Phase transition of locust bean gum-, tara gum- and guar gum-water systems   总被引:1,自引:0,他引:1  
Phase transition behaviour of neutral galactomannans, i.e., locust bean gum (LBG), tara gum (Tara-G) and guar gum (GG)-water systems is investigated. In this study, water content {Wc=(gram of water)/(gram of dry sample)} of these systems was varied from 0.2 to 3.6 g g-1. In the DSC heating curves, glass transition (Tg), cold crystallization (Tcc) and melting (Tm) were observed in all three samples. In addition, liquid crystal transition (T*) was observed in GG-water systems at a temperature higher than Tm. Using Tg, Tcc, Tm and T*, phase diagrams of each system were established. From the melting enthalpy of ice in the systems, three types of water, non-freezing water (Wnf), freezing bound water (Wfb) and free water were calculated. The maximum amount of Wnf was observed at Wc=0.7 g g-1, where Tg showed the lowest temperature. The amount of Wnf in LBG and GG is higher than that of Tara-G, whereas the highest amount of Wfb is found in GG. T* was only observed in GG-water systems. It is concluded that frequency of the side chains in the repeating unit of the main chains of these three galactomannan affects the frozen structure of the glassy state in the presence of water. Cationic amphiphilic modified pullulan’s were obtained in two steps: firstly the synthesis of diethylaminoethylpullulan (DEAE-Pullulan) with DS0 = 0.80 in water (all degrees of substitution (DS) are calculated in anhydroglucose units (AGU)) followed by the incorporation of different alkyl chains (C10, C12 and C16) using Hoffmann alkylation reaction. Three alkylated derivatives (Y-Cx-DEAE-Pullulan) were produced with a degree of substitution of alkyl chains (Y = 100 DSCx) of 40 in the case of C10 and C12 and 20 in the case of C16. The three samples contained two functional types: an amine function (DSN), dependent on pH and a quaternary ammonium function (DSN+) linked to a hydrophobic alkyl chain (DSCx). Chemical characterizations were carried out by conductimetric measurements, elemental analysis and 1H NMR. The amphiphilic and associative properties have been confirmed by flow field flow fractionation (F4) coupled on-line with multi-angle laser light scattering (MALLS). The intermolecular associations were observed for DEAE-Pullulan and Y-Cx-DEAE-Pullulan in 0.1 M LiNO3.  相似文献

A novel low-molecular-weight pectic polysaccharide was isolated from sunflower heads that are a useless side product produced from sunflower oil processing. The low-molecular-weight pectic polysaccharide was purified by using an optimized four-step procedure and named as SHPPB-1. The molecular weight of SHPPB-1 is about 1.69× 104 Da. Structure characterizations of SHPPB-1 by monosaccharide composition, methylation analysis, and Fourier transform infrared (FT-IR) spectroscopy showed that SHPPB-1 is consisted of 1,4-linked α-D-GalpA and 1,4-linked 2-OAc-5-COOMe-α-D-GalpA with rare α/β-D-Rhap, α/β-D-Manp, and α/β-D-GalpA. This was combined with NMR spectroscopic analysis to propose a structure of SHPPB-1 as: →4)-[α/β-D-monosaccharide-(1→3)]-α-D-GalpA-(1→4)-2-OAc-5-COOMe-α-D-GalpA-(1→ .  相似文献

The O-specific polysaccharide of Alteromonas addita type strain KMM 3600T is constructed of trisaccharide repeat units containing L-rhamnose, D-glucose, and D-galactose. It was established that the O-specific polysaccharide consists of trisaccharide repeat units with the structure →3)-α-D-Galp-(1→3)-α-L-Rhap-(1→3)-α-D-Glcp-(1→ based on monosaccharide analysis, Smith degradation, PMR and 13C NMR spectroscopy, and two-dimensional COSY, HSQC, and HMBC. 肝素(heparin, Hp)是目前临床应用最为广泛的抗凝剂,是由重复二糖单元组成的多硫酸化酸性直链多糖。低分子量肝素(LMWHs)是以肝素为原料,经过化学或酶降解获得的相对分子质量相对较小的肝素衍生物,相对肝素,它们的出血副作用和免疫原性更小,皮下注射时生物利用度更高。肝素及低分子量肝素具有一系列结构特点,如相对分子质量偏大且有一定分布,多种糖残基同时存在,硫酸酯位置和数量呈现多样化,以及不同工艺产生的特殊残基的种类和含量不一等。该类药物结构的复杂性对分析方法提出了巨大的挑战,也限制了其质量控制提升、工艺优化、临床用药安全和新适应证拓展等。该文以色谱分析方法为中心,从结构分析的不同角度,包括单糖、二糖、寡糖、多糖的识别、组成分析和不同层次,系统地梳理和阐述近年来肝素类药物在色谱分析方法上的进展,并对这些方法的应用范畴、创新性、局限性等进行总结。该文将为肝素类药物的结构分析、质量控制提供较系统的方法学参考,为更多新方法开发提供思路,为更深入地研究肝素类药物结构、拓展其应用提供有力支撑。  相似文献

为制备孔径为100 nm的大孔硅胶,考察了热液法和焙烧法对球形硅胶(粒径5 μm,孔径10 nm)的扩孔效果。采用热液法扩孔时,在水溶液中加入22 g/L氟化钠,可以有效增强扩孔效果,在高压釜内160℃加热48 h便可扩孔至100 nm,但孔径不均匀。采用焙烧法扩孔时,通过调节焙烧温度、时间以及复盐LiCl-NaCl的加入量可以方便地控制扩孔速度与效果;在每10 g硅胶中加入1.125 g LiCl·H2O和0.75 g NaCl,于500℃焙烧3~5 h,可得到100 nm大孔硅胶;该方法简单、高效,扩孔后的硅胶孔径分布均匀,表面形态与商品化的Fuji-1000硅胶相似。将两种扩孔方式得到的硅胶经氨基修饰后,涂覆纤维素-三(3,5-二甲基苯基氨基甲酸酯)制得了相应的手性固定相。结果表明,采用焙烧法扩孔得到的硅胶制备的固定相明显具有较好的分离选择性及分离度。  相似文献

Lentil protein isolate (LPI)-κ-carrageenan (κ-C) and -ι-carrageenan (ι-C) based microcapsules were prepared through spray-drying and freeze-drying to encapsulate flaxseed oil in order to reach final oil levels of 20% and 30%. Characteristics of the corresponding emulsions and their dried microcapsules were determined. For emulsion properties, all LPI-κ-C and LPI-ι-C emulsions remained 100% stable after 48 h, while the LPI emulsions destabilized quickly (p < 0.05) after homogenization mainly due to low emulsion viscosity. For spray-dried microcapsules, the highest yield was attributed to LPI-ι-C with 20% oil, followed by LPI-κ-C 20% and LPI-ι-C 30% (p < 0.05). Flaxseed oil was oxidized more significantly among the spray-dried capsules compared to untreated oil (p < 0.05) due to the effect of heat. Flaxseed oil was more stable in all the freeze-dried capsules and showed significantly lower oil oxidation than the untreated oil after 8 weeks of storage (p < 0.05). As for in vitro oil release profile, a higher amount of oil was released for LPI-κ-C powders under simulated gastric fluid (SGF), while more oil was released for LPI-ι-C powders under simulated gastric fluid and simulated intestinal fluid (SGF + SIF) regardless of drying method and oil content. This study enhanced the emulsion stability by applying carrageenan to LPI and showed the potential to make plant-based microcapsules to deliver omega-3 oils.  相似文献

对当归多糖ASP3的糖链结构进行分析. 分别采用0.2 mol/L三氟乙酸(Trifluoroacetic acid, TFA)和内切-α-(1→4)-聚半乳糖醛酸酶(EndoPG)对ASP3进行部分酸水解和酶水解, 并对水解前后多糖组分的1D和2D NMR光谱特征进行分析. 实验结果表明, ASP3是一种果胶多糖. GalpA和Rhap位于多糖分子的主链, 由1→4-D-GalpA相连形成的“光滑区”(半乳糖醛酸聚糖)是其主要组成部分; 由α-(1→4)-GalpA通过O4位与α-(1→2)-和α-(1→2,4)-Rhap的O2位交替连接所形成的重复单元[→4)-α-GalpA-(1→2)-α-Rhap-(1→]构成具有较高分支的“毛发区”(富含中性糖侧链的鼠李半乳糖醛酸聚糖). Galp和Araf是中性糖侧链的主要组成, 通过Rhap残基的O4位与主链相连. 非还原性末端T-β-Galp, β-(1→3)-, β-(1→3,6)-, β-(1→4)-, β-(1→4,6)-Galp聚合形成以β-(1→3,6)-Galp为分支点的β-(1→6)-半乳聚糖和以β-(1→4,6)-Galp为分支点的β-(1→4)-半乳聚糖. T-α-Araf, α-(1→5)-Araf和α-(1→3,5)-Araf聚合形成以α-(1→3,5)-Araf为分支点的α-(1→5)-阿拉伯聚糖. 此外, 由α-(1→5)-阿拉伯聚糖通过α-(1→3)连接与β-(1→6)-半乳聚糖末端聚合形成阿拉伯半乳聚糖.  相似文献
