This paper reports a microfluidic system for biophysical characterization of red blood cells (RBCs) at a speed of 100-150 cells s(-1). Electrical impedance measurement is made when single RBCs flow through a constriction channel that is marginally smaller than RBCs' diameters. The multiple parameters quantified as mechanical and electrical signatures of each RBC include transit time, impedance amplitude ratio, and impedance phase increase. Histograms, compiled from 84,073 adult RBCs (from 5 adult blood samples) and 82,253 neonatal RBCs (from 5 newborn blood samples), reveal different biophysical properties across samples and between the adult and neonatal RBC populations. In comparison with previously reported microfluidic devices for single RBC biophysical measurement, this system has a higher throughput, higher signal to noise ratio, and the capability of performing multi-parameter measurements.  相似文献

A compound having the capability of releasing NO upon exposure to visible or near-infrared (vis or NIR) light could be a potential candidate for photodynamic therapy (PDT), which is significant for humans. Here, we investigated a series of Mn(II) complexes (a-d) based on density functional theory (DFT) to illuminate the mechanism of their behavior of releasing NO. Their structural, spectroscopic, and photodissociable properties were calculated by quantum theoretical methods to give a detailed and warranted explanation of the performance of releasing NO. The results indicate that, for a-d, releasing NO was attributed to the electron transfer from d(yz)/d(xz)(Mn) orbitals to π*(NO) orbitals at the second excited triplet state (T(2)). Importantly, we confirmed the finding in the experiment that d could release NO upon exposure to the NIR region and, thus, may be a best candidate for PDT in a-d. Therefore, to take d for example, the analyses of the potential energy curves (PECs) of difference states and electron density difference between the T(2) and the ground state (S(0)) were performed to further provide evidence of ligand dissociation and release of NO at the T(2) state. Finally, we hope that our discussion can provide assistance to understand the behavior of the release of NO and design novel photodissociable transition metal nitrosyls for PDT applications.  相似文献

We conducted a combined anion photoelectron spectroscopy and density functional theory study on the structural evolution of copper-doped silicon clusters, CuSi(n)(-) (n = 4-18). Based on the comparison between the experiments and theoretical calculations, CuSi(12)(-) is suggested to be the smallest fully endohedral cluster. The low-lying isomers of CuSi(n)(-) with n ≥ 12 are dominated by endohedral structures, those of CuSi(n)(-) with n < 12 are dominated by exohedral structures. The most stable structure of CuSi(12)(-) is a double-chair endohedral structure with the copper atom sandwiched between two chair-style Si(6) rings or, in another word, encapsulated in a distorted Si(12) hexagonal prism cage. CuSi(14)(-) has an interesting C(3h) symmetry structure, in which the Si(14) cage is composed by three four-membered rings and six five-membered rings.  相似文献

A water compatible molecularly imprinted polymers (MIPs) coated stir bar for bisphenol A(BPA) was prepared with 3,3',5,5'-tetrabromobisphenol A as the dummy template molecule in this study. The dummy molecularly imprinted polymers coated stir bar (DMIPs-SB) showed better selectivity than the bars coated with polydimethylsiloxane or non-imprinted polymers when used to extract BPA and its three analogues. The saturated adsorption amount of the DMIPs coating was 3.0 times over that of the non-imprinted polymers coating. To achieve the optimum extraction performance, several parameters, including extraction and desorption time, pH value, adsorption temperature and stirring speed were investigated. The high-performance liquid chromatography combined with the DMIPs-SB was employed in the analysis of BPA in aqueous solution. The linear range of BPA concentration in aqueous medium was 0.0228-2.28 ng/mL with correlation coefficient of 0.9994 and the detection limit was about 6.84 × 10(-3) ng/mL based on three times ratio of signal to noise. This method was directly applied to the determination of trace BPA in milk with satisfactory results.  相似文献

Two 3D coordination polymers, [Co(24)(OH)(12)(SO(4))(12)(ip)(6)(DMSO)(18)(H(2)O)(6)]·(DMSO)(6)(EtOH)(6)(H(2)O)(36) (1·guests, ip = isophthalate) and [Ni(24)(OH)(12)(SO(4))(12)(ip)(6)(DMSO)(12)(H(2)O)(12)]·(DMSO)(6)(EtOH)(6)(H(2)O)(20) (2·guests), constructed with nanosized tetraicosanuclear Co(II) and Ni(II) wheels are solvothermally synthesized. Fumonisins are produced by the fungus Fusarium verticillioides, which are known to cause fatal diseases in some animals and humans. Here, we describe a sensitive, reproducible and reliable analytical method for the quantitative determination of fumonisins B(1) (FB(1)) and B(2) (FB(2)) in 112 spices and aromatic and medicinal herbs marketed in China. This method is based on high performance liquid chromatography and fluorescence detection (HPLC-FLD) coupled to a new on-line post-column derivatization using ortho-phthaldialdehyde with 2-mercaptoethanol and immunoaffinity column clean-up. Under the optimized experimental conditions, a complete separation of FB(1) and FB(2) was obtained using a Synergi C(18) column and a gradient elution at 0.8 mL min(-1) with methanol and 0.1 M phosphate buffer at pH 3.15. The limits of detection for FB(1) and FB(2) were both 40 μg kg(-1). Good recoveries were found for spiked samples with FB(1) and FB(2), ranging from 82.34% to 98.16% for FB(1) and from 72.58% to 97.10% for FB(2), with relative standard deviation (RSD) < 7.0%. 5 spices, 11 aromatic herbs and 96 medicinal herbs including 93 normal samples and 19 visibly moldy samples, which were spoiled artificially, were analyzed. The results showed that 8 (42.1%) visibly moldy samples and 8 (8.6%) normal samples were contaminated with FB(1) at mean contents of 129.0 and 165.9 μg kg(-1), and with FB(2) at 1745.0 and 256.8 μg kg(-1), respectively. Positive confirmation of detected samples was performed by liquid chromatography tandem electrospray ionization mass spectrometry (LC-ESI-MS/MS), using a triple quadrupole analyzer and operated in the multiple reaction monitoring mode.  相似文献
