthrough volume experiment. The order of the obtained breakthrough volume values is similar to that of Kow values of the phenolic compounds. In the solid-phase extraction process from a feed mixture of 0.1 ppm for 4-nitrophenol, 2,4-dichlorophenol, and 4-chloro-3-methylphenol, high concentration factors and almost complete recoveries were achieved. Moreover, by increasing the membrane volume, a larger sample volume could be processed without any deterioration in performance.  相似文献

The first facile Fmoc-based synthetic procedure for peptide thioacids was developed. Successful application of the resulting thioacids to sequential native chemical ligation (NCL) in the N to C direction was achieved. A novel procedure for the determination of nine selected fungicides (metalaxyl-M, azoxystrobin, myclobutanil, flusilazole, penconazole, tebuconazole, propiconazole, diniconazole and difenoconazole) in wine samples is presented. Sample enrichment and purification is simultaneously performed using mixed-mode, anion exchange and reversed-phase, OASIS MAX solid-phase extraction (SPE) cartridges. Analytes were determined by liquid chromatography coupled to tandem mass spectrometry using atmospheric pressure electrospray ionization (LC-ESI-MS/MS). Parameters affecting the chromatographic determination and the extraction-purification processes were thoroughly investigated. Under optimized conditions, 10 mL of wine were firstly diluted 1:1 with ultrapure water and then passed through the mixed-mode SPE cartridge at a flow of ca. 5 mLmin(-1). After a washing step with 5 mL of an aqueous NH(4)OH solution (5%, w:v), analytes were recovered with just 1 mL of methanol and injected in the LC-MS/MS system without any additional purification. The selective extraction process avoided significant changes in the ionization efficiency for red and white wine extracts in comparison with pure standards in methanol. Performance of the method was good in terms of precision (RSDs<11%) and accuracy (absolute recoveries>72%, determined against pure standards in methanol) reporting method LOQs in the range of 0.01-0.79 ngmL(-1) for target compounds, which are far below the EU maxima residue levels (MRLs) for fungicides in vinification grapes and wine. Several commercial wines from different geographic areas in Spain were analyzed. In most samples, metalaxyl-M and azoxystrobin were found at concentrations up to several ngmL(-1).  相似文献

A simple sugaring-out assisted liquid-liquid extraction method combined with high-performance liquid-chromatography with fluorescence detection (HPLC-FL) was developed for the extraction and determination of sulfonamides in honey. Sample preparation consisted of acid hydrolysis to release sugar-bound sulfonamides. After derivatization with fluorescamine, the derivatives were partitioned into the organic layer under the honey (sugar)/water/acetonitrile system. The clear organic extract obtained by centrifugation could be injected into the HPLC system either directly or after dilution. Linearity was obtained with the coefficient of determination (R(2)) higher than 0.998 from 2 to 200 ng/mL. Under the optimal conditions, recoveries were determined for honey fortified at three levels (5, 20, and 100 ng/g) were 80.9-99.6% with coefficients of variation of 0.3-4.4%. Limits of detection for the sulfonamides studied were found to range from 0.6 to 0.9 ng/g.  相似文献

This study presents an efficient strategy based on microwave‐assisted extraction (MAE), HPLC‐DAD‐MS/MS and high‐speed counter‐current chromatography (HSCCC) for the rapid extraction, identification, separation and purification of active components from the traditional Chinese medicine Fructus Aurantii Immaturus. An LC‐DAD‐MS/MS method was applied for the screening and structural identification of main components in crude extract, and five components were preliminarily identified as neoeriocitrin, narirutin, naringin, hesperidin and neohesperidin according to their UV and mass spectra. An efficient MAE method for the extraction of the three most abundant components (narirutin, naringin and neohesperidin) was optimized by the combination of univariate and multivariate approaches. The crude extract was then separated and purified by HSCCC and a total of 61.6 mg of narirutin, 207.3 mg of naringin and 159.5 mg of neohesperidin at high purities of 98.1, 97.2 and 99.5%, respectively, were obtained from 1.42 g of crude extract. For the first time, an automatic sample pre-treatment/detection method is proposed for the multiclass determination of UV filters (namely, benzophenone-3, ethylhexylmetoxycinnamate, butylmethoxydibenzoylmethane and homosalate) in environmental samples. The new methodology comprises in-line solid-phase extraction (SPE) of the target analytes by exploiting the bead injection (BI) concept in a mesofluidic lab-on-valve (LOV) format, with subsequent determination by liquid chromatography (LC). The proposed microanalytical system, using a multisyringe burette as propulsion unit, automatically performed the overall SPE steps, including the renewal of the sorbent in each analytical cycle to prevent sample cross-contamination and the post-extraction adjustment of the eluate composition to prevent chromatographic band broadening effects. In order to expedite the LC separation, a C18 monolithic column was applied and an accelerated isocratic elution was carried out by using a cationic surfactant as mobile phase additive. The LOV-BI-LC method was proven reliable for handling and analysis of complex matrices, e.g., spiked swimming pool water and seawater, with limits of detection ranging between 0.45 and 3.2 μg L−1 for 9 mL sample volume. Linear calibration was attained up to 160 μg L−1 for homosalate and up to 35 μg L−1 for the other target analytes, with good reproducibility (RSD < 13%, for 5 different SPE columns). The hyphenated scheme is able to process a given sample simultaneously and within the same time frame than the chromatographic separation/determination of the formerly pre-treated sample, providing concentration values every 9 min. Hence, the sample throughput was enhanced up to 33 times when compared with previously reported off-line SPE methods. A drastic reduction in reagent consumption and effluent production was also attained, contributing to the development of an environment-friendly analyzer.  相似文献

Polyurethane (PU) foams were applied for stir bar sorptive extraction of five triazinic metabolites (desethyl-2-hydroxyatrazine, desisopropylatrazine, desethylatrazine, 2-hydroxyatrazine and desethylterbuthylazine) in water matrices, followed by liquid desorption and high performance liquid chromatography with diode array detection (SBSE(PU)-LD/HPLC-DAD). The optimum conditions for SBSE(PU)-LD were 5 h of extraction (1000 rpm) and 5% (v/v) of methanol for the analysis of desethyl-2-hydroxyatrazine and 2-hydroxyatrazine, 15% (w/v) of sodium chloride for the remaining compounds and acetonitrile as back-extraction solvent (5 mL) under ultrasonic treatment (60 min). The methodology provided recoveries up to 26.3%, remarkable precision (RSD < 2.4%), excellent linear dynamic ranges between 5.0 and 122.1 μg/L (r2 > 0.9993) and convenient detection limits (0.4-1.3 μg/L). The proposed method was applied in the analysis of triazinic metabolites in tap, river and ground waters, with remarkable performance and negligible matrix effects. The comparison of the recoveries obtained by PU and commercial stir bars was also performed, where the yields achieved with the former were up to ten times higher proving that PU is appropriate for analysis at trace level of this type of polar compounds in water matrices.  相似文献

A stir bar microextraction (SBSE) procedure for the determination of seven strobilurin fungicides in fruit samples using liquid chromatography (LC) and diode array detection (DAD) has been developed. The samples were sonicated in the presence of ethanol before submitting the extracts to SBSE. The incorporation of drazoxolon as an internal standard before SBSE allowed calibration without the need to use the standard additions method. Under the optimized conditions, detection limits were in the 0.3–2 ng g−1 range, corresponding to trifloxystrobin and metominostrobin, respectively. The SBSE–LC–DAD procedure showed good repeatability (RSD below 11% in all cases) and provided recoveries of 80–105% from spiked samples. The method was applied to fifteen fruit samples, and low levels of pyraclostrobin and trifloxystrobin were found in two of them.  相似文献

A fast and flexible multi-residue procedure for effective extraction of benzimidazole fungicides and the related transformation products in various raw agricultural commodities is developed for direct and simultaneous analysis by liquid chromatography–tandem mass spectrometry (LC–MS/MS). The new sample preparation method, introduced to allow direct extraction of the labile fungicides as the intact forms in complex matrices, is achieved using a conservative homogenizing extraction and multifunction adsorption cleanup (CHEMAC), which basically involves salting-out partitioning/extraction with acetate-buffered acetonitrile at low-temperature and sequential rapid solid-phase dispersive cleanup with a ternary sorbent mixture. The CHEMAC procedure was optimized and further modified by incorporating several pretreatment variables influencing sample stability and process efficiency, such as pH, temperature, salt and sorbent utilized. By using CHEMAC, a noteworthy improvement in extraction recoveries was obtained for the problematic fungicides, while no significant differential matrix effects were detected on the LC–MS/MS analysis in all 9 matrices. The in-source fragmentation of benomyl occurred but caused no cross-talk interference in multi-component analysis. Thus the CHEMAC-based LC–MS/MS strategy can serve as an attractive approach to satisfactory overall process efficiencies (70–92%) with acceptable repeatability (relative standard deviations below 16%) for all the analyte–matrix combinations. Mean accuracies were obtained within the range of 70–110% at fortified levels of 1–500 ng/g, with intra-day and inter-day variations less than 15 and 20%, respectively. The successful practical application of the proposed method to real samples has also been demonstrated.  相似文献
