目的:应用全基因组DNA芯片技术分析低盐冷刺激作用下副溶血弧菌基因的转录表达变化.方法:分别采用"低盐持续刺激培养(continuous growth,CTG)"和"中间转入低盐环境培养(shift growth,STG)".CTG和STG下.分别采用含NaCl浓度为2%和0.66%的MV-5培养基孵育副溶血弧菌,收集菌体,提取RNA,应用全基因组DNA芯片分别比较两个不同的转录表达谱基因变化特点,分析其作用规律.同时,应用实时定量逆转录多聚酶联反应对芯片结果进行验证.结果:和对照组相比,STG实验中,共有205个基因的转录表达发生显著性变化,上调的基因占优势地位;CTG实验中,总计有298个基因的转录表达发生显著性变化,上、下涮的基因总体基本趋于平衡状态,没有明显差异.实时定量逆转录多聚酶联反应结果证实其和芯片数据结果有很强的相关性.结论:在低盐这一"胁迫环境"下,副溶血弧菌利用其存在的独特而精细的应对机制,能够顽强的生存下来并繁衍生殖,这一过程中,节能调节处于调控的核心地位.  相似文献

Historically, chemists have explored chemical space in a highly uneven and unsystematic manner. As an example, the shape diversity of existing fragment sets does not generally reflect that of all theoretically possible fragments. To assess experimentally the added value of increased three dimensionality, a shape-diverse fragment set was designed and collated. The set was assembled by both using commercially available fragments and harnessing unified synthetic approaches to sp3-rich molecular scaffolds. The resulting set of 80 fragments was highly three-dimensional, and its shape diversity was significantly enriched by twenty synthesised fragments. The fragment set was screened by high-throughput protein crystallography against Aurora-A kinase, revealing four hits that targeted the binding site of allosteric regulators. In the longer term, it is envisaged that the fragment set could be screened against a range of functionally diverse proteins, allowing the added value of more shape-diverse screening collections to be more fully assessed.  相似文献

In the cores of some clusters of galaxies the hot intracluster plasma is dense enough that it should cool radiatively in the cluster's lifetime, leading to continuous 'cooling flows' of gas sinking towards the cluster centre, yet no such cooling flow has been observed. The low observed star-formation rates and cool gas masses for these 'cool-core' clusters suggest that much of the cooling must be offset by feedback to prevent the formation of a runaway cooling flow. Here we report X-ray, optical and infrared observations of the galaxy cluster SPT-CLJ2344-4243 (ref. 11) at redshift z = 0.596. These observations reveal an exceptionally luminous (8.2?×?10(45)?erg?s(-1)) galaxy cluster that hosts an extremely strong cooling flow (around 3,820 solar masses a year). Further, the central galaxy in this cluster appears to be experiencing a massive starburst (formation of around 740 solar masses a year), which suggests that the feedback source responsible for preventing runaway cooling in nearby cool-core clusters may not yet be fully established in SPT-CLJ2344-4243. This large star-formation rate implies that a significant fraction of the stars in the central galaxy of this cluster may form through accretion of the intracluster medium, rather than (as is currently thought) assembling entirely via mergers.  相似文献

We present the first example of the phase transition occurring via the different kinetic mechanisms, displacive or diffusive, competing with each other in quartz-like alpha-GeO2 single crystals. Upon room-pressure heating, alpha-GeO2 transforms to the rutile-type phase (the alpha-->r transition) via the diffusive mechanism. With increase of the treatment pressure the diffusive mode of the temperature-induced alpha-->r transition is substituted at approximately 4 GPa by a displacive-like mode, and then at approximately 6 GPa the transition type changes from the alpha-->r sequence to a displacive martensitic-like transition to a distorted rutile-like phase (alpha-->r'. A crossover between diffusive and displacive transition modes suggests a new way to control the meso- and nanometer-scale morphology of high-pressure phases.  相似文献

The photoluminescence (PL) at ∼2.2-2.3 eV from Ge-based nanocrystalline materials is described in the literature as nanocrystal size-independent. We have observed visible luminescence from two different types of stain-etched Ge samples, one prepared after Sendova-Vassileva et al. (Thin Solid Films 255 (1995) 282) in a solution of H2O2:HF at 50:1 volume ratio, and the other in a solution of HF:H3PO4:H2O2 at 34:17:1 volume ratio. Energy dispersive X-ray analysis (EDX), Raman and FTIR spectroscopy, and the near edge X-ray absorption structure (XANES), indicate that the chemically etched Ge layers of the former type of samples are composed of non-stoichometric Ge oxides, i.e. GeOx (0<x<2), and free from any Ge nanoconstructions. It is also suggested from XANES that the latter type of chemically etched Ge samples comprise 8-9 nm nanocrystals of Ge, surface-covered with mainly oxygen. Photoluminescence occurred at ∼2.3 eV for all samples. The PL behavior of the latter type of chemically etched Ge on annealing in different chemical environments (air or H) allowed us to conclude that the PL from these materials, as well as that from those Ge-based nanocrystalline materials reported in the literature, is from GeOxs.  相似文献

Abstract

The present status of rapid IR multicomponent control analysis is that of a rapidly changing dynamic field of investigation in which the state-of-the-art is largely reflected in meeting abstracts, instrument manufacturer's seminars, brochures, private communications, and widely scattered articles in rather obscure journals. There are no definitive texts. These facts notwithstanding, the actual state of affairs is that several hundred investigators are very active in this field, partially in research but many in quality control laboratories. The dearth of information of the usual more formal sort reflects in part the very pragmatic and proprietary use to which the IR instruments capable of doing this work are being employed. The results do not always represent publishable material. However, when viewed from a suitable breadth of viewpoint, there is genuine merit in the multicomponent measurements.  相似文献

Silver and chromium particles in the size range 10 nm to 25 nm formed by deposition in ultra-high vacuum on graphite have been studied using synchrotron radiation. We have determined the island size by measuring the spectral dependence of the reflectivity of each sample using light in the photon energy range 35 eV to 190 eV. Comparing this measurement to a calculation of the reflectivity modified by the size-dependent extinction, determined using Mie theory, gives the average particle radius. Using this sizing technique we find that, for the growth of silver at room temperature, the average radius increases linearly with the cube root of evaporation time. Deposition with the substrate at elevated temperatures produces, for similar coverages, an increase in the average size for temperatures between 200°C and 225°C followed by a rapid decrease above 225°C. Chromium shows size effects in the photoemission spectra from the 3s and 3p core levels and the 3d valence band. With decreasing radius a decrease in the multiplet splitting of the 3s core level and a large change in the relative intensity among the 3p multiplets occurs indicating increased electron correlation as the system becomes smaller. A narrowing of the valence band also occurs which we attribute to a structural change at the surface of the particles.  相似文献

The fluctuating field of a heated jet excited by a sound pulse is simulated numerically. The fluctuations in both the flow field and the far field are studied. The flow field results depend crucially on the stability properties of the heated jet. The altered stability properties due to the heating cause a reduction in the overall fluctuations and a shift of the instability waves into lower frequencies. These changes cause a similar downward shift in the far field spectrum and a reduction in the total far field noise. The results provide a partial explanation in terms of stability theory of experimentally observed properties of the noise of heated jets.  相似文献
