根据B组轮状病毒(GBRV)WH-1株nsp2基因的全序列,设计引物,用PCR的方法扩增得到nsp2基因的编码区.将其片段克隆到原核表达载体pGEX-KG上,经IPTG诱导,在E.coliDH5α菌株中得到高效表达.表达的GST融合蛋白大小为61×103,经SDS-PAGE分离纯化,免疫小白鼠制备了多克隆抗体.抗体经1∶3 000倍稀释后用于Western Blot分析,获得特异性显色信号.所表达的蛋白和制备的抗体可用于蛋白结构和功能的进一步研究.  相似文献

We present a compact integrated fiber laser with more than 200 mW of output power. It combines polarized fiber output with very narrow linewidth of less than 2 kHz. The coherence length of the laser is measured to be longer than 5 km. The laser features high mode stability of less than /spl plusmn/10 MHz over hours. The relative intensity noise (RIN) spectrum is dominated by a peak at the relaxation oscillation frequency and is shot-noise limited otherwise. The RIN peak at 1 MHz is reduced to /spl sim/-130 dB/Hz by integrating a negative feedback circuit. In addition to thermal wavelength tuning, the laser frequency can be modulated at a bandwidth of up to 10 kHz via the piezoelectric effect.  相似文献

In this work, the nonisothermal crystallization and subsequent melting behaviors of polypropylene (PP) nucleated with different nucleating agents (NAs) have been studied. α‐phase NA 1,3:2,4‐bis (3,4‐dimethylbenzylidene) sorbitol (DMDBS, Millad 3988), β‐phase NA aryl amides compound (TMB‐5), and their compounds were introduced into PP matrix, respectively. The results show that the nonisothermal crystallization behaviors and crystalline structures of PP with compounded NAs are dependent on the composition of NAs. In the sample of PP with 0.1 wt % DMDBS and 0.1 wt % TMB‐5, the nucleation efficiency (NE) of TMB‐5 is much higher than that of DMDBS and PP crystallizes mainly nucleated by TMB‐5, and in this condition, β‐phase PP is the main crystallization structure. For the sample of PP with 0.2 wt % DMDBS and 0.2 wt % TMB‐5, 0.2 wt % DMDBS has higher NE than 0.2 wt % TMB5, and α‐phase is the main crystalline structure. The cooling rate is proved to be very important in controlling the nonisothermal crystallization behavior and the final crystalline structure of nucleated PP. © 2008 Wiley Periodicals, Inc. We present a novel means of transporting molecules in solution by applying a zero-time-average alternating motive force to the molecules, and perturbing the molecular drag coefficient synchronously with the applied force, thus causing a net drift in a direction determined by the phase of the alternating drag perturbation relative to the alternating force. We apply an electrophoretic form of the method to transport and concentrate DNA in a gel, such that all molecules migrate on average away from the nearest electrode and toward a central region. Since an electrode does not occupy this central region, this method presents the possibility of transporting and focusing DNA and other charged molecules in regions free from electrodes and the associated electrochemistry.  相似文献

A new heterometallic MOF compound, [ZnNa(m-BDC)2].NH2(CH3)2 (MOF-CJ2), has been solvothermally synthesized and characterized by single-crystal X-ray diffraction, X-ray powder diffraction, ICP, TGA, IR, and photoluminescence spectroscopy analyses. It crystallizes in monoclinic space group C2/c (No. 15) with a=13.277(6) A, b=14.323(7) A, c=11.328(6) A, and beta=111.73(2) degrees. Its framework is constructed from M-O-C (M=Na and Zn) rods composed of alternating six-coordinated Na(I) centers and four-coordinated Zn(II) centers. These rods are linked by m-BDC links to form primitive cubic (pcu) type rod packing. This framework exemplifies the first open-framework heterometallic MOF structure based on the assembly of infinite rod building units.  相似文献

The ESR of diluted Er in Au thin films of about 1.000 Å thick, epitaxially grown on NaCl (001) faces, has been measured. The observed anisotropy on the g value and linewidth of the Er resonance are interpreted in terms of strains on the film induced by the substrate. A lower limit for the second order orbit-lattice coupling parameter is estimated.  相似文献

The concern regarding the effect of chemicals on the environment has increased considerably in recent years. Nowadays, technological developments in the coating industry are largely influenced by environmental issues and subsequent legislation. One of these issues is the tendency to replace cobalt as a catalyst with more environmentally friendly alternatives, because studies have indicated possible carcinogenicity. Not much knowledge is available on the effects of catalysts (driers) on the in-depth drying of alkyd coatings. Therefore we have studied the effect of cobalt as a primary drier combined with Ca and Zr as secondary driers on the in-depth curing of high solid solvent-borne alkyds. The profiling of the curing of alkyd coatings is performed with a new high-spatial-resolution NMR setup. In this study, two effects observed in the solvent-borne alkyd coatings are investigated. One is that when Ca and Zr are added as secondary driers the speed of the observed cross-linking front increases. Second, in the deeper un-cross-linked region below the front, the signal of the NMR profiles was found to decrease proportional to . This could be explained by the presence of slowly reacting species that diffuse into the deeper uncured region of the coating, after which they cross-link. The model describing the effect of these reactive species also indicates that the signal decrease is inversely proportional to coating thickness L, which was confirmed by additional measurements.  相似文献

The mechanical properties of plasma-enhanced magnetron sputtering Si-C-N hard coatings with various compositions are characterized. The effect of chemical composition on the microstructure and properties of coating is investigated. The results show that the microstructure and mechanical properties of Si-C-N coatings are very sensitive to chemical composition. The nanocrystalline/amorphous composite structure is beneficial to the coating's mechanical properties. It also reveals that Si-C-N coating with low Si and high C concentrations has the highest hardness (≥40 GPa) and the best wear property with dry friction coefficient about 0.2.  相似文献
