Studies on the metabolism and detectability of the emerging drug of abuse diphenyl‐2‐pyrrolidinemethanol (D2PM) in rat urine using GC‐MS and LC‐HR‐MS/MS

In the last years, the number of new psychoactive substances, so‐called ‘legal highs’, has enormously increased. They are sold via online shops often with inaccurate and false information about the content. The aim of this work was to study the metabolism and the detectability of the drug of abuse diphenyl‐2‐pyrrolidinemethanol (D2PM) in rat urine using gas chromatography‐mass spectrometry and liquid chromatography‐high resolution‐tandem mass spectrometry. Five phase I and two phase II metabolites were identified suggesting hydroxylation at the pyrrolidine and diphenyl part as the main metabolic steps. Assuming similar kinetics, an intake of D2PM should be detectable in human urine mainly via its metabolites. Copyright © 2013 John Wiley & Sons, Ltd.     

Screening for exogenous androgen anabolic steroids in human hair by liquid chromatography/orbitrap-high resolution mass spectrometry

A method for the screening of various anabolic steroids and their esters in human hair, based on liquid-chromatography–high resolution mass spectrometry using an Exactive benchtop Orbitrap mass spectrometer, has been set up and validated. This method involved methanolic incubation of 30 mg of hair and analysis of the relevant extract in HPLC using a C18 column. The mass detector, with nominal resolving power of 100,000, operated in full scan mode in APCI under positive ionization mode. Analytes were identified by exact mass, correspondence of isotopic cluster and retention times.     

Doped Nano‐Sized Copper(I) Oxide (Cu2O) on Melamine?Formaldehyde Resin: a Highly Efficient Heterogeneous Nano Catalyst for ‘Click’ Synthesis of Some Novel 1H‐1,2,3‐Triazole Derivatives Having Antibacterial Activity

A facile and simple protocol for the 1,3‐dipolar cycloaddition of organic azides with terminal alkynes catalyzed by doped nano‐sized Cu₂O on melamine? formaldehyde resin (nano‐Cu₂O? MFR) as a new and convenient heterogeneous catalyst is described. In this method, ‘click’ cycloaddition of various structurally diverse β‐azido alcohols and alkynes in the presence of nano‐Cu₂O? MFR in H₂O/THF 1 : 2 furnished the corresponding 1,4‐disubstituted 1H‐1,2,3‐triazole adducts 1a – 1o in good to excellent yields at room temperature (Scheme and Table 3). The nano‐Cu₂O? MFR was characterized by scanning electron microscopy (SEM), X‐ray diffraction (XRD), inductively coupled plasma (ICP) analysis, and FT‐IR. The nano‐Cu₂O? MFR could be easily recovered and recycled from the reaction mixture and reused for many consecutive trials without significant decrease in activity (Table 4). The in vitro antibacterial activities of all synthesized compounds were tested on several Gram‐positive and/or Gram‐negative bacteria (Table 5). The results demonstrate the promising antibacterial activity for some of the synthesized compounds.     

Selective formation of non-conjugated olefins by samarium(II)-mediated elimination/isomerization of allylic benzoates

Aromatic allylic benzoates can be selectively transformed to the corresponding benzoate eliminated olefin by the action of samarium diiodide. Depending on the substrate and the elimination conditions, high selectivity for the non-conjugated alkene product can be achieved.     

Evaluation of Pesticide Contamination of Ground Water in Two Agricultural Areas of Portugal

In two Portuguese agricultural areas, "Beira Litoral" and "Ribatejo e Oeste", several pesticides regularly applied in vineyards, maize, potato, tomato for industry, apple, pear and rice were detected in ground water. Atrazine was the most frequently detected, being found in 70% of the total of 79 sites selected in the year 2000, followed by its metabolites desethylatrazine and desisopropylatrazine with frequencies of detection, respectively, of 56% and 48% and by simazine (37%), alachlor (25%), metolachlor (24%) and metribuzin (15%). Other pesticides and metabolites i.e. 3,4-dichloroaniline, dimethoate, f and g -endosulfan, lindane, molinate and prometryn were also detected but at lower occurrences. Pesticides were detected mainly in ground water wells used for irrigation purposes, although in some locations they were also found in water wells used for human consumption. In this study, it was also observed a seasonal variation of pesticide residues in ground water of shallow and deep wells.     

Synthesis and computational study of two new glycoluril clips containing benzocrown ether side walls

Two new glycoluril-derived molecular clips containing benzocrown ether side walls have been synthesized via reaction of a glycoluril scaffold with two bromomethylated benzocrown ethers. The molecular geometry of their most stable structures were investigated with density functional theory at the B3LYP level of theory using STO-3G, 6-31G, and 6-311G basis sets. Then based on the obtained computer-optimized structures, the binding properties of one clip with some 5-substituted resorcinols have been calculated.     

Synthesis, computational study and binding properties of some structurally rigid glycoluril-derived molecular clips

Several new clip-shaped molecules, with different degrees of steric hindrance around their cavities as well as more rigidity on their glycoluril scaffold, have been synthesized and the molecular geometry of their most stable structures has been investigated and optimized with density functional theory (DFT) at the B3LYP level of theory using 6-311G basis set. The affinity of these molecular clips for some dihydroxybenzene derivatives (guests) has been computationally and experimentally studied. The clips having dimethyl-substituted aromatic side walls, showed the best interaction energies towards the mentioned guests. Also, the computational results revealed that the more electron-deficient guest interacts more strongly with the clips (hosts).
