The following review describes the development of mass spectrometry off-line and on-line coupled with liquid chromatography to the analysis of food proteins. It includes the significant results recently obtained in the field of milk, egg and cereal proteins. This paper also outlines the research carried out in the area of food protein hydrolysates, which are important components in foodstuffs due to their functional properties. Silicone rubber (SIR) shows superior performance when used outdoors, but its surface can be transformed frominherently hydrophobic to hydrophilic by the adsorption of contaminants. Al(OH)_3, Al_2O_3, quartz powder and active carbonwere selected as authentic contaminants. Hydrophobicity of the surface was determined using contact angle measurement.The results indicate that the adsorbability of the contaminants can strongly affect the hydrophobicity of contaminated SIRsurface. The increasing rate of contact angle of specimens contaminated by Al(OH)_3 was much faster than that by Al_2O_3 andquartz due to the adsorption of migrated low molecular weight (LMW) polydimethylsiloxanes. Specimens contaminated byactive carbon could achieve sunde hydrophobicity within 15 min because active carbon has high adsorbability. Surfaces ofcontaminated ultrapure SIR, polytetrafluoroethylene (PTFE) and glass remain hydrophilic because they contain no mobileLMW components. Mass spectrometry (MS) was used for the characterization of recombinant human α-2 interferon (α-2 IFN) produced in E. coli. After purification by monoclonal antibody affinity chromatography, α-2 IFN showed two major peaks in reversed-phase liquid chromatography (RP- LC). Each component was digested with trypsin and Staphylococcus aureus protease V8, separately or in tandem, and the peptide mixture was analysed by MS without further purification. The first peak corresponded to the 165 amino acid sequence of human α-2 IFN and the main component of the second peak was the acetylated Cys1 α-2 IFN. It was also possible to verify by MS the location of the SS bonds in α-2 IFN and the occurrence of incorrect SS bridges in the products of some renaturation processes. The best renaturation process for obtaining a product without adducts or scrambling of disulphide bonds could be found by using RP-LC and fast atom bombardment MS. 相似文献

Several structures of pi complexes of isocyanuric acid and of several thio derivatives with anions have been computed by using high level ab initio calculations. The nature of the complexes has been studied by means of the method of molecular interaction potential with polarization (MIPp) and Bader's theory of atoms-in-molecules. These molecules form favorable complexes with anions and can be used as binding units for building receptors for the molecular recognition of anions. Multiphoton excitation and dissociation of SO(2) have been investigated in the wavelength range from 224 to 232 nm. Strong evidence is found for two-photon excitation to the H Rydberg state, followed by dissociation to SO + O and ionization of the SO product by absorption of a third photon. The two-photon excitation is resonantly enhanced via the C (1)B(2) intermediate state, and the two-photon yield spectrum thus bears a strong resemblance to the spectrum of this intermediate. Imaging of the O((3)P(2)), S((1)D(2)), and SO products suggests that, following dissociation of SO(2) from the H state, SO is produced in the A and B electronic states. S((1)D(2)) is produced both from two-photon dissociation of SO(2) to give S((1)D(2)) + O(2) and by single-photon dissociation of SO(+).
