We synthesized a novel 1,3,5-substituted triethylbenzene derivative with a benzimidazole moiety as a binding and signalling subunit. The triethylbenzene platform was judiciously incorporated into the receptor design such that all the fluorophores are in close proximity to each other in order to facilitate stacking. The advantage of this approach lies in the fact that the receptor will offer a dual channel emission for the multiple ratiometric determination of metal ions. The sensor was tested in a CH3CN/H2O (7:3, v/v, pH 7.1) HEPES buffered solution and successfully developed for the ratiometric and simultaneous estimation of Fe3+ and Cu2+ ions in aqueous samples.  相似文献

The cannabinoid CB1 receptor is a class A G protein‐coupled receptor (GPCR) that is the most widely expressed GPCR in the brain. Many GPCRs contain allosteric binding sites for endogenous and/or synthetic ligands, which are topographically distinct from the agonist‐binding site that is known as the orthosteric site. While both endogenous and synthetic ligands that act at the CB1 orthosteric site have been known for some time, compounds that act at a CB1 allosteric site have only recently been discovered. The most studied of these is 5‐chloro‐3‐ethyl‐1H‐indole‐2‐carboxylic acid [2‐(4‐piperidin‐1‐ylphenyl)ethyl]amide (Org27569). Because allosteric ligands are thought to act through conformational changes in the receptor that are transmitted from the allosteric to the orthosteric site, computational studies of the structural and dynamic interactions of Org27569 with the CB1 receptor are crucial to achieve a molecular level understanding of the basis of action of this important new class of compounds. To date, such computational studies have not been possible due to the lack of a complete set of molecular mechanics force field parameters for Org27569. Here, we present the development of missing CHARMM force field parameters for Org27569 using previously published methods and the validation and application of these new parameters using normal mode analysis and molecular dynamics simulations combined with experimental infrared measurements. © 2011 Wiley Periodicals, Inc. Six new compounds: [M(dps)(dca)2] (1, M = Cd; 2, M = Fe), [Cu(dps)(dca)2(CH3OH)2] (3), [M(dps)2(SCN)2(H2O)2] (4, M = Cd; 5, M = Fe), [Cu(dps)4(SCN)2] (6), (dps = 4,4′-dipyridyl sulfide, dca = dicyanamide) were synthesized, characterized by FT-IR, element analysis and TG. Their structures were determined by single crystal X-ray diffraction. According to the structure analysis, compounds 1 and 2 are three-dimensional microporous frameworks constructed by Cu-dps-Cu and Cu-dca-Cu bridges; compound 3 shows a zigzag chain constructed by a Cu-dps-Cu bridge; compounds 4 and 5 show double-stranded chains constructed by a Cu-(dps)2-Cu bridge while thiocyanate anions act as terminal groups; compound 6 is a mononuclear compound. Moreover, compound 1 shows visible yellow luminescence under UV light irradiation and such phenomenon is mainly attributed to inter-ligand transitions.  相似文献

The membrane‐bound tumor‐associated glycoprotein MUC1 is aberrantly glycosylated in cancer cells compared with normal cells, and is therefore considered an attractive target for cancer immunotherapy. However, tumor‐associated glycopeptides from MUC1 do not elicit a sufficiently robust immune response. Therefore, antitumor vaccines were developed, which consist of MUC1 glycopeptides as the B epitopes and immune‐stimulating toll‐like receptor 2 (TLR 2) lipopeptide ligands. These fully synthetic vaccine candidates were prepared by solid‐phase synthesis of the MUC1 glycopeptides. The Pam3Cys lipopeptide, also synthesized on solid‐phase, was C‐terminally coupled to oligovalent lysine cores, which N‐terminally incorporate O‐propargyl oligoethylene glycol acyl side chains. The MUC1 glycopeptides and lipopeptide lysine constructs were then conjugated by click chemistry to give oligovalent synthetic vaccines. Oligovalent glycopeptide–lipopeptide conjugates are considered more immunogenic than their monovalent analogues.  相似文献

This paper reports an automated analytical method for rapid determination of plutonium isotopes (239Pu and 240Pu) in environmental solid extracts. Anion exchange chromatographic columns were incorporated in a sequential injection (SI) system to undertake the automated separation of plutonium from matrix and interfering elements. The analytical results most distinctly demonstrated that the crosslinkage of the anion exchanger is a key parameter controlling the separation efficiency. AG 1-×4 type resin was selected as the most suitable sorbent material for analyte separation. Investigation of column size effect upon the separation efficiency revealed that small-sized (2 mL) columns sufficed to handle up to 50 g of environmental soil samples. Under the optimum conditions, chemical yields of plutonium exceeded 90% and the decontamination factors for uranium, thorium and lead ranged from 103 to 104. The determination of plutonium isotopes in three standard/certified reference materials (IAEA-375 soil, IAEA-135 sediment and NIST-4359 seaweed) and two reference samples (Irish Sea sediment and Danish soil) revealed a good agreement with reference/certified values. The SI column-separation method is straightforward and less labor intensive as compared with batch-wise anion exchange chromatographic procedures. Besides, the automated method features low consumption of ion-exchanger and reagents for column washing and elution, with the consequent decrease in the generation of acidic waste, thus bearing green chemical credentials.  相似文献
