Fibroblast growth factor-21 (FGF21) is a hepatic hormone that plays a critical role in metabolism, stimulating fatty acid oxidation in the liver and glucose uptake in adipose tissue. In this study, we produced tobacco plants expressing human recombinant FGF21 (hFGF21) via Agrobacterium-mediated transformation using a potato virus X (PVX)-based vector (pgR107). The vector contained the sequence encoding the human FGF21 gene fused with green florescence protein and a histidine tag. The recombinant plasmid was introduced into leaf cells of Nicotiana benthamiana (a wild Australian tobacco) via Agrobacterium-mediated agroinfiltration. As determined by fluorescence and Western blot of leaf extracts, the hFGF21 gene was correctly translated in tobacco plants. Seven days after agroinfection, the recombinant hFGF21 had accumulated to levels as high as 450 μg g−1 fresh weight in leaves of agroinfected plants. The recombinant hFGF21 was purified from plant tissues by Ni–NTA affinity chromatography, and the purified hFGF21 stimulated glucose uptake in 3T3/L1 cells. This indicated that the recombinant hFGF21 expressed via the PVX viral vector in N. benthamiana was biologically active.  相似文献

This work describes a novel methodology for unequivocal identification of chemical constituents in Zhimu‐Baihe herb‐pair (ZMBHHP). Compounds were removed from ZMBHHP by ultrasonic extraction with 70% ethanol, and then analyzed by fast high‐performance liquid chromatography (HPLC) coupled with time‐of‐flight mass spectrometry (TOFMS). The accurate‐mass capability of the TOF analyzer allowed reliable confirmation of the identity of the detected compounds, normally with mass errors below 3 ppm in routine analysis. This mass accuracy was sufficient to verify the elemental compositions of the chemical constituents in ZMBHHP. With dynamic adjustment of fragmentor voltage in TOFMS, an efficient ion transmission was achieved to obtain the best sensitivity and abundant fragmentation. By accurate mass measurements for each molecular ion and subsequent fragment ions, a reliable identification and differentiation of 24 saponins, 3 xanthones, 1 anthraquinone and 2 alkaloids was described here, including four groups of isomers. It is concluded that this fast and sensitive HPLC/ESI‐TOFMS technique is powerful in qualitative analysis of complex herbal medicines in terms of sensitivity, selectivity, resolving power, time savings and lower solvent consumption. Furthermore, the data gathered in this study may be helpful for understanding the synergistic nature of this herb pair in traditional Chinese medicine (TCM) and further pharmacokinetic studies of ZMBHHP. Cellulose nanofibers (CNFs) were isolated from four kinds of plant cellulose fibers by a chemical-ultrasonic treatment. The chemical composition, morphology, crystalline behavior, and thermal properties of the nanofibers and their intermediate products were characterized and compared. The CNFs extracted from wood, bamboo, and wheat straw fibers had uniform diameters of 1040 nm, whereas the flax fibers were not uniformly nanofibrillated because of their initially high cellulose content. The chemical composition of each kind of nanofibers was mainly cellulose because hemicelluloses and lignin were significantly removed during chemical process. The crystallinity of the nanofibers increased as the chemical treatments were applied. The degradation temperature of each kind of nanofiber reached beyond 330 °C. Based on the properties of the CNFs, we expect that they will be suitable for use in green nanocomposites, filtration media and optically transparent films.  相似文献

The Pd-catalyzed cross-coupling of racemic tertiary allylic carbonates and allylboronates is described. This reaction generates all-carbon quaternary centers in a highly regioselective and enantioselective fashion. The outcome of these reactions is consistent with a process that proceeds by way of 3,3'-reductive elimination of bis(η(1)-allyl)palladium intermediates. Strategies for distinguishing the product alkenes and application to the synthesis of (+)-α-cuparenone are also described.  相似文献

The aggregation-induced emission (AIE) of a 1,2-diphenyl-1,2-di(p-tolyl)ethene (TPE) was explored as a novel fluorescence method for probing the assembling/disassembling of amphiphilic molecules. The fluorescence intensity was able to monitor the formation of micelles and determine the critical micelle concentration (CMC) of surfactants. The temperature-dependent micellization of the pharmaceutically important PEO-PPO-PEO copolymer, Pluronic F127, was further studied by using the TPE fluorescence spectrum intensity. Our results showed good agreement with those reported in the literature by using other methods. The special advantage of the AIE probe method was further explored to determine the assembling/disassembling process of the colored amphiphilic molecule, 1-[4-(3-phenylazophenoxy)butyl]triethylamine bromide (AzoC4), whose CMC value has not previously been described. Since the TPE fluorescence signal mainly comes from the aqueous phase, not from the inside of hydrophobic core, it provides a possible platform to study the CMC of those colored surfactants. Based on the novel fluorescence properties of TPE in the aggregated and dispersed states, one can conclude that the TPE method is a promising method for the determination of the CMC and critical micellization temperature (CMT), particularly having a special advantage to determine the assembling/disassembling process of colored amphiphilic molecules.  相似文献

建立了反相离子对色谱(RP-IPC)与电感耦合等离子体质谱(ICP-MS)联用技术快速分离测定化妆品中Cr(III)和Cr(VI)的方法。样品经乙二胺四乙酸钠盐(EDTA)50 ℃水浴中提取后,采用XDB-C18色谱柱分离,以5%(v/v)甲醇-2.0 mmol/L四正丁基铵盐(TBA)水溶液(pH 6.0)作为流动相,流速为1.0 mL/min,进样量为100 μL。采用碰撞池技术消除ICP-MS测定时40Ar12C+、35Cl16O1H+对52Cr+的光谱学干扰。Cr(III)和Cr(VI)的分离过程在5 min内完成。样品中加标量为0.01~0.50 μg时,其回收率为82.7%~107.2%,相对标准偏差(RSD)小于5.62%。该方法操作简便、灵敏度高、重现性好,适合于化妆品中Cr(III)和Cr(VI)的同时分析。  相似文献

A cobalt(11) complex containing mixed-ligands of 1,10-phenathroline(phen) and glycollic acid(GA).|Co(GA)2(phen)| was synthesized and its structure was characterized by X-ray diffraction.The interaction of the complex with DNA was investigated by electronic absorption spectra and electrochemical methods.Electronic absorption spectrum experiments showed that after interaction with DNA.the characteristic absorption peaks of |Co(GA)2(phen)| underwent hypochromic effect as well as redshift. Also,the binding strength of 3.8×104L/mol was estimated by titration method.Electrochemical assays revealed that the redox peak currents of the complex decreased obviously accompanied by a positive shift of the formal potential after association with DNA.All these results revealed that the synthesized cobalt complex bound with DNA via an intercalation mode.  相似文献

A series of naphthopyrans with hydrazone unit ( 8a – 8m ), were synthesized and characterized by 1H NMR, 13C NMR, IR and HRMS. The photochromic properties were investigated under continuous irradiation, in particular regard to the fatigue resistance and the lifetime of the colored open form in solution and polymers. The results showed that these compounds had both good photochromic properties and high fatigue resistance. Detailed studies showed that representative compound 8d (3,3‐di‐4‐methoxybenzoic acid methylenehydrazino‐[3H]‐naphtho [2,1‐b]pyran) had good photochromic properties in THF solution, in solid state, and in polymers, and exhibited a significant bathochromic shift in the spectra of the open forms compared to known naphthopyrans 9 (3,3‐diphenyl‐[3H]‐naphtho[2,1‐b]pyran). On the other hand, the higher melting points of target compounds are promising for the polymer film preparation through hot‐melt method.  相似文献

To search for more potent antitumor agent,a series of novel nitric oxide-donating colchicine(Col) derivatives(6a-f,8a and b) were synthesized by coupling nitrates with N-methyl colchiceinamide.Their cytotoxicity against four human cancer cell lines in vitro were evaluated by MTT assay.It was found that many of the derivatives displayed significant activity,particularly,compounds 6c,8a and 8b showed more potent cytotoxic activities than Col.  相似文献
