FAE1启动子的克隆和转γ-TMT基因大豆的获得

脂肪酸链延长酶组分1(FAE1)是广泛存在于植物中催化脂肪酸碳链延长的酮脂酰CoA合成酶,该反应是脂肪酸碳链延长的限速步骤,在油料种子胚中特异性表达.以拟南芥基因组DNA为模板,设计引物,扩增到该基因的启动子片段.序列分析表明,该片段长度为943 bp,与GenBank注册序列AF355982达99.5%同源,并含有与之完全相同的种子特异性表达元件,据此认定它具有种子特异性启动活性.将其与甘蓝型油菜γ-生育酚甲基转移酶(γ-TMT)基因连接,构建成表达载体pFAE1-TMT,通过农杆菌介导的方法转化大豆胚尖,获得28株卡那霉素抗性筛选植株,进一步PCR检测获得2株转γ-TMT基因植株.     

二球悬铃木叶片离体植株再生

研究不同浓度细胞分裂素BA或TDZ与生长素NAA或IBA组合对二球悬铃木种子苗所繁殖植株的叶片分化不定芽的影响.结果表明,诱导二球悬铃木种子苗所繁殖植株的叶片形成不定芽的最佳激素组合为3 mg/L BA和0.5 mg/L IBA;不定芽的诱导效果与植株的繁殖代数有关,在60 d的培养过程中,繁殖6代、10代、15代和20代的不定芽形成率分别为47%、44%、40%和38%.所有长达1 cm的不定芽在含2 mg/LIBA和1 g/L活性炭的培养基上培养20 d均生根良好.经过炼苗后,再生植株的移栽成活率达到92%.     

前体物质和化学诱导子对云南红豆杉细胞生长和产生紫杉醇的影响

研究了几种前体物质和化学诱导子对云南红豆杉细胞生长产生紫杉醇的影响,结果表明,在培养第１２ｄ培养其中添加１ｍｍｏｌ．Ｌ＾－１或２ｍｍｏｌ．Ｌ＾－１苯丙氨酸、５ｍｍｏｌ．Ｌ＾－１丙酮酸钠和１ｇ．Ｌ＾－１牛儿醇可显提高细胞中紫杉醇的含量,同对照相比,紫杉醇产量分别增加了４０．０％、９５．７％和１２１．３％;使用高盐渗透胁迫剂（３００ｍｍｏｌ．Ｌ＾－１）高糖渗透胁迫剂（２５ｇ．Ｌ＾－１蔗糖和２５ｇ．Ｌ＾－１甘露醇）、桂皮酸（３ｍｍｏｌ．Ｌ＾－１）和硫酸钒酰（１０ｍｇ．Ｌ＾－１）可显促进细胞中紫杉醇的合成,同时对照相比,紫杉醇产量分别增加了２３．０％、８８．０％、４２．８％和１０３．０％。     

浮子式波浪能转换装置的浮子受力分析

为提高现有波浪发电系统的稳定性和效率,提出了一种新型的浮子式波浪发电系统,介绍了其具体结构和特点.同时,采用解析法详细分析了波浪能转换装置(WEC)所受的重力、浮力、系链拉力等情况.根据天津波浪特性,对所提出的波浪发电装置进行了仿真分析.分析结果表明,所设计的波浪能转换装置具有较好的单向周期性稳定输出转矩特性,且其效率较传统的浮子式WEC提高了10%左右.     

A microchip laser feedback interferometer with nanometer resolution and increased measurement speed based on phase meter

We introduce a new signal processing method based on phase meter into heterodyne microchip Nd:YAG laser feedback interferometer. The nanometer resolution and a higher measurement speed are realized. The factors determining the accuracy are analyzed. The displacements of the Physik Instrumente nanopositioning system and two piezoelectric transducers were measured. Experimental results indicate laser feedback interferometer’s ability of measuring nanoscale displacement and present promising application prospects in noncooperative targets measurement.     

Synthesis and target identification of hymenialdisine analogs

Hymenialdisine (HMD) is a sponge-derived natural product kinase inhibitor with nanomolar activity against CDKs, Mek1, GSK3beta, and CK1 and micromolar activity against Chk1. In order to explore the broader application of the pyrrolo[2,3-c]azepine skeleton of HMD as a general kinase inhibitory scaffold, we searched for additional protein targets using affinity chromatography in conjunction with the synthesis of diverse HMD analogs and profiled HMD against a panel of 60 recombinant enzymes. This effort has led to nanomolar to micromolar inhibitors of 11 new targets including p90RSK, KDR, c-Kit, Fes, MAPK1, PAK2, PDK1, PKCtheta, PKD2, Rsk1, and SGK. The synthesis of HMD analogs has resulted in the identification of compounds with enhanced and/or dramatically altered selectivities relative to HMD (28n) and in molecules with antiproliferative activities 30-fold higher than HMD (28p).     

具有一级磁相变的不同系列NiMnGa合金其电子浓度对相变温度的影响

分析了具有一级磁相变的Ni2 x yMn1-xGa1-y,Ni2 xMn1-xGa两个合金系列中,合金电子浓度e/a对结构和磁转变温度的影响。对于Ni2 x yMn1-xGa1-y系列合金,随着电子浓度的增加,马氏体相变温度TM增大,居里温度TC减小,在e/a=7.68处相交,继续增加e/a,TM和TC同时减小;而对于Ni2 xMn1-xGa系列合金,随x的增加,电子浓度增加,TM增大,TC减小,在e/a=7.635处相交,x继续增大,TM和TC仍然缓慢增加。不同NiMnGa合金系列中TM,TC与合金的e/a之间不同的变化关系说明,e/a对TM和TC的影响在不同的NiMnGa合金系列中是相同的。     

Direct metal-metal interaction contributions to quadratic hyperpolarizability: a study on dirhenium complexes

We present a comparative study of the metal-metal interaction effect on the static quadratic hyperpolarizabilities of two typical dinuclear rhenium clusters. The electronic structures, excitation spectra, dipolar moments, static polarizabilities, and quadratic hyperpolarizabilities of the two complexes with direct metal-metal interactions have been computed and analyzed with the use of high-level DFT/TDDFT methods. The geometries and the first intense excitations agree with the relevant reported measurements. The orbital decomposition scheme ( J. Phys. Chem. A 2006, 110, 1014-1021) has been applied to analyze the relationship between the electronic structures and nonlinear optical (NLO) properties of these two complexes. We propose an unprecedented NLO response mechanism featuring the contribution of the direct metal-metal interaction transition process in these dinuclear rhenium complexes. This contribution positively enhances the quadratic hyperpolarizability and relates to the intensity of the metal-metal interactions of the complexes. The results are helpful to the development of NLO chromophores in polynuclear metal clusters through the molecular design technique.     

生产性服务业规模对制造业效率的影响研究——分地区分行业的视角

基于2007—2016年全国29个省份面板数据,以生产性服务业发展规模为主要解释变量,本文分析生产性服务业发展规模对制造业效率的影响。实证结果表明,生产性服务业发展规模对制造业效率有明显的积极影响;分地区分析,影响程度排序为:西部东部中部;分行业分析,交通运输、仓储邮政业和金融业发展对制造业效率具有显著的积极影响。根据分析提出对策,全国要营造有利的政策环境;东部要注重制造业增加值的提升;中部应促进制造业外包;西部应重点发展区位影响小的生产性服务业。     

MS培养基凝固效果和高温灭菌后pH值变化的研究

研究了培养基的pH值、琼脂浓度与MS培养基凝固的关系 ,以及不同成分对高温灭菌后MS培养基pH值的影响 .结果表明 ,调高培养基的pH值或增加琼脂浓度均可改善培养基的凝固效果 ;高温灭菌后 ,MS基本培养基的pH值由灭菌前的 5 .80下降为 5 .5 1 ,造成pH值下降的最主要原因是高温灭菌过程中EDTA铁盐与其它微量元素发生相互作用所致 ;同基本培养基相比 ,培养基中加入 3 0 .0g·L- 1 蔗糖、食用白糖、葡萄糖或果糖使高温灭菌后的培养基的pH值分别下降了 0 .0 6、0 .1 0、0 .2 8和 0 .98个单位 ,而添加 2 .0g·L- 1 水解酪蛋白、蛋白胨、酵母提取物或 1 .0～ 5 .0g·L- 1 活性炭则使高温灭菌后的培养基的pH值分别上升了 0 .2 3 ,0 .1 8,0 .3 9和 0 .1 9～ 0 .72个单位
