Chiral separation of 1,1'-bi-2-naphthol and its analogue on molecular imprinting monolithic columns by HPLC

Two molecular imprinting polymer (MIP) monolithic columns with (S)-(-)-1,1'-bi-2-naphthol and (R)-(+)-5,5',6,6',7,7',8,8'-octahydro-1,1'-bi-2-naphthol as the templating molecules, respectively, have been prepared by in situ polymerization using 4-vinylpyridine and ethylene dimethacrylate as functional monomer and cross-linker, respectively. The columns with good flow-through properties were obtained by changing the molar ratio of the functional monomer and the template molecule. The effects of mobile-phase composition on separation of enantiomers were systematically investigated. The results indicate that hydrophobic interaction in aqueous solution and hydrogen-bonding interaction in ACN between the enantiomers and polymers could play important roles in the retention and resolution. The effects of chromatographic conditions, such as flow rate, column temperature, sample loading, on the enantioseparation were also studied. Further, these two MIP columns show a cross-reactivity.     

抗人绒毛膜促性腺激素（HCG）单克隆抗体的研制和特性分析

以HCG为抗原,免疫BALB/c小鼠,取其脾细胞与小鼠Sp2/0—Ag14骨髓瘤细胞融合,经反复筛选和再克隆化,获得8株能稳定分泌抗HCG特异单克隆抗体的杂交瘤细胞株。对其中6个高效价单抗进行了特异性和抗原决定簇检测,选出两个抗相距较远不同决定簇的单抗(2D_4,4F_4)作进一步特性分析。结果显示:免疫球蛋白亚类鉴定,2D_4为IgGl亚类,4F_4为IgG2a亚类;亲和力常数(K_(affi)),2D_4为5.41±0.94×10~(-10)mol/L,4F_4为1.37±0.33×10~(-9)mol/L。证实该两种单克隆抗体可用于研制有关临床诊断试剂盒。     

白腐菌降解氯代农药的机理

确定了白腐菌在水中的静置培养条件,探讨了在该条件下白腐菌对水中微量氯代农药的生物降解情况及其降解机理．实验结果表明,白腐菌在含有ρ为０．０２％的葡萄糖、０．０３％的酒石酸铵和适量无机盐的培养液中,于３５℃,经５～７ｄ的静置培养可获得良好的菌丝,并对氯代农药有最大的降解效率（９０％以上）．对降解的部分中间产物的ＧＣ－ＭＳ分析表明,不同类氯代农药有不同的降解过程和机理

The molecular mechanism of stabilization of proteins by TMAO and its ability to counteract the effects of urea

Trimethylamine n-oxide (TMAO) is a naturally occurring osmolyte that stabilizes proteins and offsets the destabilizing effects of urea. To investigate the molecular mechanism of these effects, we have studied the thermodynamics of interaction between TMAO and protein functional groups. The solubilities of a homologous series of cyclic dipeptides were measured by differential refractive index and the dissolution heats were determined calorimetrically as a function of TMAO concentration at 25 degrees C. The transfer free energy of the amide unit (-CONH-) from water to 1 M TMAO is large and positive, indicating an unfavorable interaction between the TMAO solution and the amide unit. This unfavorable interaction is enthalpic in origin. The interaction between TMAO and apolar groups is slightly favorable. The transfer free energy of apolar groups from water to TMAO consists of favorable enthalpic and unfavorable entropic contributions. This is in contrast to the contributions for the interaction between urea and apolar groups. Molecular dynamics simulations were performed to provide a structural framework for the interpretation of these results. The simulations show enhancement of water structure by TMAO in the form of a slight increase in the number of hydrogen bonds per water molecule, stronger water hydrogen bonds, and long-range spatial ordering of the solvent. These findings suggest that TMAO stabilizes proteins via enhancement of water structure, such that interactions with the amide unit are discouraged.     

New diphosphite ligands for enantioselective asymmetric hydroformylation

A series of new diphosphite ligands have been easily prepared from BINOL derivatives; moderate enantioselectivities (up to 80% ee) and excellent regioselectivities (b/l up to 98/2) have been achieved in the Rh-catalyzed asymmetric hydroformylation of vinyl acetate.
