Direct current electrokinetic systems generally require Faradaic reactions to occur at a pair of electrodes to maintain an electric field in an electrolyte connecting them. The vast majority of such systems, e.g. electrophoretic separations (capillary electrophoresis) or electroosmotic pumps (EOPs), employ electrolysis of the solvent in these reactions. In many cases, the electrolytic products, such as H+ and OH? in the case of water, can negatively influence the chemical or biological species being transported or separated, and gaseous products such as O? and H? can break the electrochemical circuit in microfluidic devices. This article presents an EOP that employs the oxidation/reduction of the conjugated polymer poly(3,4-ethylenedioxythiophene), rather than electrolysis of a solvent, to drive flow in a capillary. Devices made with poly(3,4-ethylenedioxythiophene) electrodes are compared with devices made with Pt electrodes in terms of flow and local pH change at the electrodes. Furthermore, we demonstrate that flow is driven for applied potentials under 2?V, and the electrodes are stable for potentials of at least 100?V. Electrochemically active electrodes like those presented here minimize the disadvantage of integrated EOP in, e.g. lab-on-a-chip applications, and may open new possibilities, especially for battery-powered disposable point-of-care devices.  相似文献

A range of surfaces coated with spherical silica particles, covering the size range from nanometer to micrometer, have been produced using Langmuir-Blodgett (LB) deposition. The particles were characterized both in suspension and in the Langmuir trough to optimize the surface preparation procedure. By limiting the particle aggregation and surface layer failures during the preparation steps, well-defined monolayers with a close-packed structure have been obtained for all particle sizes. Thus, this procedure led to structured surfaces with a characteristic variation in the amplitude and spatial roughness parameters. In order to obtain robust surfaces, a sintering protocol and an AFM-based wear test to determine the stability of the deposited surface layer were employed. Hydrophobization of the LB films followed by water contact angle measurements showed, for all tested particle sizes, the same increase in contact angle compared to the contact angle of a flat hydrophobic surface. This indicates nearly hexagonal packing and gives evidence for nearly complete surface wetting of the surface features.  相似文献

We here report a remarkably slow rearrangement of binding modes for a binuclear ruthenium(II) complex upon interaction with DNA. It has been previously shown that Delta,Delta-[mu-(11,11'-bidppz)(phen)4Ru2]4+ binds to DNA in one of the grooves. However, we find that this is only an initial, metastable, binding mode, which is extremely slowly reorganized into an intercalative binding geometry. The slow rearrangement and dissociation, revealed by flow linear dichroism and fluorescence spectroscopy, are concluded to be a result from the complex being threaded through the DNA, with one of the bridging aromatic dppz ligands intercalated between the base pairs of the DNA, placing one metal center in the minor groove and one in the major groove. A negative LD, a high luminescence quantum yield, and long luminescence lifetimes, similar to the intercalating complex Delta-[Ru(phen)2dppz]2+, indicate intercalation of the bidppz moiety. The unique slow dissociation of the complex in its final DNA-binding mode suggests that this class of threading, partially intercalated binuclear complexes may be interesting in the context of cancer therapy. Also, their unique optical and photophysical properties could make such complexes, either alone or scaffolded by DNA structures, of interest for the development of nanometer-sized molecular optoelectronic devices.  相似文献

Enzymatic hydrolysis of cellulose provides a renewable source of monosaccharides for production of variety of biochemicals and biopolymers. Unfortunately, the enzymatic hydrolysis of cellulose is often incomplete, and the reasons are not fully understood. We have monitored enzymatic hydrolysis in terms of molecular density, ordering and autofluorescence of cellulose structures in real time using simultaneous CARS, SHG and MPEF microscopy with the aim of contributing to the understanding and optimization of the enzymatic hydrolysis of cellulose. Three cellulose-rich substrates with different supramolecular structures, pulp fibre, acid-treated pulp fibre and Avicel, were studied at microscopic level. The microscopy studies revealed that before enzymatic hydrolysis Avicel had the greatest carbon-hydrogen density, while pulp fibre and acid-treated fibre had similar density. Monitoring of the substrates during enzymatic hydrolysis revealed the double exponential SHG decay for pulp fibre and acid-treated fibre indicating two phases of the process. Acid-treated fibre was hydrolysed most rapidly and the hydrolysis of pulp fibre was spatially non-uniform leading to fractioning of the particles, while the hydrolysis of Avicel was more than an order of magnitude slower than that of both fibres.  相似文献

The mechanism for methane formation in methyl-coenzyme M reductase (MCR) has been investigated using the B3LYP hybrid density functional method and chemical models consisting of 107 atoms. The experimental X-ray crystal structure of the enzyme in the inactive MCR(ox1)(-)(silent) state was used to set up the initial model structure. The calculations suggest a mechanism not previously proposed, in which the most remarkable feature is the formation of an essentially free methyl radical at the transition state. The reaction cycle suggested starts from a Michaelis complex with CoB and methyl-CoM coenzymes bound and with a squareplanar coordination of the Ni(I) center in the tetrapyrrole F(430) prosthetic group. In the rate-limiting step the methyl radical is released from methyl-CoM, induced by the attack of Ni(I) on the methyl-CoM thioether sulfur. In this step, the metal center is oxidized from Ni(I) to Ni(II). The resulting methyl radical is rapidly quenched by hydrogen-atom transfer from the CoB thiol group, yielding the methane molecule and the CoB radical. The estimated activation energy is around 20 kcal/mol, which includes a significant contribution from entropy due to the formation of the free methyl. The mechanism implies an inversion of configuration at the reactive carbon. The size of the inversion barrier is used to explain the fact that CF(3)-S-CoM is an inactive substrate. Heterodisulfide CoB-S-S-CoM formation is proposed in the final step in which nickel is reduced back to Ni(I). The suggested mechanism agrees well with experimental observations.  相似文献

The relative partial molar enthalpies of the components in the system N-pentanol-sodium N-octanoate-water (C5OH-NaC8-H2O) have been determined in the lamellar (D-) and hexagonal (E-) liquid crystalline phases formed in this system at 25°C. The enthalpies are correlated with Bragg spacings and earlier nmr studies. They indicate that (a) the minimum amount of water needed to hydrate the polar groups limits the region of existence of both D- and E-phase towards low water contents, (b) the interaction between -OH and -COO is especially important in stabilizing the D-phase containing large amounts of water, (c) the Bragg spacings and the enthalpies both change their concentration dependence markedly when the maximum amount of water that can be bound by primary hydration of the polar groups and the counter ions in the D-phase is exceeded, (d) enthalpies of transition from one phase to another are small compared to enthalpy changes with composition within the phases; the same holds for Bragg spacings. Hence, the composition of the sample (above all, the mole fraction of NaC8) is more important than the phase structure in determining these properties. It is obvious that further studies of enthalpies in similar systems can give important information on the factors governing phase stabilities and structural changes within the phases.  相似文献

Powder pattern indexing can still be a challenge, despite the great recent advances in theoretical approaches, computer speed and experimental devices. More plausible unit cells, belonging to different crystal systems, are frequently found by the indexing programs, and recognition of the correct one may not be trivial. The task is, however, of extreme importance: in case of failure a lot of effort and computing time may be wasted. The classical figures of merit for estimating the unit‐cell reliability {i.e.M20 [de Wolff (1968). J. Appl. Cryst. 1 , 108–113] and FN [Smith & Snyder (1979). J. Appl. Cryst. 12 , 60–65]} sometimes fail. For this reason, a new figure of merit has been introduced in N‐TREOR09, the updated version of the indexing package N‐TREOR [Altomare, Giacovazzo, Guagliardi, Moliterni, Rizzi & Werner (2000). J. Appl. Cryst. 33 , 1180–1186], combining the information supplied by M20 with additional parameters such as the number of unindexed lines, the degree of overlap in the pattern (the so‐called number of statistically independent observations), the symmetry deriving from the automatic evaluation of the extinction group, and the agreement between the calculated and observed profiles. The use of the new parameters requires a dramatic modification of the procedures used worldwide: in the approach presented here, extinction symbol and unit‐cell determination are simultaneously estimated. N‐TREOR09 benefits also from an improved indexing procedure in the triclinic system and has been integrated into EXPO2009, the updated version of EXPO2004 [Altomare, Caliandro, Camalli, Cuocci, Giacovazzo, Moliterni & Rizzi (2004). J. Appl. Cryst. 37 , 1025–1028]. The application of the new procedure to a large set of test structures is described.  相似文献

Chemically crosslinked gels made by polymerization and copolymerization of trimethylolpropane trimethacrylate have been characterized. Bimodal pore size distributions were obtained. The radii of the large pores (100 Å to 1 μm) varied with concentration while the small ones (20 to 30 Å) were largely independent of the concentration of TRIM. A mechanism of formation of the pores is outlined. Physically crosslinked gels of polyacrylonitrile with highly porous structures were made by cooling polymer solutions in DMF containing varying amounts of water. By controlling the thermal treatment the pore sizes could be varied from 0.1 to 1 μm. Finally the surface modification of cellulose fibers with derivatives of dichlorotriazine is described. It is shown that derivatives containing double bonds in the side chain can form covalent bonds across the interface between cellulose and an unsaturated polyester.  相似文献
