Bacillus anthracis is an endospore-forming bacterium that causes inhalational anthrax. Key virulence genes are found on plasmids (extra-chromosomal, circular, double-stranded DNA molecules) pXO1 (ref. 2) and pXO2 (ref. 3). To identify additional genes that might contribute to virulence, we analysed the complete sequence of the chromosome of B. anthracis Ames (about 5.23 megabases). We found several chromosomally encoded proteins that may contribute to pathogenicity--including haemolysins, phospholipases and iron acquisition functions--and identified numerous surface proteins that might be important targets for vaccines and drugs. Almost all these putative chromosomal virulence and surface proteins have homologues in Bacillus cereus, highlighting the similarity of B. anthracis to near-neighbours that are not associated with anthrax. By performing a comparative genome hybridization of 19 B. cereus and Bacillus thuringiensis strains against a B. anthracis DNA microarray, we confirmed the general similarity of chromosomal genes among this group of close relatives. However, we found that the gene sequences of pXO1 and pXO2 were more variable between strains, suggesting plasmid mobility in the group. The complete sequence of B. anthracis is a step towards a better understanding of anthrax pathogenesis.  相似文献

The dissolution reaction of WO3 in pure molten K2S2O7 and in molten K2S2O7-K2SO4 mixtures is studied under static equilibrium conditions in the XWO3(0) = 0-0.33 mol fraction range at temperatures up to 860 °C. High temperature Raman spectroscopy shows that the dissolution leads to formation of W(VI) oxosulfato complexes, and the spectral features are adequate for inferring the structural and vibrational properties of the complexes formed. The band characteristics observed in the W=O stretching region (band wavenumbers, intensities, and polarization characteristics) are consistent with a dioxo W(=O)2 configuration as a core unit within the oxosulfato complexes formed. A quantitative exploitation of the relative Raman intensities in the binary WO3-K2S2O7 system allows the determination of the stoichiometric coefficient, n, of the complex formation reaction WO3 + nS2O7(2-) --> C(2n-). It is found that n = 1; therefore, the reaction WO3 + S2O7(2-) > WO2(SO4)2(2-) with six-fold W coordination is proposed as fully consistent with the observed Raman features. The effects of the incremental dissolution and presence of K2SO4 in WO3-K2S2O7 melts point to a WO3 · K2S2O7 · K2SO4 stoichiometry and a corresponding complex formation reaction in the ternary molten WO3-K2S2O7-K2SO4 system according to WO3 + S2O7(2-) + SO4(2-) --> WO2(SO4)3(4-). The coordination sphere of W in WO2(SO4)2(2-) (binary system) is completed with two oxide ligands and two chelating sulfate groups. A dimeric [{WO2(SO4)2}2(μ-SO4)2](8-) configuration is proposed for the W oxosulfato complex in the ternary system, generated from inversion symmetry of aWO2(SO4)3(4-) moiety resulting in two bridging sulfates. The most characteristic Raman bands for the W(VI) oxosulfato complexes pertain to W(=O)2 stretching modes (i) at 972 (polarized) and 937 (depolarized) cm(-1) for the ν(s) and ν(as) W(=O)2 modes of WO2(SO4)2(2-), and (ii) at 933 (polarized) and 909 (depolarized) cm(-1) for the respective modes of [{WO2(SO4)2}2(μ-SO4)2](8-).  相似文献

Isotope dilution inductively coupled plasma mass spectrometry (ID-ICP-MS) was applied to the certification of Pb in four levels of NIST blood SRM, 955a. This standard reference material (SRM) represents a significant improvement over the previous blood reference material and will greatly aid method development. The lowest level, 47.76 ng/g Pb was determined with analysis uncertainty (95% CI, ID-ICP-MS uncertainties) of less than 1% and the highest level, 517.9 ng/g Pb to 0.3%. Uncertainty in the lowest level was due to sample inhomogeneity and variability in the analytical blank as the RSD on ratio measurements was typically better than 0.2%. Properly applied isotope dilution coupled with careful isotope ratio measurements on the ICP-MS offers precision and accuracy for blood Pb analyses beyond what is currently obtainable with routine methods.  相似文献

Nuclear inelastic scattering (NIS) of synchrotron radiation has been used to investigate the dynamics of tin ions chelated by DNA. Theoretical NIS spectra have been simulated with the help of density functional theory (DFT) calculations using 12 models for different binding sites of the tin ion in (CH3)Sn(DNAPhosphate)2. The simulated spectra are compared with the measured spectrum of the tin–DNA complex.  相似文献

Mixed oxides in the system S-Ce-Co-O were prepared by solid state reaction and by freeze-drying of precursor compounds followed by thermal treatment. Two types of perovskite oxides exist in the system: Solid solutions of the type Sr1 − yCeyCoO3 − x and mixed oxides of the type (1 − y)SrCeO3 − ySrCoO3 − x. Microstructures and phase compositions were determined by electron microscopy and X-ray diffraction. SrCoO3 − x forms a solid solution of ceria on the A-site in the strontium cobaltite lattice up to 0.15 mol Ce. This solid solution corresponds to the high-temperature structure of pure SrCoO3 − x and is characterized by high oxygen exchange and electrical conductivity. The oxygen deficiency x was measured by solid electrolyte coulometry. The oxygen deficiency of solid solutions Sr1 − yCeyCoO3 − x increases with temperature and decreases with pO2 in the ambient atmosphere and with increasing Ce dopant concentration. The pO2-T-x diagram of the solid solution was determined. The T, pO2 and dopant concentration dependencies of electrical conductivity were measured by a four-point d.c. technique. By Ce doping strontium cobaltite becomes a stabilized high-conductive material (maximum conductivity: 500 S cm−1 at 400 °C, Ea = 0.025 eV, p-type). Above this temperature the T-coefficient of the conductivity changes from positive (semiconducting) to negative values.  相似文献
