Seed oils of 12 Salvia species collected from different regions in Turkey (S. Albimaculata Hedge & Hub.-Mor., S. Candidisima Vahl., S. Cedronella Boiss., S. Cryptantha Montbret & Aucher ex Bentham, S. Forskahlei L., S. Fruticosa Miller (Sin. S. Triloba L. Fil), S. Halophila Hedge, S. Hypargeia Fisch. & Mey., S. Sclarea L., S. Tomentosa Miller, S. Tchihatcheffii (Fisch. & Mey.) Boiss., S. Virgata Jacq.) were obtained by Soxhlet apparatus using hexane. The oil yields were found to be between 2.0% and 20.9%. Fatty acids in the oils were converted to methyl esters and determined by GC/MS in methyl ester form. The main fatty acid components of S. halophila, S. Hypargeia, and S. Sclarea are unsaturated oleic, linoleic, and linolenic acids. In others except S. Candidissima, the dominant acids are oleic, linoleic and palmitic acids.  相似文献

Effect of environmental factors and carbohydrate on gellan gum production   总被引:3,自引:0,他引:3  
Submerged culture fermentation studies were carried out in batch mode for optimizing the environmental parameters and carbon source requirement by Pseudomonas elodea for the production of gellan gum. The maximum production of gellan gum was obtained with 16-h-old culture and 8% inoculum at 30°C and pH 7.0 after 52 h of incubation (6.0 g/L). Of the various carbon sources tested, 2% sucrose, glucose, and soluble starch yielded considerably high amounts of gellan. Studies on the concentration of various carbohydrates on gellan gum production indicated that the optimum concentration of glucose and starch was 3%, whereas for sucrose it was 4%. The addition of glucose in the medium above 3% had a detrimental effect on gellan yield. The investigation of intermediate two-step addition of glucose under identical conditions of fermentation showed an enhanced production of gellan (8.12 g/L) as compared with the control (6.0 g/L). To optimize the recovery of gellan from fermented broth, different solvents were tested for precipitation of gellan gum. Among the various solvents tested, tetrahydrofuran gave better recovery of gellan (82%) as compared with the conventional solvent isopropanol (49%).  相似文献

Two methods of protein extraction for soybean seeds were evaluated in terms of preservation of the metal ions bound to proteins after the extraction and separation procedures. The proteins were firstly separated according to their molar masses by polyacrylamide gel electrophoresis. Then, the protein bands were mapped by synchrotron radiation X-ray fluorescence in order to establish which metal ions were present in each one. Finally, some mapped protein bands were decomposed by microwave-assisted combustion and Ca, Cu, K, Mg, Mn, and Zn were quantified by inductively coupled plasma mass spectrometry or inductively coupled plasma optical emission spectrometry. The extraction methods studied were Method A (based on the treatment of ground soybean seeds with hexane and their extraction with Tris–HCl and β-mercaptoethanol) and Method B (based on the treatment of ground soybean seeds with petroleum ether and their extraction with Tris–HCl, dithiothreitol, phenylmethanesulfonyl fluoride, sodium dodecyl sulfate and potassium chloride). The best method was Method B, in which a 78% higher extraction efficiency was obtained when compared to Method A. Additionally, the metal-protein interactions were more appropriately preserved when Method B was applied, where the most affected ions were those that are bound weakly to proteins, such as Ca, K, and Mg.  相似文献

The uniform-sized spherical molecularly imprinted polymers were successfully prepared through molecular imprinting technology by two-step seed swelling and mini-emulsion polymerization in the aqueous condition using quinine as template molecules and methacrylic acid (MAA) as functional monomer. The polymers were characterized by IR spectra, thermal-weight analysis, scanning electron microscope and laser particle size analysis. The properties of imprinted polymers were investigated in different organic phases and aqueous media. In the organic media, results suggested that polar interactions (hydrogen bonding, ionic interactions) between acidic monomer/polymer and template molecules are mainly responsible for the binding and recognition; whereas in the aqueous medium, a considerable recognition effect was also obtained where the ionic (electrostatic) interaction and hydrophobic interaction play an important role. Eupatilin (5,7-dihydroxy-3',4',6-trimethoxy flavone) is an active ingredient of an ethanol extract of Artemisia asiatica (DA-9601) that is used in the treatment of gastritis. In vitro and in vivo metabolism of eupatilin in the rats has been studied by LC-electrospray mass spectrometry. Rat liver microsomal incubation of eupatilin in the presence of NADPH and UDPGA resulted in the formation of four metabolites (M1-M4). M1, M2, M3 and M4 were tentatively identified as 3'- or 4'-O-demethyl-eupatilin glucuronide, eupatilin glucuronide, 6-O-demethyleupatilin and 3'- or 4'-O-demethyl-eupatilin, respectively. Those metabolites from in vitro study were also characterized in bile, plasma or urine samples after an intravenous administration of eupatilin to rats. In rat bile, plasma and urine samples, eupatilin glucuronide (M2) was a major metabolite, whereas M3, M4 and M4 glucuronide (M1) were the minor metabolites.  相似文献

The oxime of artemisia ketone is smoothly converted to the pyrazole derivative by sodium nitrite and acetic acid in CHCl3. The structure of the pyrazole was found by x-ray structural analysis. Absolute stereochemistry of dihydroarteannuin B 5 obtained by the reduction of arteannuin B 3 with Ni2B, NaBH4 or CdCl2-Mg-MeOH-H2O has been established by 2D NMR and single crystal X-ray diffraction studies. The acetogenins are strongly bioactive natural compounds present in the bark, roots, leaves, and seeds of manyAnnonaceae plants. They are modified fatty acids and their cytotoxicities have been determined for different biological models including the in vitro growth inhibition of several human cancer cell lines. Very low acetogenin yield (< 0.1 g%) has been found previously in native phytobiomass, and we have now investigated the nonpredatory exploitation of the seeds as acetogenin sources characterizing the seed triacylglycerols (dominant fraction; > 90% of the whole lipid extracts) as potential valuable by-products.  相似文献
