Galactose oxidase (GO) is an enzyme that catalyzes two-electron oxidations. Its active site contains a copper atom coordinated to a tyrosyl radical, the biogenesis of which requires copper and dioxygen. We have recently studied the properties of electrochemically generated mononuclear Cu(II)-phenoxyl radical systems as model compounds of GO. We present here the solution chemistry of these ligands under various copper and dioxygen statuses: N(3)O ligands first chelate Cu(II), leading, in the presence of base, to [Cu(II)(ligand)(CH(3)CN)](+) complexes (ortho-tert-butylated ligands) or [(Cu(II))(2)(ligand)(2)](2+) complexes (ortho-methoxylated ligands). Excess copper(II) then oxidizes the complex to the corresponding mononuclear Cu(II)-phenoxyl radical species. N(2)O(2) tripodal ligands, in the presence of copper(II), afford directly a copper(II)-phenoxyl radical species. Addition of more than two molar equivalents of copper(II) affords a Cu(II)-bis(phenoxyl) diradical species. The donor set of the ligand directs the reaction towards comproportionation for ligands possessing an N(3)O donor set, while disproportionation is observed for ligands possessing an N(2)O(2) donor set. These results are discussed in the light of recent results concerning the self-processing of GO. A path involving copper(II) disproportionation is proposed for oxidation of the cross-linked tyrosinate of GO, supporting the fact that both copper(I) and copper(II) activate the enzyme.  相似文献

Photoinduced charge separation and recombination in a carotenoid-porphyrin-fullerene triad C-P-C(60)(1) have been followed by multifrequency time-resolved electron paramagnetic resonance (TREPR) at intermediate magnetic field and microwave frequency (X-band) and high field and frequency (W-band). The electron-transfer process has been characterized in the different phases of two uniaxial liquid crystals (E-7 and ZLI-1167). The triad undergoes photoinduced electron transfer, with the generation of a long-lived charge-separated state, and charge recombination to the triplet state, localized in the carotene moiety, mimicking different aspects of the photosynthetic electron-transfer process. Both the photoinduced spin-correlated radical pair and the spin-polarized recombination triplet are observed starting from the crystalline up to the isotropic phase of the liquid crystals. The W-band TREPR radical pair spectrum has allowed unambiguous assignment of the spin-correlated radical pair spectrum to the charge-separated state C(.+)-P-C(60)(.-). The magnetic interaction parameters have been evaluated by simulation of the spin-polarized radical pair spectrum and the spin-selective recombination rates have been derived from the time dependence of the spectrum. The weak exchange interaction parameter (J = +0.5 +/- 0.2 G) provides a direct measure of the dominant electronic coupling matrix element V between the C(.+)-P-C(60)(.-) radical pair state and the recombination triplet state (3)C-P-C(60). The kinetic parameters have been analyzed in terms of the effect of the liquid crystal medium on the electron-transfer process. Effects of orientation of the molecular triad in the liquid crystal are evidenced by simulations of the carotenoid triplet state EPR spectra at different orientations of the external magnetic field with respect to the director of the mesophase. The order parameter (S = 0.5 +/- 0.05) has been evaluated.  相似文献

X-ray crystal structures are reported of a free host compound 1, comprising two diphenylmethanol terminal groups attached to a central 9,10-ethynyl substituted anthracene unit, and of three inclusion compounds of a fluoren-9-ol substituted analogous host 2 with acetone, dimethyl sulphoxide (DMSO) and dimethylformamide (DMF) as guest, respectively. Despite the presence of two hydroxyl groups in 1, there is no O–H...O hydrogen bond between the molecules in the guest free crystal – only weaker C–H...O interactions and van der Waals' type connections. In the inclusion compounds of 2, H-bonded 1:2 host–guest associates are formed, where each of the host hydroxyl groups binds to a guest oxygen atom. The orientations of the host–guest connections in these complexes vary, being E for acetone and Z for both DMSO and DMF guests, relative to the host anthracene unit. The DMSO and DMF inclusion compounds of 2 proved to be isostructural.  相似文献

All methionine aminopeptidases exhibit the same conserved metal binding site. The structure of this site with either Co2+ions or Zn2+ions was investigated using density functional theory. The calculations showed that the structure of the site was not influenced by the identity of the metal ions. This was the case for both of the systems studied; one based on the X-ray structure of the human methionine aminopeptidase type 2 (hMetAP-2) and the other based on the X-ray structure of the E. colimethionine aminopeptidase type 1 (eMetAP-1). Another important structural issue is the identity of the bridging oxygen, which is part of either a water molecule or a hydroxide ion. Within the site of hMetAP-2 the results strongly indicate that a hydroxide ion bridges the metal ions. By contrast, the nature of the oxygen bridging the metal ions within the metal binding site of eMetAP-1 cannot be determined based on the results here, due to the similar structural results obtained with a bridging water molecule and a bridging hydroxide ion.  相似文献

The first systematic study on the use of tricyanoborates as ligands is presented. The tricyanoborates [RB(CN)3]- (R = oct and Ph) can be prepared by direct cyanation of RBCl2 precursors as well as by thermolysis of the corresponding isocyanides [RB(NC)3]-. The first organo-cyanogallates [RGa(CN)3]- (R = Bu, C6H2-2,4,6-Me3) were prepared from the corresponding dichloride, the structure of Et4N[mesGa(CN3] being confirmed crystallographically. The reaction of equimolar [RB(CN)3]- (R = oct, Ph) and [Cu(MeCN)4]+ afforded two-dimensional polymers [RB(CN)3Cu(NCMe)]. The sheets arise via conjoined hexagonal B3Cu3(CN)6 rings with chair conformations. The reaction of excess [PhB(CN)3]- and [Cu(MeCN)4]+ gives the polymer [K(18-crown-6)]{Cu[PhB(CN)3]2}. Treatment of [PhB(CN)3]- with [Cu(PCy3)2(NCMe)x]PF6 gave the one-dimensional polymer [PhB(CN)3Cu(PCy3)2], wherein two of the three BCN substituents are coordinated.  相似文献

Inelastic neutron scattering spectra of KHCO3 and CsH(NO3)2 have been obtained in the region 400 → 2400 and 400 → 2800 cm?1 respectively. The in- and out-of-phase bending vibrations of the hydrogen bonds have been observed and assigned. For CsH(NO3)2 the two bending modes are closer in frequency than in KHCO3 and they are not resolved from the antisymmetric stretch.  相似文献

Dipole-moment derivatives, calculated by both the CNDO/2 method with different parameterizations and the INDO method, are compared to the experimental values determined from absolute infrared intensity measurements for the IR active modes of methane, ethane, ethylene and acetylene. A parameter refinement procedure is introduced in which the CNDO/2 molecular orbital parameters are adjusted through a damped least-squares treatment to give best agreement with the observed dipole-moment derivatives. It is found that the refinement does not substantially improve the agreement obtained with the original CNDO/2 parameterization. The INDO method gives somewhat poorer agreement than the CNDO/2 calculations. As an example of the applicability of the molecular orbital methods toward reproducing relative infrared intensities, the spectrum of cyclopropane in the gasphase is examined.
Zusammenfassung Die Ableitungen des Dipol-Moments, die nach der CNDO/2-Methode mit verschiedenen Parametrisierungen sowie der INDO-Methode berechnet wurden, werden mit den experimentellen Ergebnissen aus Messungen der absoluten Infrarot-Intensitäten für die IR-aktiven Schwingungen von Methan, Äthan, Äthylen und Azetylen verglichen. Die CNDO/2-Parameter werden mit einer Methode der kleinsten Quadrate den beobachteten Dipol-Moment-Ableitungen angepaßt. Die Ergebnisse sind jedoch nicht wesentlich von denen der ursprünglichen CNDO/2-Methode verschieden. Die INDO-Ergebnisse sind nicht so gut wie die CNDO/2-Ergebnisse. Als Beispiel der Anwendbarkeit der MO-Methoden zur Berechnung von relativen IR-Intensitäten wird das Spektrum des Cyclopropans in der Gasphase untersucht.

Résumé Les dérivées du moment dipolaire, calculées par la méthode CNDO/2 avec différentes paramétrisations et par la méthode INDO, sont comparées aux valeurs expérimentales déterminées à partir de mesures d'intensité absolue pour les modes actifs dans l'infra-rouge dans le méthane, l'éthane, l'éthylène et l'acétylène. Les paramètres sont ajustés de manière à donner le meilleur accord avec les dérivées du moment dipolaire. Cet ajustement n'améliore pas sensiblement l'accord obtenu avec la paramétrisation CNDO/2 originale. La méthode INDO donne des résultats moins bons que les calculs CNDO/2. Le spectre du cyclopropane en phase gazeuse est étudié comme exemple de l'applicabilité de la méthode des orbitales moléculaires au calcul des intensités relatives infra-rouge.

The synthesis of dimetallic olefin complexes of the type L1M1C7H7M2L2 (M1 = Fe, Co, Rh; M2 = Rh, Ir, Pd; L1 = CO, C5H5; L2 = diene, allyl, P(OR)3) is described. The fluxional structures were investigated by 13C-, 57Fe- and 103Rh-NMR.-spectroscopy, and a cisoid dimetallic coordination, including a (metal, metal)-bond, can be deduced for the C7H7-ring. 57Fe- and 103Rh-chemical shifts give indications for the charge distribution in the 34e-complexes. The homodimetallic complex (Cp)Rh(tropone)Rh(Cp) ( 13 , Cp = cyclopentadienyl) and the corresponding 2-methoxytropone complex 14 were synthesized in addition to the above mentioned complexes. A fluxional bis(1-3-η-allyl)-coordination of the two Rh-atoms was derived from the temperature-dependent 13C-NMR.-spectra. A spin simulation of the (Cp)-multiplets of 12 and 13 yields information about (Rh, Rh)-spin-coupling which amounts to ≈5 Hz at 30°.  相似文献

A two-step, general synthesis of 1,4-benzodiazepine-2,5-diones (BZDs) is presented. This synthesis employs an Ugi four-component condensation using a convertible isocyanide (1-isocyanocyclohexene), followed by an acid-activated cyclization reaction. This synthesis represents a dramatically improved route to BZDs over those currently in the literature. In addition, since amino acids are not used as inputs, the potential for molecular diversity is much greater than that with existing syntheses. It was also found that BZDs substituted with methylenes at the C-3 and N-4 positions display conformational isomerism in the NMR spectra at room temperature. Variable-temperature NMR experiments support this observation and offer the interesting conclusion that the BZD core structure, in certain examples, might not be as rigid as previously supposed.  相似文献

A multivariate approach to the quantitative determination of 2-hydroxybenzaldehyde (salicylaldehyde), 3-hydroxybenzaldehyde, and 4-hydroxybenzaldehyde in their mixtures is described. The method is based on second order data generated in a flow injection analysis system with a pH gradient and photodiode-array detection. Each injection gave rise to an 89 (times) x 101 (wavelength) matrix, containing both the acidic and the basic characteristics of the sample injected. A least-squares algorithm based on Lambert-Beers law was used for the prediction of concentrations in unknown samples. No assumptions concerning the qualitative mixture composition of the hydroxybenzaldehydes were necessary to perform concentration predictions. The following four data types were used in the least-squares modelling: (1) unfolded raw data, (2) acidic spectra, (3) basic spectra, and (4) first spectral derivative of the raw data. The prediction errors obtained were comparable to literature results. A graphic method, based on the model residuals for detecting erroneous samples, was developed.  相似文献
