Crystals of Saccharomyces cerevisiae inorganic pyrophosphatase suitable for X-ray diffraction study were grown by cocrystallization of the enzyme with cobalt chloride and imidodiphosphate. Saccharomyces cerevisiae is a metal-dependent enzyme which catalyzes hydrolysis of inorganic pyrophosphate to orthophosphate. The three-dimensional structure of this enzyme was solved by the molecular-replacement method and refined at 1.8 Å resolution to an R factor of 19.5%. Cobalt and phosphate ions were revealed in the active centers of both identical subunits (A and B) of the pyrophosphatase molecule. In subunit B, a water molecule was found between two cobalt ions. It is believed that this water molecule acts as an attacking nucleophile in the enzymatic cleavage of the pyrophosphate bond. It was demonstrated that cobalt ions and a phosphate group occupy only part of the potential binding sites (two chemically identical and crystallographically independent subunits have different binding sites). The arrangement of ligands and the structure of the nucleophile-binding site are discussed in relation to the mechanism of action of the enzyme and the nature of the metal activator.  相似文献

A 1.3 ?m laser in a compact disc package is coupled to single-mode fibre using two graded-index lenses in a nearly confocal scheme. An increase in the alignment tolerance at the lens-lens interface by a factor of six suggests the design for a potentially very low-cost connectorised laser package. The coupling efficiency of approx. 6% (?12 dB) is shown to be adequate for local loop applications.  相似文献

Pyramidal- and wedge-absorber materials are used extensively in anechoic measurement chambers to attenuate stray signals. Typical absorber layouts result in large absorber walls in which the absorber tips and bases are roughly aligned in the same plane. Such a quasi-periodic configuration produces a strong coherent specular reflection which dominates the absorber scattered field. Based on the multisection impedance transformer concept, one can divide absorber elements into different levels (layers) so that this coherence can be destroyed to reduce the specular absorber scattering level. The synthesis of this desired behavior can be implemented by the Chebyshev transformer technique, which provides the largest bandwidth given a passband ripple threshold. The resulting reflected field is then the product of the original absorber response times the Chebyshev reduction factor, which is independent of polarization and absorber properties. Various measured results are used to show that more than a 10-dB improvement can be achieved at the critical low end of the frequency band using this approach. This improvement cannot be achieved using conventional design concepts unless the absorber size is doubled  相似文献

Let Ψ be any adaptive sampling algorithm that can run in real time on a tapeless multichannel electrocardiogram (ECG) Holter system. Simple methods which can significantly improve Ψ's fidelity are described and their results are compared in this paper. It is shown that by adding some simple tests to Ψ, the signals reconstructed by Ψ can be improved as much as 5.45 dB. It is also shown that under the same data rate, a good data compressor with slowly sampled input ECG is preferable to a bad data compressor with highly sampled input ECG  相似文献

Using an enhanced multiple output domino logic (EMODL) implementation of a carry lookahead adder (CLA), sums of several consecutive bits can be built in one nFET tree with a single carry-in. Based on this result, a new sparse carry chain architecture is proposed for the CLA adder. We demonstrate the design approach using a 32-b adder, and show that only four carries are sufficient for generating all sums, with a consequent reduction in the number of stage delays. Using a 1.2-μm CMOS technology, we verify our simulation procedures by fabrication and measurement of a 2.7 ns critical path  相似文献

A series active power filter working as a sinusoidal current source, in-phase with the mains voltage, has been developed and tested. The amplitude of the fundamental current in the series filter is controlled through the error signal generated between the load voltage and a pre-established reference. The control allows an effective correction of power factor, harmonic distortion and load voltage regulation. Compared with previous methods of control developed for series active filters, this method is simpler to implement because it is only required to generate a sinusoidal current, in-phase with the mains voltage, the amplitude of which is controlled through the error in the load voltage. The proposed system has been studied analytically and tested using computer simulations and experiments. In the experiments, it has been verified that the filter keeps the line current almost sinusoidal and in-phase with the line voltage supply. It also responds very quickly under sudden changes in load conditions, reaching its steady-state in about two cycles of the fundamental  相似文献

We describe the design and operation of a critically phase-matched femtosecond optical parametric oscillator based on KTA. By employing a small pump-signal noncollinear angle, tuning of the idler to beyond 4 μm is achieved using a Ti:sapphire pump laser. A Gaussian-beam model is described which can be used to identify the optimal noncollinear phase-matching geometry. Idler and signal pulses are characterized fully both temporally and spectrally and interferometric autocorrelation data at 3.5 μm showing idler pulses of only eight optical cycles duration are presented  相似文献

We have investigated the optical output of the free-electron laser for infrared experiments (FELIX) when it is driven by an electron beam with a ramped energy. We show that the applied slow ramp on the electron beam energy leads to a frequency chirp on each picosecond optical pulse. Typical values for the chirp are 0.2% frequency sweep across a 1.5-ps-long optical pulse. The optical pulses were analyzed with a double-grating pair and with a second-order autocorrelator. The pulse duration was reduced in the double-grating pair by 20%. A linear dependence of the chirp on the cavity desynchronization was measured  相似文献
