The purpose of this study was to develop mixed polymeric micelles with high drug loading capacity to improve the oral bioavailability of icaritin with Soluplus® and Poloxamer 407 using a creative acid-base shift (ABS) method, which exhibits the advantages of exclusion of organic solvents, high drug loading and ease of scaling-up. The feasibility of the ABS method was successfully demonstrated by studies of icaritin-loaded polymeric micelles (IPMs). The prepared IPMs were characterized to have a spherical shape with a size of 72.74 ± 0.51 nm, and 13.18% drug loading content. In vitro release tests confirmed the faster release of icaritin from IPMs compared to an oil suspension. Furthermore, bioavailability of icaritin in IPMs in beagle dogs displayed a 14.9-fold increase when compared with the oil suspension. Transcellular transport studies of IPMs across Caco-2 cell monolayers confirmed that the IPMs were endocytosed in their intact forms through macropinocytosis, clathrin-, and caveolae-mediated pathways. In conclusion, the results suggested that the mixed micelles of Soluplus® and Poloxamer 407 could be a feasible drug delivery system to enhance oral bioavailability of icaritin, and the ABS method might be a promising technology for the preparation of polymeric micelles to encapsulate poorly water-soluble weakly acidic and alkaline drugs.  相似文献

Sigma-1 (σ-1) receptor agonists are considered as potential treatment for stroke. TS-157 is an alkoxyisoxazole-based σ-1 receptor agonist previously discovered in our group. The present study describes TS-157 profile in a battery of tests for cerebral ischemia. Initial evaluation demonstrated the compound’s safety profile and blood–brain barrier permeability, as well as its ability to induce neurite outgrowth in vitro. The neurite outgrowth was shown to be mediated via σ-1 receptor agonism and involves upregulation of ERK phosphorylation (pERK). In particular, TS-157 also significantly accelerated the recovery of motor function in rats with transient middle cerebral artery occlusion (tMCAO). Overall, the results herein support the notion that σ-1 receptor agonists are potential therapeutics for stroke and further animal efficacy studies are warranted.  相似文献

Antiphospholipid syndrome (APS) is an autoimmune disease characterized by the production of β2-glycoprotein I (β2GPI)-dependent autoantibodies, with vascular thrombosis or obstetrical complications. Around 20% of APS patients are refractory to current treatments. Crassolide, a cembranoid diterpene extracted from soft corals, is a potential therapeutic candidate. Here, to examine the anti-inflammatory properties of crassolide, we first determined its effects on bone marrow-derived and splenic dendritic cells (DC). Specifically, we applied lipopolysaccharide (LPS) or β2GPI stimulation and measured the expressions of CD80 and CD86, and secretions of cytokines. We also determined in the OT-II mice, if bone marrow-derived DC was able to stimulate antigen-specific T cells. Moreover, we examined the therapeutic potential of crassolide postimmunization in a murine model of APS that depended on active immunization with β2GPI. The vascular manifestations were evaluated in terms of fluorescein-induced thrombi in mesenteric microvessels, whereas the obstetric manifestations were evaluated based on the proportion of fetal loss after pregnancy. We also measured blood titers of anti-β2GPI antibody, splenic cell proliferative responses and cytokine secretions after β2GPI stimulation ex vivo. Finally, we determined in these mice, hematological, hepatic and renal toxicities of crassolide. Crassolide after LPS stimulation suppressed DC maturation and secretion of tumor necrosis factor (TNF)-α, interleukin (IL)-6, IL-12 and IL-23, and downstream T cell activation. Crassolide could partially ameliorate both the vascular and obstetric manifestations of APS in BALB/c mice. Both blood titers of anti-β2GPI antibody and splenic cell proliferation after β2GPI stimulation were reduced. Splenic Th1 and Th17 responses were also lowered after β2GPI stimulation. Finally, within therapeutic doses of crassolide, we found no evidence of its toxicity. In conclusion, we showed the ability of crassolide to suppress DC and downstream T cell responses. Crassolide is therefore a potential candidate for adjunctive therapy in APS.  相似文献

Chitosan, a biopolymer possessing numerous interesting bioactivities and excellent technological properties, has received great attention from scientists in different fields including the food industry, pharmacy, medicine, and environmental fields. A series of recent studies have reported exciting results about improvement of the properties of chitosan using the Maillard reaction. However, there is a lack of a systemic review about the preparation, bioactivities and applications in food industry of chitosan-based Maillard reaction products (CMRPs). The presence of free amino groups in chitosan allows it to acquire some stronger or new functional properties via the Maillard reaction. The present review aims to focus on the current research status of synthesis, optimization and structural identification of CMRPs. The applications of CMRPs in the food industry are also discussed according to their biological and technological properties such as antioxidant, antimicrobial activities and inducing conformational changes of allergens in food. Some promising directions for future research are proposed in this review, aiming to provide theoretical guidance for the further development of chitosan and its derivatives.  相似文献

水资源匮乏是现代化发展中面临的全球性问题,太阳能界面水汽转换(Interfacial Solar Steam Generation, ISSG)是一种高效、绿色、低成本进行海水淡化和废水处理的方法。ISSG使用绿色的太阳能作为热源,通过光热转换并将热限制在水气界面上以高效产生蒸气,然后经过冷凝收集获得清洁水。设计和构筑具有强光吸收的光热转换材料是ISSG的技术核心。Ti3C2-MXene是一种新型二维碳化钛材料,具有比表面积大、水分散性好和光热转换效率高等优点,在ISSG领域具有巨大的应用潜力。本文介绍了ISSG技术和MXene,总结了光热转换材料的设计原则,论述了Ti3C2-MXene复合材料在ISSG领域的研究进展,其中包括二维MXene薄膜、三维MXene气凝胶和水凝胶、生物基-MXene复合材料的构筑和性能等,并分析了Ti3C2-MXene所面临的挑战和发展前景。  相似文献

Hierarchical porous tubular biochar (PTBC) was prepared by selectively removing lignin simply according to reverse the pyrolysis sequence of cellulose. The properties of the PTBC sample were characterized by XRD, SEM, TEM, Raman spectra, cyclic voltammetry and electrochemical impedance spectroscopy. The electrochemical performances of PTBC modified on the screen-printing electrode illustrated excellent detection of lead ions (lead (II)) in water with the linear range (0.5–120 μg/L) and the detection limit (0.02 μg/L) by in-situ bismuth film square wave anode stripping voltammetry.  相似文献

Effectively improving the mechanical properties and thermal resistance of epoxy shape‐memory polymers (ESMPs) without affecting their shape‐memory performance is necessary to expand these polymers in practical applications. In this article, modified multi‐walled carbon nanotubes (MWCNTs) were prepared and used as efficient reinforcement for enhancing the comprehensive properties of ESMPs. Increases of nearly 289% to 444% for impact strength and 112% to 184% for tensile force were obtained by adding only 0.1 to 1 wt% epoxy‐modified MWCNTs. The addition of unmodified and carboxyl‐modified MWCNTs was also investigated but showed less impact on the mechanical properties of the ESMPs than epoxy‐modified MWCNTs. Thermogravimetry analysis (TGA) and dynamic mechanical analyses (DMA) showed that less than 1 wt% modified MWCNTs can enhance the heat resistance of ESMPs greatly. Although the shape recovery time for composite materials increased upon adding the MWCNTs, the entire recovery time was still less than 1 minute, and the shape recovery rate was relatively high, nearly 100%.  相似文献

Lenticular printing technique provides a promising way to realize stereoscopic displays,especially,when microscopic optical structures are integrated into light-emitting materials/devices.Here,we fabricated large-area periodic structures with a spatial resolution at a wavelength scale from hybrid perovskite materials via a space-confined solution growth method.It takes advantages of both high refractive index contrast and high luminescence brightness,which allows the optical modulation on not only the reflection of illumination,but also the light emission from hybrid perovskites.The distributed feedback within these periodic structures significantly improves the degree of polarization and directionality of laser actions while their threshold is also reduced.These findings enable us to present a prototype of lenticular printing laser displays that vary emission colors at different view angles,which may find applications in creating high-resolution and high-contrast holographical images.  相似文献

CO2 is the main component of greenhouse gases and also an important carbon source. The hydrogenation of CO2 to methane using Ni-based catalysts can not only alleviate CO2 emissions but also obtain useful fuels. However, Ni-based catalysts face one major problem of the sintering of Ni nanoparticles in the process of CO2 methanation. Thus, this work has synthesized a series of efficient and robust nickel silicate catalysts (NiPS−X) with different nickel content derived from nickel phyllosilicate by the hydrothermal method. It was found that the Ni loading plays a critical role in the structure and catalytic performance of the NiPS−X catalysts. The catalytic performance gradually increases with the increase of Ni loading. In particular, the highly dispersed NiPS-1.6 catalyst with a high Ni loading of 34.3 wt% could obtain the CO2 conversion greater than 80%, and the methane selectivity was close to 100% for 48 h at 330 °C and the GHSV of 40,000 mL g−1 h−1. The excellent catalytic property can be assigned to the high dispersion of Ni nanoparticles and the strong interaction between the active component and the carrier, which is derived from a unique layered silicate structure with lots of nickel phyllosilicate and a large number of Lewis acid sites.  相似文献
