NMR spectroscopy plays an important part in the determination of the structures of organic compounds. The parameters of importance here are the chemical shifts of the 1H and 13C nuclei and the spin-spin interactions both between 1H nuclei and between 1H and 13C nuclei. Couplings between 13C nuclei were almost completely neglected until a few years ago, since they were extremely difficult to observe because of the low natural abundance of 13C. However, it is these couplings which afford information directly on the carbon-carbon connectivities in the molecule. It is now possible to use a special NMR pulse sequence to make these couplings more readily visible: the result of using this sequence is a 13C-NMR spectrum from which the carbon skeleton concerned can be directly read off. Two-dimensional spectra in particular are very easy to evaluate. The pulse sequence involved, which bears the somewhat puzzling name INADEQUATE, produces double-quantum coherences from which the NMR signals of the coupled carbon nuclei can be obtained. In this article the principle of double-quantum coherence is described and a number of examples for the application of the INADEQUATE pulse sequences to problems in synthetic organic chemistry, biosynthesis and natural products chemistry are presented; in addition, the possibility of applying the INADEQUATE method to other nuclei is considered. 相似文献

Chromatography with supercritical fluids unites the features of both gas chromatography and liquid chromatography, yet retains special characteristics of its own. The diffusion coefficient and particularly the viscosity of fluid phases may approach values for low-pressure gases, while the solvent power may be similar to that of liquids. However, with supercritical fluids it is possible to control chromatographic separations very effectively by pressure programming, since the solubility increases with increasing density. Temperature programming, on the other hand, can have the opposite effect to that in gas- or liquid-chromatography since the density decreases with increasing temperature at a given pressure. Supercritical fluid chromatography is primarily of interest for the separation of higher molecular weight compounds. The efficiency of this method of separation is demonstrated on several homologous series. Thus, a styrene oligomer with nominal Mw=2200 can be resolved by a pressure and temperature program into 40 species. 相似文献

Herein, we discuss the analyses and quantification of the different components in porphyrin mixtures, prepared from p-anisaldehyde, p-tolualdehyde, and 5-(4-bromophenyl)-dipyrromethane with acid catalysis, using NMR and HPLC. The advantages and disadvantages of these analytical methods are emphasized. Due to the similar size of a bromine atom and a methyl group it was possible to grow crystals suitable for X-ray crystallographic studies from a mixture of porphyrins, where the 4-position of the meso-phenyl rings was either substituted with methyl groups or bromine atoms. We also show that X-ray studies are inferior to NMR analysis for determining the components in a porphyrin mixture. 相似文献

Our 1D + 1D model of DMFC reveals a new effect. At infinitely small total current in the cell, near the channel inlet forms a “bridge”, a narrow region with finite local current density. The bridge short-circuits the electrodes, thus reducing cell open-circuit voltage. In our previous work the effect is described for the case of equal methanol λa and oxygen λc stoichiometries. In this Letter, we analyze the general case of arbitrary λa and λc. In the case of λa > λc current may occupy finite domain of the cell surface. Asymptotic solution for the case of λa λc shows, that the size of this domain is proportional to oxygen stoichiometry. In the opposite limit of λa λc local current exponentially decreases with the distance along the channel. Asymptotic solutions suggest that the bridge forms regardless of the relationship between λa and λc. In all cases local current density in the bridge increases with the rate of methanol crossover and decreases with the growth of the “rate-determining” stoichiometry. Summary Analytical peak-shape equations were derived for first-order reversible reactions occurring in a chromatographic reactor by
treating the reversible reactions as consecutive reactions with alternating products. The results of the analytical peak-shape
equations were compared with those from a numerical solution of the partial differential equation system modeling the chromatographic
reactor. For small to medium conversions the correspondence was found to be sufficiently close to enable substitution of the
numerical solution in fitting procedures for the determination of rate constants.

Roasting of Coffea arabica L. seeds gives rise to chemical reactions that produce more than 800 compounds, some being responsible for the desired organoleptic properties for which the beverage called “coffee” is known. In the industry, the “roasting profile,” that is, the times and temperatures applied, is key to influence the composition of roasted coffee beans and the flavour of the beverage made from them. The impact of roasting on the chemical composition of coffee has been the subject of numerous studies, including by nuclear magnetic resonance (NMR) spectroscopy. However, the roasting equipment and profiles applied in these studies are often far from real industrial conditions. In this work, the effects of two critical technological parameters of the roasting process, namely, the “development time” (the period of time after the “first crack,” a characteristic noise due to seed disruption) and the final roasting temperature on coffee extracts, were investigated. Seeds were roasted at pilot scale according to 13 industrial roasting profiles and extracted in D2O. The extracts were analysed by 1H NMR experiments. The NMR spectra were compared using (a) quantitative analysis of main signals by successive orders of magnitude and (b) chemometric tools (principal component analysis, partial least squares and sparse-orthogonal partial least squares analysis). This allowed to identify compounds, which may serve as markers of roasting and showed that changes in chemical composition can be detected even for slight change in final temperature (~1°C) or in total roasting time (~25 s). 相似文献
