Xenotransplantation, the transplantation of cells, tissues or organs between individuals of different species, would resolve the current shortage of organs, but rejection remains the major hurdle to successful xenotransplantation. In the present study, we analyzed mixed lymphocyte reactions (MLRs) and used 51Cr release assays in order to identify the proliferation and expansion of mouse CD8+ cytotoxic T lymphocyte cells against PK15, PK15/pIL-18 or PK15/mIL-18 cells. In addition, we identified T cell populations in mouse splenocytes and lymph node cells using two-color flow cytometry. It was found that the CD8+ T cells of xenograft recipients proliferated extensively and that the survival rates of populations of PK15/mIL-18 or PK15/pIL-18 cells were higher than untransfected controls. Moreover, CD3+ T cells were increased in mice injected with PK15 cells or PK15/pIL-18 cells but PK15/pIL-18 cell numbers were lower in lymph nodes than untransfected controls. CD8+ T cells numbers were reduced in the lymph nodes of PK15/pIL-18 injected mice. These results suggest that porcine IL-18 regulates anti-pig cellular rejection in C57BL/6 mice.  相似文献

ERCC1 is a DNA repair gene and has been associated with resistance to DNA damaging agents. In this study we hypothesized that a polymorphism of ERCC1 Asn118Asn (C -> T) might affect the platinum-resistance of epithelial ovarian cancer patients to platinum-taxane chemotherapy administered postoperatively. Using the SNapShot assay, we assessed this polymorphism in ERCC1 in 60 ovarian cancer patients. Platinum-resistance was defined as progression on platinum-based chemotherapy or recurrence within 6 months of completing therapy. Although not significant, platinum-resistance was less frequently observed in patients with the C/T+T/T genotype (P=0.064). Multivariate analysis showed that the C/T+T/T genotypes constituted an independent predictive factor of reduced risk of platinum-resistance in ovarian cancer (odds ratio 0.17, 95% confidence interval 0.04-0.74, P=0.018, Fisher's exact test). No significant correlation was observed between overall survival and the ERCC1 polymorphism. Our results suggest that genotyping of the ERCC1 polymorphism Asn118Asn may be useful for predicting the platinum-resistance of epithelial ovarian cancer patients. However, these findings require prospective confirmation.  相似文献

A photoinduced charge transfer (PCT)-based 1,3-alternate calix[4]crown fluoroionophore containing two cation recognition sites, a crown ether ring and two facing pyreneamide groups, is synthesized. Upon addition of K+, Pb2+, or Cu2+, wavelength changes are observed in both the fluorescence and absorption spectra, but with different binding modes. With K+, fluorescence emissions of the ligand scarcely change, while addition of Pb2+ or Cu2+ produces a remarkable change in both the excimer and monomer emissions. The observed data indicate that the metal cation is encapsulated in the crown-5 ring for K+ and by the two facing amide groups in the latter case, which is verified by a metal ion exchange experiment. The wavelength shifts in both fluorescence and absorption spectra upon addition of Cu2+ show that, in contrast to Pb2+, Cu2+ interacts with the nitrogen atoms of the amide groups, resulting in a PCT mechanism.  相似文献

A chiral C2-symmetric NCN ligand, (5R,7R)-1,3-bis(6,6-dimethyl-5,6,7,8-tetrahydro-5,7-methanoquinolin-2-yl)benzene has been synthesized. A direct cyclometalation of this ligand with K2MCl4 (M = Pt, Pd) in dry acetic acid offered the corresponding pincer complexes, [(5R,7R)-1,3-bis(6,6-dimethyl-5,6,7,8-tetrahydro-5,7-methanoquinolin-2-yl)phenyl]platinum(II) chloride 5a and its palladium(II) analogue 5b. The Pt(II) and Pd(II) complexes 5 were characterized by NMR spectroscopy, and X-ray crystal structure analysis was done for the Pt(II) complex. The NMR data for both the complexes and X-ray crystal structural data for the chloro-Pt(II) complex indicate the existence of intramolecular C-H?Cl hydrogen bonding both in solution and in solid states. Chloride abstraction from 5a by treatment with silver triflate resulted in the corresponding triflate complex 6a, which generates the corresponding cationic aqua complex 7a in the presence of water molecules. The Pt(II) complex 6a/7a was used as asymmetric catalyst in the aldol reaction between methyl isocyanoacetate and aldehydes and also in the silylcyanation of aldehydes.  相似文献

In this paper, we describe design of a simple taste analyzing system using sensory system based on a multi-array chemical sensor (MACS) and personal digital assistant (PDA) for visual and quantitative analysis of different tastes using pattern recognition techniques. The sensory system is communicated with PDA, which has several interesting benefits for data analysis and display, via wireless using the Bluetooth. A various pattern recognition techniques are adapted including spider map, principal component analysis (PCA) and fuzzy C-means (FCM) algorithm to classify visually data patterns detected by the sensory system. The proposed techniques can be determined the cluster centers and membership grade of patterns through the unsupervised way. The membership grade of an unknown pattern, which does not shown previously, can be visually and analytically determined. Throughout the experimental trails, the taste analyzing system is demonstrated robust performance through data acquisition via wireless communication and visual and quantitative analysis of different tastes for the liquids. The system, which is implemented as a simple hand-held taste analyzing instrument, can be applicable to on-site taste monitoring.  相似文献

A porous hybrid inorganic/organic material, NaNi3(OH)(SIP)2 [SIP = 5-sulfoisophthalate][1], is shown to strongly bind molecular hydrogen at coordinatively unsaturated metal sites. A combination of H2 sorption isotherms, temperature programmed desorption, and inelastic neutron scattering spectroscopy show the existence of a considerable number of such strong binding sites in [1] along with other sites where hydrogen is more weakly physisorbed. The overall capacity for hydrogen of this material as well as the much stronger binding of hydrogen than in typical porous material represent an important step toward a possible utilization of porous media for hydrogen storage.  相似文献

Zeolite-intercalated semiconductor quantum dots (QDs) have long been proposed to give very high third-order nonlinear optical (3NLO) responses. However, measurements of their 3NLO responses have not been possible due to the lack of methods to prepare optically transparent QD-incorporating zeolite films supported on optically transparent substrates and to confine QDs only within zeolite interiors. We found that the zeolite-Y films grown on indium-tin-oxide-coated glass plates (Ygs) remain firmly bonded to the substrates during ion exchange with Pb2+ ions, drying, and formation of PbS QDs by treating Pb2+ ions with H2S. A series of Ygs encapsulating different numbers (n = 0, 8, 14, 23, and 33) of PbS in a unit cell [(PbS)n-Yg] were prepared. The PbS QDs were expelled by adsorbed moisture to the external surfaces, and the expelled QDs formed large QDs. Coating of the (PbS)n-Ygs with octadecyltrimethoxysilane results in effective confinement of the QDs within the internal pores. The zeolite-encapsulated PbS QDs showed remarkably high 3NLO activities at 532 and 1064 nm which are unparalleled by other PbS QDs dispersed in other matrixes.  相似文献
