采用非水毛细管电泳方法对2’-AMP、3’-AMP和5’-AMP 3种单磷酸腺苷进行分离研究,考察了电泳溶液pH*值、非水介质、缓冲溶液对分离的影响。以含50%乙腈的Tris-H3BO3体系为缓冲溶液,在pH*10.0、压差进样(50 mbar,5 s)、柱温25℃、25 kV恒压下进行分离,在波长260 nm处负极检测,各组分可达到基线分离。在质量浓度为1~100 mg/L范围内,3种单磷酸腺苷的线性关系良好,平均回收率为88%~106%,RSD小于4%。该方法应用于核苷样品的测定,结果满意。  相似文献

Efficient basic hydrotalcite (HT)-supported gold nanoparticle (AuNP) catalysts have been developed for the aerobic oxidative tandem synthesis of methyl esters and imines from primary alcohols catalyzed under mild and soluble-base-free conditions. The catalytic performance can be fine-tuned for these cascade reactions by simple adjustment of the Mg/Al atomic ratio of the HT support. The one-pot synthesis of methyl esters benefits from high basicity (Mg/Al=5), whereas moderate basicity greatly improves imine selectivity (Mg/Al=2). These catalysts outperform previously reported AuNP catalysts by far. Kinetic studies show a cooperative enhancement between AuNP and the surface basic sites, which not only benefits the oxidation of the starting alcohol but also the subsequent steps of the tandem reactions. To the best of our knowledge, this is the first time that straightforward control of the composition of the support has been shown to yield optimum AuNP catalysts for different tandem reactions.  相似文献

Allylated biphenol neolignans contain a variety of chemopreventive entities that have been used as anti-tumor drug leads. Herein, 37 allylated biphenols were evaluated for anti-proliferative activity by the MTT assay and inhibitory effect on the migration and tube formation of HUVECs featuring anti-angiogenic properties. 3-(2-Methylbut-3-en-2-yl)-3′,5′-bis(trifluoromethyl)-[1,1′-biphenyl]-4-ol (5c) exerted an inhibitory effect on HUVECs compared to honokiol (IC?? = 47.0 vs. 52.6 μM) and showed significant blocking effects on the proliferation of C26, Hela, K562, A549, and HepG2 (IC?? = 15.0, 25.0, 21.2, 29.5, and 13.0 μM, respectively), superior to those of honokiol (IC?? = 65.1, 62.0, 42.0, 75.0, and 55.4 μM, respectively). Importantly, compound 5c inhibited the migration and capillary-like tube formation of HUVECs in vitro.  相似文献

The effect of heavy metal ions, Cd(2+), Hg(2+) and Pb(2+) on (+)-catechin binding to bovine serum albumin (BSA) has been investigated by spectroscopic methods. The results indicated that the presence of heavy metal ions significantly affected the binding modes and binding affinities of (+)-catechin to BSA, and the effects depend on the types of heavy metal ion. One binding mode was found for (+)-catechin with and without Cd(2+), while two binding modes - a weaker one at low concentration and a stronger one at high concentration were found for (+)-catechin in the presence of Hg(2+) and Pb(2+). The presence of Cd(2+) decreased the binding affinities of (+)-catechin for BSA by 20.5%. The presence of Hg(2+) and Pb(2+) decreased the binding affinity of (+)-catechin for BSA by 8.9% and 26.7% in lower concentration, respectively, and increased the binding affinity of (+)-catechin for BSA by 5.2% and 9.2% in higher concentration, respectively. The changed binding affinity and binding distance of (+)-catechin for BSA in the presence of Cd(2+), Hg(2+) and Pb(2+) were mainly because of the conformational change of BSA induced by heavy metal ions. However, the quenching mechanism for (+)-catechin to BSA was based on static quenching combined with non-radiative energy transfer irrespective of the absence or presence of heavy metal ions.  相似文献

The synthesis, crystal structure, magnetic properties, and single-molecule conductance of two new trinuclear metal string complexes, [Ni(3)(dzp)(4)(NCS)(2)] (2) and [Co(3)(dzp)(4)(NCS)(2)] (3), containing the rigid Hdzp ligand (1, 1,9-diazaphenoxazine) are reported. X-ray structural analyses show that compounds 2 and 3 exhibit smaller torsion angles and longer metal-metal distances than those exhibited by the corresponding dpa(-) analogues (dpa(-) = dipyridylamido anion) due to the rigidity of Hdzp ligands. The longer metal-metal distance observed for 2 and 3 results in variations in their magnetic properties. The exchange interaction (J = -160 cm(-1)) between two high spin (HS) Ni(II) ions in 2 decreases slightly in comparison with those of trinickel dpa(-) analogues. The doublet-quartet gap of 3 is smaller than that of [Co(3)(dpa)(4)(NCS)(2)] (4), which causes compound 3 to show spin-crossover behavior even at low temperature.  相似文献

Streptocarbazoles A (1) and B (2), two novel indolocarbazoles featuring unprecedented cyclic N-glycosidic linkages between 1,3-carbon atoms of the glycosyl moiety and two indole nitrogen atoms of the indolocarbazole core, were isolated from the marine-derived actinomycetes strain Streptomyces sp. FMA. Their structures were established by spectroscopic methods, CD spectra, and ECD quantum mechanical calculations. Compound 1 was cytotoxic on HL-60 and A-549 cell lines and could arrest the cell cycle of Hela cells at the G(2)/M phase.  相似文献

A series of triphenylamine-centered starburst quinolines (1a-1g) have been synthesized by Friedl?nder condensation of the 4,4',4'-triacetyltriphenylamine (2) and 2-aminophenyl ketones (3a-3g) in the presence of catalytic sulfuric acid and characterized well. They are thermally robust with high glass transition temperatures (above 176.4 °C) and decomposition temperatures (above 406 °C). These compounds emit blue fluorescence with λ(max)(Em) ranging from 433 to 446 nm in dilute toluene solution and 461 to 502 nm in the solid-state and have a relatively high efficiency (Φ(u) = 0.98-0.57). 1a-1g have estimated ionization potentials (IP) of 4.54 to 6.45 eV which are significantly near or higher than those of well-known electron transport materials (ETMs), including tris(8-hydroxyquinoline)aluminium (Alq(3)) (IP = 5.7-5.9 eV), and previously reported oligoquinolines (IP = 5.53-5.81 eV). Quantum chemical calculations using DFT B3LYP/6-31G* showed the highest occupied molecular orbital (HOMO) of -5.05 to -4.81 eV, which is close to the work function of indium tin oxide (ITO). These results demonstrate the potential of 1a-1g as hole-transporting/light-emitting/electron-transport materials and the host-materials of a dopant for hole-injecting for applications in organic light-emitting devices.  相似文献

Annexin V (1) specifically binds to phosphatidylserine on apoptotic and necrotic cells as well as certain cancer cells, making it an attractive vehicle for the delivery of therapeutically-relevant conjugates to such sites. The wild-type protein possesses a single thiol at Cys316, which is difficultly accessible to site-specific labeling by simple maleimides. By contrast, 1,4-benzoquinone site-specifically labels annexin V in minutes. The resulting conjugate (5) serves as an intermediate for crosslinking annexin molecules, which can be accomplished within hours either directly for linking annexin V-128 (19), or via an extended sequence involving the crosslinking of two units of (5) by the symmetrical α,ω-dithiol (20). Besides its ability to mediate protein dimer formation while retaining annexin V's ability to bind phosphatidylserine, (5) possesses classic 1,4-benzoquinone reactivity. Various nucleophiles and Diels-Alder dienes form adducts with (5) in reactions that may have general utility for the synthesis of novel biologically active entities. The present work presents the first example of thiol-specific crosslinking of proteins by 1,4-quinone-based methodology designed to exploit the reactivity of this versatile chemical entity.  相似文献

In the present study, we aimed to develop a nucleic acid lateral-flow method for the rapid and sensitive detection of multiple bacteria that contaminate platelet concentrations (PCs). Polymerase chain reaction (PCR) amplicons were produced by a set of board-range primers that recognize the conserved region of bacteria 16S rDNA, followed by hybridization with both an FITC (fluorescein isothiocyanate)-labelled probe and biotin-labelled probe, and then a nucleic acid lateral-flow dipstick (LFD) assay. The LFD accurately identified 7 species of bacteria, but had no cross-reactivity with human genomic DNA. The limit of detection (LOD) of the LFD assay was as low as 10(1) copies/μL of 16S rDNA for plasmid. In the case of spiked PCs without enrichment, the detection limit of LFD for K. pneumonia was 5 CFU/mL, 6.5 × 10(4) CFU/mL for the S. epidermidis and 35 CFU/mL for P. aeruginosa.  相似文献
