Responsive Exosome Nano‐bioconjugates for Synergistic Cancer Therapy

Exosomes hold great potential in therapeutic development. However, native exosomes usually induce insufficient effects in vivo and simply act as drug delivery vehicles. Herein, we synthesize responsive exosome nano‐bioconjugates for cancer therapy. Azide‐modified exosomes derived from M1 macrophages are conjugated with dibenzocyclooctyne‐modified antibodies of CD47 and SIRPα (aCD47 and aSIRPα) through pH‐sensitive linkers. After systemic administration, the nano‐bioconjugates can actively target tumors through the specific recognition between aCD47 and CD47 on the tumor cell surface. In the acidic tumor microenvironment, the benzoic‐imine bonds of the nano‐bioconjugates are cleaved to release aSIRPα and aCD47 that can, respectively, block SIRPα on macrophages and CD47, leading to abolished “don't eat me” signaling and improved phagocytosis of macrophages. Meanwhile, the native M1 exosomes effectively reprogram the macrophages from pro‐tumoral M2 to anti‐tumoral M1.     

Irreversible Amide‐Linked Covalent Organic Framework for Selective and Ultrafast Gold Recovery

Design of stable adsorbents for selective gold recovery with large capacity and fast adsorption kinetics is of great challenge, but significant for the economy and the environment. Herein, we show the design and preparation of an irreversible amide‐linked covalent organic framework (COF) JNU‐1 via a building block exchange strategy for efficient recovery of gold. JNU‐1 was synthesized through the exchange of 4,4′‐biphenyldicarboxaldehyde (BA) in mother COF TzBA consisting of 4,4′,4′′‐(1,3,5‐triazine‐2,4,6‐triyl)trianiline (Tz) and BA with terephthaloyl chloride. The irreversible amide linked JNU‐1 gave good stability, unprecedented fast kinetics, excellent selectivity and outstanding adsorption capacity for gold recovery. X‐ray photoelectron spectroscopy along with thermodynamic study and quantum mechanics calculation reveals that the excellent performance of JNU‐1 for gold recovery results from the formation of hydrogen bonds C(N)?H???Cl and coordinate interaction of O and Au. The rational design of irreversible bonds as both inherent linkage and functional groups in COFs is a promising way to prepare stable COFs for diverse applications.

Plastic strain and dislocation structure of the KDP group crystals

Plastic strain of the KDP, DKDP, RDP, ADP crystals has been studied by the methods of uniaxial compression, selective etching, and indentation. It has been found that at room temperature the crystals had little plasticity, while at high temperature their plasticity was large. Rows of dislocations and dislocation rosettes have been observed. Glide systems and activation parameters of the process of plastic glide have been determined.

An ionothermal synthetic approach to porous polyoxometalate-based metal-organic frameworks

Pretty porous POMs: Ionothermal synthesis was applied to prepare porous POM-based open frameworks. The structural integrity remains unchanged until 300?°C; most importantly, the bulky tetrabutylammonium cations within their nanosized channels can be replaced by transition-metal ions through a cation-exchange process, and subsequent gas adsorption measurements confirm their permanent porosity.     

Recognition hysteresis of the acetylcholine receptor of torpedo Californica

The nicotinic acetylcholine receptor (nAcChR) of the electric organ of Torpedo californica fish exhibits a pronounced hysteresis loop in the high affinity binding of the neurotransmitter acetylcholine (AcCh). When increasing amounts of AcCh are added (pulse mode) an extremely long-lived, metastable conformer distribution is obtained (lower hysteresis branch) between low affinity AcCh binding states (R_l) and high (R_h) and very high (R_vh) affinity states. Dialysis conditions always lead to the equilibrium binding curve (upper hysteresis branch; K̄_A = 5 × 10⁻⁹M, 4°C; one A bound to the R-monomer of M_r ≈ 290 000). Cyclic, pulse mode addition and dilution of AcCh results in scanning loops within the main hysteresis. The kinetic analysis of the changes in free and bound AcCh during the open-system conditions of dialysis, that releases the metastability, shows that the AcCh (A) binding proceeds along an induced-fit pathway according to A+R_h ⇋ AR_n ⇋ AR_vh. The rate constant of the step AR_h → AR_vh is k₂ = 6 × 10⁻³s⁻¹ and that of the reverse step is k₋₂ = 3 × 10⁻⁴s⁻¹. Direct binding of A to free R_vh can be excluded. Therefore, the state R_vh does not preexist, it is induced and only stable, as AR_vh, by bound AcCh. The metastability can be described in terms of long-lived AR_vh ·R₁ hybrid dimers. Physiologically, the metastable hybrid may be viewed as a saving device: the functionally important, channel-active R₁ conformer is, at low AcCh-concentrations [A] < 1μM, prevented to convert to the desensitized states R_h and AR_vh. Furtheron, AcCh enhances the phosphorylation of phosphatidyl inositol and the auto-phosphorylation of the receptor. If the AcCh binding hysteresis causes a phosphorylation hysteresis the desensitized nAcChR may serve as a memory molecule of the transsynaptic information signalling of the neurotransmission.
