Currently, there are no reliable biomarkers available that can aid early differential diagnosis of reactive arthritis (ReA) from other inflammatory joint diseases. Metabolic profiling of synovial fluid (SF)—obtained from joints affected in ReA—holds great promise in this regard and will further aid monitoring treatment and improving our understanding about disease mechanism. As a first step in this direction, we report here the metabolite specific assignment of 1H and 13C resonances detected in the NMR spectra of SF samples extracted from human patients with established ReA. The metabolite characterization has been carried out on both normal and ultrafiltered (deproteinized) SF samples of eight ReA patients (n = 8) using high-resolution (800 MHz) 1H and 1H─13C NMR spectroscopy methods such as one-dimensional 1H CPMG and two-dimensional J-resolved1H NMR and homonuclear 1H─1H TOCSY and heteronuclear1H─13C HSQC correlation spectra. Compared with normal SF samples, several distinctive 1H NMR signals were identified and assigned to metabolites in the 1H NMR spectra of ultrafiltered SF samples. Overall, we assigned 53 metabolites in normal filtered SF and 64 metabolites in filtered pooled SF sample compared with nonfiltered SF samples for which only 48 metabolites (including lipid/membrane metabolites as well) have been identified. The established NMR characterization of SF metabolites will serve to guide future metabolomics studies aiming to identify/evaluate the SF-based metabolic biomarkers of diagnostic/prognostic potential or seeking biochemical insights into disease mechanisms in a clinical perspective. 相似文献

The alkylative carboxylation of allenamide catalyzed by an N‐heterocyclic carbene (NHC)–copper(I) complex [(IPr)CuCl] with CO2 and dialkylzinc reagents was investigated. The reaction of allenamides with dialkylzinc reagents (1.5 equiv) and CO2 (1 atm.) proceeded smoothly in the presence of a catalytic quantity of [(IPr)CuCl] to afford (Z)‐α,β‐dehydro‐β‐amino acid esters in good yields. The reaction is regioselective, with the alkyl group introduced onto the less hindered γ‐carbon, and the carboxyl group introduced onto the β‐carbon atom of the allenamides. The first step of the reaction was alkylative zincation of the allenamides to give an alkenylzinc intermediate followed by nucleophilic addition to CO2. A variety of cyclic and acyclic allenamides were found to be applicable to this transformation. Dialkylzinc reagents bearing β‐hydrogen atoms, such as Et2Zn or Bu2Zn, also gave the corresponding alkylative carboxylation products without β‐hydride elimination. The present methodology provides an easy route to alkyl‐substituted α,β‐dehydro‐β‐amino acid ester derivatives under mild reaction conditions with high regio‐ and stereoselectivtiy. 相似文献

Spectrum sensing is the important function of cognitive radio and energy detection is the most popular technique used for spectrum sensing. Detection of the availability of unused spectrum for the secondary user becomes difficult when the channel is affected by composite multipath/shadowed fading. In this paper, the performance analysis of an Energy Detector in Hoyt/gamma composite fading channel with Maximum Ratio Combining employing micro-diversity is analyzed. Analytical expressions for performance parameters, i.e., the average probability of detection and the average area under the receiver operating characteristics curve are evaluate. The tetrapeptide (Bz-Phe(p-NPh2)-l-DOPA(protected)-l-Phe-l-Phe-OMe was designed to incorporate seven phenyl rings so that it’s conformation, self-assembly and application in Hg2+ ions sensing could be studied. Peptide molecules adopted an overlapping β-turn of type III/III conformation in crystals. The peptide showed a highly selective turn-on response towards mercuric ion over other metal ions with a 10-fold enhancement in fluorescence intensity. This intensity change coupled with the selectivity of the peptide towards mercury allowed us to demonstrate simple colorimetric dip sensing of Hg2+ ions. The technique provides a highly selective and effective way to detect Hg2+ ions. The peptide also self-assembled into nanospheres with diameter ranges from 100 to 500?nm. Mercuric ion coordination enabled these peptide nanospheres to aggregate into well-defined nanoparticles. The enhanced fluorescence upon Hg2+ addition demonstrates that peptide scaffolds can be exploited in the development of different selective sensors. 相似文献

Multiplicative fading statistics usually encountered in different radio propagation environments. In this context, we evaluate and analyse the performance of a wireless communication system over the nonidentical cascaded generalised Gamma Fading Channels, also known as generalised Bessel‐K fading channel. To this end, the closed‐form expressions for the amount of fading (AOF), the outage probability (OP), the average symbol error probability (SEP), and the channel capacity are derived. In addition approximate expressions for the average SEP with maximal ratio combining (MRC) diversity are also provided. The low‐ and high‐power solutions for the channel capacity are also provided. Furthermore, simplified asymptotic average SEP expressions for MRC and selection combining (SC) are presented to gain the system performance with diversity. The proposed methodologies provide more flexibility to accommodate different radio propagation scenarios. To examine the accuracy of the proposed solutions, numerical and simulation results are compared and shown to fit for variety of fading parameters. 相似文献
