The article describes a systematic study to overcome the matrix effect during chromatographic analysis of gemfibrozil, rivastigmine, telmisartan and tacrolimus from biological fluids using LC–ESI–MS/MS. All four methods were thoroughly developed by the appropriate choice of analytical column, elution mode and pH of mobile phase for improved chromatography and overall method performance. Matrix effect was assessed by post-column analyte infusion, slope of calibration line approach and post-extraction spiking. The best chromatographic conditions established were: Acquity BEH C18 (50 × 2.1 mm, 1.7 μm) column with 5.0 mm ammonium acetate, pH 6.0–methanol as the mobile phase under gradient program for gemfibrozil; Luna CN (50 × 2.0 mm, 3 μm) column with a mobile phase consisting of acetonitrile–10 mm ammonium acetate, pH 7.0 (90:10, v/v) for rivastigmine; Inertsustain C18 (100 × 2.0 mm, 5 μm) column using methanol–2.0 mm ammonium formate, pH 5.5 (80: 20, v/v) as the mobile phase for isocratic elution of telmisartan; and Acquity BEH C18 (50 × 2.1 mm, 1.7 μm) with methanol–10 mm ammonium acetate, pH 6.0 (95:5, v/v) as mobile phase for tacrolimus. The methods were thoroughly validated as per European Medicines Agency and US Food and Drug Administration guidance and were successfully applied for pharmacokinetic studies in healthy subjects.  相似文献

A practical, sensitive, and robust UPLC–MS/MS method was developed and validated to quantify lacosamide in human plasma. A simple one-step protein precipitation was used to extract lacosamide and labeled lacosamide-13C, D3 as an internal standard (IS) from 150-μL plasma. The extracts were analyzed on an Eclipse Plus C18 column (50 × 2.1 mm, 1.8 μm) using 0.1% formic acid in water and methanol:acetonitrile (50:50, v/v) under gradient conditions. The extracts were quantified on LCMS-8040 using electrospray ionization source operated in positive ionization and multiple reaction monitoring modes. The method showed good linearity from 0.02 to 20 μg/mL, which was adequate to cover lacosamide concentration assayed in formulations with different strengths. The bioanalytical assay was fully validated as per current regulatory guidelines. The intra-batch and inter-batch precision values of lacosamide were less than 4.6%. Lacosamide was found to be stable at different storage conditions. The extraction recoveries and IS-normalized matrix factors for lacosamide ranged from 97.17 to 99.68% and from 0.973 to 1.012, respectively. The validated method was successfully applied to a pharmacokinetic study with three lacosamide formulations (50, 100, and 200 mg) in 36 healthy subjects. The assay reliability was determined by reanalysis of 81 subject samples.  相似文献

Imbalance of zinc ion (Zn2+) in human body causes diseases like Alzheimer’s and Parkinson’s and therefore Zn2+ estimation in biological fluids has diagnostic values. Fluorescence “off-on” sensors have advantages of high sensitivity and in situ application over other sensors. A new fluorescent “off-on” Zn2+ sensor, N-benzoate-N′ salicylaldehyde ethynelediamine (L), has been synthesisied. In 1:1(v/v) CH3OH:PBS (PBS?=?phosphate buffer solution), L shows ca. 20 times enhancement in fluorescence intensity on interaction with Zn2+, due to snapping of photoinduced electron transfer (PET) process, which is selective over metal ions - Na+, K+, Ca2+, Ni2+, Cu2+, Cd2+, Hg2+ and Pb2+. These metal ions either individually or all together does not interfere the sensing ability of L towards Zn2+. A 1:1 interaction between L and Zn2+ ion with binding constant 104.25 has been established from spectroscopic data.  相似文献

We have successfully deposited nanostructured cadmium selenide (CdSe) inside anodic aluminum oxide (AAO) templates from aqueous electrolyte containing cadmium sulfate, selenium oxide and mercuric chloride by using high frequency alternating current (19 Vrms, 200 Hz). Addition of mercury ions aids in the deposition of CdSe inside anodic oxide pores. Scanning and transmission electron microscopy studies of the deposit etched in phosphoric acid showed the presence of end standing nanostrips. Energy dispersive X-ray analysis of the deposit confirmed the presence of cadmium and selenium in the deposit. X-ray diffraction (XRD) studies of the deposit showed small but broad diffraction peaks consistent with the presence of hexagonal CdSe. Optical studies revealed blue shift in band gap energy due to quantum confinement.  相似文献

Present investigation demonstrates a very simple seed-mediated route, using hydroxypropyl methyl cellulose (HPMC) as stabilizing agent, for the synthesis of silver nanodiscs in aqueous solution. Central to the concept of seed-mediated growth of nanoparticles is that small nanoparticle seeds serve as nucleation centres to grow nanoparticles to a desired size and shape. It is found that the additional citrate ions in the growth solution play the pivotal role in controlling the size of silver nanodiscs. Similar to the polymers in the solution, citrate ions could be likewise dynamically adsorbed on the growing silver nanoparticles and promote the two-dimensional (2D) growth of nanoparticles. Morphological, structural, and spectral changes associated with the seed-mediated growth of the nanoparticles in the presence of HPMC are characterized using UV–vis and TEM spectroscopic studies. Metal nanoparticles have received increasing attention for their peculiar capability to control local surface plasmon resonance (LSPR) when interacting with incident light waves. Extensive simulation study of the UV–vis extinction spectra of our synthesized silver nanodiscs has been carried out using discrete dipole approximation (DDA) methodology.  相似文献

An efficient synthesis of N-Boc-tubuphenylalanine benzyl ester (N-Boc-Tup-OBn, 1a) and N-Boc-epi-tubuphenylalanine benzyl ester (N-Boc-epi-Tup-OBn, 1b) is reported herein. Regioselective aziridine 4 ring opening with carbon nucleophiles followed by hydroboration of 1,1-substituted aminoalkene 3 using 9-BBN and subsequent oxidation in an alkaline medium are used as the key steps to provide N-tosyl 1,4-aminoalcohols. The 1,4-aminoalcohols are successfully transformed into the desired products with an overall yield of 23% for 1a and 11% for 1b over 8 consecutive steps separately.  相似文献
