A previously elaborated capillary electrophoresis (CE) method used for the determination of glyphosate and aminomethylphosphonic acid (AMPA) was slightly modified in order to improve the sensitivity. However, detection limits attained (5 μg mL−1 for glyphosate and 4 μg mL−1 for AMPA) were still not satisfactory for analytical purposes, thus the addition of a preconcentration step before the CE analysis was proposed. AMBERLITE®IRA-900, a strong anion-exchange resin, was used to preconcentrate both analytes in environmental aqueous samples. The experimental conditions optimised in a previous work were readapted, by decreasing the eluent concentration due to CE limitations. Satisfactory results were attained when spiked ultrapure water was applied, with recoveries from 84 to 87% for glyphosate (R.S.D. < 6%) and from 85 to 98% for AMPA (R.S.D. < 5%). Enrichment factors up to 65 were achieved with this system, allowing the determination of 85 ng mL−1 of glyphosate and 60 ng mL−1 of AMPA. The extraction efficiency varied when four different natural water samples of varying conductivity were applied. Especially the strong dependence on ion concentration in samples on AMPA recovery was found. For glyphosate, good recoveries (86-99%) were obtained for samples of low and medium conductivity (0-800 μS). The effect of sample salt content on extraction efficiency was studied and a linear relationship could be established for AMPA (r2 = 0.996). An important improvement on recoveries was observed when lower volumes of sample were treated.A HPLC method with UV-vis detection and pre-column derivatisation with p-toluensulphonyl chloride was compared to the CE method. No significant differences in results were found when t- and F-statistical tests were applied.  相似文献

A method for the accurate determination of Cd and Pb isotope compositions in sediment samples is presented. Separation of Cd and Pb was designed by using an anionic exchange chromatographic procedure. Measurements of Cd isotopic compositions were carried out by multi-collector inductively coupled plasma mass spectrometer (MC-ICPMS), by using standard-sample bracketing technology for mass bias correction and Pb isotopic ratios were determined by thermal ionization mass spectrometry (TIMS). The factors that affect the accurate and precise Cd isotope compositions analysis, such as instrumental mass fractionation and isobaric interferences, were carefully evaluated and corrected. The Cd isotopic results were reported relative to an internal Cd solution and expressed as the δ114/110Cd. Five Cd reference solutions and one Pb standard were repeatedly measured in order to assess the accuracy of the measurements. Uncertainties obtained were estimated to be lesser than 0.11‰ (2s) for the δ114/110Cd value. Analytical uncertainties in 2s for Pb isotopic ratios were better than 0.5‰. The method has been successfully applied to the investigation of Cd and Pb isotope compositions in sediment samples collected from North River in south China.  相似文献

Human serum albumin (HSA) was explored for use as a stationary phase and ligand in affinity microcolumns for the ultrafast extraction of free drug fractions and the use of this information for the analysis of drug–protein binding. Warfarin, imipramine, and ibuprofen were used as model analytes in this study. It was found that greater than 95% extraction of all these drugs could be achieved in as little as 250 ms on HSA microcolumns. The retained drug fraction was then eluted from the same column under isocratic conditions, giving elution in less than 40 s when working at 4.5 mL/min. The chromatographic behavior of this system gave a good fit with that predicted by computer simulations based on a reversible, saturable model for the binding of an injected drug with immobilized HSA. The free fractions measured by this method were found to be comparable to those determined by ultrafiltration, and equilibrium constants estimated by this approach gave good agreement with literature values. Advantages of this method include its speed and the relatively low cost of microcolumns that contain HSA. The ability of HSA to bind many types of drugs also creates the possibility of using the same affinity microcolumn to study and measure the free fractions for a variety of pharmaceutical agents. These properties make this technique appealing for use in drug-binding studies and in the high-throughput screening of new drug candidates.  相似文献

Organic–inorganic anion-exchange membranes based on poly(vinylidene fluoride)/SiO2 were prepared and quaternary ammonium groups were introduced by the reaction of the epoxy groups of glycidyl methacrylate with trimethylamine. Various membranes were prepared with different weight fraction of nano-sized SiO2 particles. The effect of silica content on the performance of these anion-exchange membranes was extensively characterized in terms of transport properties (TSP). The hydrophilic nature, swelling and TSP of these anion-exchange membranes were dependent on nanoparticles content in the membrane matrix. Higher TSP values of these membranes were obtained, which increased with increasing silica content. The hydrophilic property of the membranes was improved in presence of silica. Results of the effect of silica content on the membrane conductivity were confirmed by the analysis of phenomenological coefficients using non-equilibrium thermodynamic principles. It can be concluded that these anion-exchange membranes prepared with 2% loading with nanoparticles exhibited better transport properties, which may be used for their application in electro-driven separation or for other electrochemical processes.  相似文献

A new specific stationary phase based on poly(1-allylimidazole)-grafted silica has been synthesized and characterized, by infrared spectra, elemental analysis, thermogravimetric analysis and X-ray photoelectron spectroscopy. The results of test showed that poly(1-allylimidazole) can effectively mask the residual silanol groups and reduce the adverse effect of residual silanol. Using this stationary phase, phenol compounds, aniline compounds, and polycyclic aromatic hydrocarbons were successfully separated with symmetric peak shapes in the reversed-phase chromatography. Inorganic anions (IO3, BrO3, Br, NO3, I, SCN) were also separated completely in the anion-exchange chromatography using sodium chloride solution as the mobile phase. The effects of pH and the concentration of eluent on the separation of inorganic anions were studied. The separation mechanism appears to involve the mixed interactions of hydrogen bonding, hydrophobic, π–π, electrostatic, and anion-exchange interactions.  相似文献

Anion-exchange (AEX) columns were prepared by on-column polymerization of acrylates and methacrylates containing tertiary amino or quaternary ammonium groups on monolithic silica in a fused silica capillary modified with anchor groups. The columns provided a plate height (H) of less than 10 μm at optimum linear velocity (u) with keeping their high permeability (K = 9–12 × 10−14 m2). Among seven kinds of AEX columns, a monolithic silica column modified with poly(2-hydroxy-3-(4-methylpiperazin-1-yl)propyl methacrylates) (HMPMA) showed larger retentions and better selectivities for nucleotides and inorganic anions than the others. The HMPMA column of 410 mm length produced 42 000–55 000 theoretical plates (N) at a linear velocity of 0.97 mm/s with a backpressure of 3.8 MPa. The same column could be employed for a fast separation of inorganic anions in 1.8 min at a linear velocity of 5.3 mm/s with a backpressure of 20 MPa. In terms of van Deemter plot and separation impedance, the HMPMA column showed higher performance than a conventional particle-packed AEX column. The HMPMA column showed good recovery of a protein, trypsin inhibitor, and it was applied to the separation of proteins and tryptic digest of bovine serum albumin (BSA) in a gradient elution, to provide better separation compared to a conventional particle-packed AEX column.  相似文献

Microcolumns have a stacked structure composed of an electron emitter, electron lens (source lens), einzel lens, and a deflector manufactured using a micro electro-mechanical system process. The electrons emitted from the tungsten field emitter mostly pass through the aperture holes. However, other electrons fail to pass through because of collisions around the aperture hole. We used Raman scattering measurements and X-ray photoelectron spectroscopy analyses to investigate the influence of electron beam bombardment on a Si electron lens irradiated by acceleration voltages of 0, 20, and 30 keV. We confirmed that the crystallinity was degraded, and carbon-related contamination was detected at the surface and edge of the aperture hole of the Si electron lens after electron bombardment for 24 h. Carbon-related contamination on the surface of the Si electron lens was verified by analyzing the Raman spectra of the carbon-deposited Si substrate using DC sputtering and a carbon rod sample. We report the crystallinity and the origin of the carbon-related contamination of electron Si lenses after electron beam bombardment by non-destructive Raman scattering and XPS analysis methods.  相似文献

Several prototypes of multi-modal ligands suitable for the capture of negatively charged proteins from high conductivity (28 mS/cm) mobile phases were coupled to Sepharose 6 Fast Flow. These new prototypes of multi-modal anion-exchangers were found by screening a diverse library of multi-modal ligands and selecting anion-exchangers resulting in elution of test proteins at high ionic strength. Candidates were then tested with respect to breakthrough capacity of BSA in a buffer adjusted to a high conductivity (20 mM Piperazine and 0.25 M NaCl, pH 6.0). The recovery of BSA was also tested with a salt step (from 0.25 to 2.0 M NaCl using 20 mM Piperazine as buffer, pH 6.0) or with a pH-step to pH 4.0. We have found that non-aromatic multi-modal anion-exchange ligands based on primary or secondary amines (or both) are optimal for the capture of proteins at high salt conditions. Furthermore, these new multi-modal anion-exchange ligands have been designed to take advantage not only of electrostatic but also hydrogen bond interactions. This has been accomplished through modification of the ligands by the introduction of hydroxyl groups in the proximity of the ionic group. Experimental evidence on the importance of the relative position of the hydroxyl groups on the ligand in order to improve the breakthrough capacity of BSA has been found. Compared to strong anion-exchangers such as Q Sepharose Fast Flow the new multi-modal weak anion-exchangers have breakthrough capacities of BSA at mobile phases of 28 mS/cm and pH 6.0 that are 20-30 times higher. The new multi-modal anion-exchangers can also be used at normal anion-exchange conditions and with either a salt step or a pH-step to acidic pH can accomplish the elution of proteins. In addition, the functional performance of the new anion-exchangers was found to be intact after treatment in 1.0 M sodium hydroxide solution for 1 week. A number of multi-modal anion-exchange ligands based on aromatic amines exhibiting high breakthrough capacity of BSA have been found. With these ligands recovery was often found to be low due to strong non-electrostatic interactions. However, for phenol derived anion-exchange media the recovery can be improved by desorption at high pH.  相似文献

The local structures of Cl- and Br- in anion-exchange resins have been studied by X-ray absorption fine structure (XAFS), and separation selectivity is discussed on the basis of results. When two different anion-exchange resins having trimethylammonium and dimethylammonium groups as anion-exchange groups are employed for ion-exchange experiments, slightly higher Br- selectivity has been obtained with the former. XAFS has indicated that the average hydration numbers for a given anion is not affected by the structure of the ion-exchange group, but that the extent of ion-association between the anion and the ion-exchange groups depends on the type of the ion-exchange group. Shorter interaction distance (and in turn stronger ion-association) has been confirmed for the dimethylammonium-type resin, and is consistent with lower Br- selectivity of this resin.  相似文献
