Auranofin (AF, hereafter) is an orally administered chrysotherapeutic agent approved for the treatment of rheumatoid arthritis that is being repurposed for various indications including bacterial infections. Its likely mode of action involves the impairment of the TrxR system through the binding of the pharmacophoric cation [AuPEt3]+. Accordingly, a reliable strategy to expand the medicinal profile of AF is the replacement of the thiosugar moiety with different ligands. Herein, we aimed to prepare the AF analogue bearing the acetylcysteine ligand (AF-AcCys, hereafter) and characterize its anti-staphylococcal activity. Biological studies revealed that AF-AcCys retains an antibacterial effect superimposable with that of AF against Staphylococcus aureus, whereas it is about 20 times less effective against Staphylococcus epidermidis. Bioinorganic studies confirmed that upon incubation with human serum albumin, AF-AcCys, similarly to AF, induced protein metalation through the [AuPEt3]+ fragment. Additionally, AF-AcCys appeared capable of binding the dodecapeptide Ac-SGGDILQSGCUG-NH2, corresponding to the tryptic C-terminal fragment (488–499) of hTrxR. To shed light on the pharmacological differences between AF and AF-AcCys, we carried out a comparative experimental stability study and a theoretical estimation of bond dissociation energies, unveiling the higher strength of the Au–S bond in AF-AcCys. From the results, it emerged that the lower lipophilicity of AF-AcCys with respect to AF could be a key feature for its different antibacterial activity. The differences and similarities between AF and AF-AcCys are discussed, alongside the opportunities and consequences that chemical structure modifications imply.  相似文献

Three analytical methods have been developed and compared for the quality control of a new formulation (Soymen GN(R) capsules) containing soy extract and melatonin for the treatment of menopausal symptoms. The first method is based on MEKC with diode-array detection, using a mixture of basic carbonate buffer (95%) and methanol (5%), containing 55 mM SDS, as the BGE. The second method is an HPLC method with UV detection at 260 nm. The third method is an HPLC method coupled to amperometric detection which is carried out at an oxidation potential of +0.8 V. In both HPLC systems, the chromatographic separation is obtained on an RP C18 column using a mixture of ACN and an acidic phosphate buffer (25:75 v/v) as the mobile phase. A feasible pretreatment procedure with a methanol/water mixture has been implemented to achieve the quantitative extraction of the main soy isoflavones and of melatonin from the capsules. The results obtained with the three methods are in good agreement with each other and satisfactory in terms of linearity (r(2) >0.9996), precision (RSD <5.4%) and accuracy (recovery >97%). Thus, each of the three analytical methods seems to be suitable for the simultaneous analysis of the main soy isoflavones and melatonin in the new commercial formulation.  相似文献

We study the diabatic crossing of two electronic surfaces from the time-dependent point of view, looking at the time and frequency-resolved spontaneous emission spectra and to the total population of the excited state, which for a pump and-probe experiment with two δ-like pulses, is proportional to the total emission. The total M shell and the Mk (k = ξ, αβ, γ, m) X‐ray production cross sections for 66Dy have been measured at incident photon energies across its Lj (j = 1–3) subshell absorption edge energies, ranging 7.8–9.2 keV. This study aims to investigate the evolution of the probability for cascade decay of Lj subshell vacancies as the tunable incident energy ionizes progressively different 66Dy Lj subshells. The experimental X‐ray production cross sections have been compared with theoretical ones calculated using the nonrelativistic Hartree–Fock–Slater (HFS) model‐based photoionization cross sections; three sets of the X‐ray emission rates, fluorescence and Coster–Kronig yield based on the nonrelativistic Hartree–Slater (NRHS) model, Dirac–Hartree–Slater (DHS) model and Dirac–Fock (DF) model; the Lj (j = 1–3) subshell to the Mi (i = 1–5) subshell vacancy transfer probabilities evaluated in the present work. Presently measured total M shell and the Mαβ X‐ray production cross sections are found to be significantly lower than the theoretical ones evaluated using physical parameters based on the relativistic Dirac–Fock/Dirac–Hartree–Slater model calculations, whereas a much better agreement is observed with respect to the NRHS model‐based calculations; however, the measured X‐ray production cross sections are still systematically lower than the NRHS values.  相似文献

We extend a new method to measure possible variation of the speed of light by using Baryon Acoustic Oscillations and the Hubble function onto an inhomogeneous pressure model of the universe. The method relies on the fact that there is a simple relation between the angular diameter distance maximum and the Hubble function (H) evaluated at the same maximum‐condition redshift, which includes the speed of light c. One limit of such a method was the assumption of the vanishing of spatial curvature (though, as it has been shown, a non‐zero curvature has negligible effect). In this paper, apart from taking into account an inhomogeneity, we consider non‐zero spatial curvature and calculate an exact relation between and H. Our main result is the evaluation if current or future missions such as Square Kilometer Array (SKA) can be sensitive enough to detect any spatial variation of c which can in principle be related to the recently observed spatial variation of the fine structure constant (an effect known as α‐dipole).  相似文献

We prove multiplicity of periodic solutions for a scalar second order differential equation with an asymmetric nonlinearity, thus generalizing previous results by Lazer and McKenna (1987) [1] and Del Pino, Manasevich and Murua (1992) [2]. The main improvement lies in the fact that we do not require any differentiability condition on the nonlinearity. The proof is based on the use of the Poincaré-Birkhoff Fixed Point Theorem.  相似文献

We provide a semilocal convergence analysis for Broyden’s method for approximating locally unique solutions of nonlinear operator equations. Using the majorant principle we show that under the same or weaker hypotheses, in combination with our new idea of recurrent functions, we can find weaker sufficient conditions for the convergence of Broyden’s method as well as finer error bounds on the distances involved, and a more precise information on the location of the solution than before (Broyden, Math. Comput. 19:577–593, 1965; Chen, Ann. Inst. Stat. Math. 42:387–401, 1990; Dennis, Nonlinear Functional Analysis and Applications, pp. 425–472, Academic Press, San Diego, 1971; Li and Fukushima, Ann. Oper. Res. 103:71–97, 2001). Numerical examples are also provided involving polynomial, integral, and differential equations.  相似文献
