The cytosolic conversion of therapeutically relevant nucleosides into bioactive triphosphates is often hampered by the inefficiency of the first kinase‐mediated step. Nucleoside monophosphate prodrugs can be used to bypass this limitation. Herein we describe a novel cyclic‐disulfide class of nucleoside monophosphate prodrugs with a cytosol‐specific, reductive release trigger. The key event, a charge‐dissipating reduction‐triggered cyclodeesterification leads to robust cytosolic production of the cyclic 3′,5′‐monophosphate for downstream enzymatic processing. The antiviral competence of the platform was demonstrated with an O‐benzyl‐1,2‐dithiane‐4,5‐diol ester of 2′‐C‐methyluridine‐3′,5′‐phosphate. Both in vitro and in vivo comparison with the clinically efficacious ProTide prodrug of 2′‐deoxy‐2′‐α‐fluoro‐β‐C‐methyluridine is provided. The cytosolic specificity of the release allows for a wide range of potential applications, from tissue‐targeted drug delivery to intracellular imaging.  相似文献

To establish the structure–catalytic property relationships of heterogeneous catalysts, a detailed characterization of the three‐dimensional (3D) distribution of active sites on a single catalyst is essential. Single‐particle catalysis of a modular multilayer catalytic platform that consists of a solid silica core, a mesoporous silica shell, and uniformly distributed Pt nanoparticles sandwiched in between these layers is presented. The first 3D high‐resolution super‐localization imaging of single fluorescent molecules produced at active sites on the core‐shell model nanocatalysts is demonstrated. The 3D mapping is aided by the well‐defined geometry and a correlation study in scanning electron microscopy and total internal reflection fluorescence and scattering microscopy. This approach can be generalized to study other nano‐ and mesoscale structures.  相似文献

A positron emission tomography (PET) tracer composed of 18F‐labeled maltohexaose (MH18F) can image bacteria in vivo with a sensitivity and specificity that are orders of magnitude higher than those of fluorodeoxyglucose (18FDG). MH18F can detect early‐stage infections composed of as few as 105 E. coli colony‐forming units (CFUs), and can identify drug resistance in bacteria in vivo. MH18F has the potential to improve the diagnosis of bacterial infections given its unique combination of high specificity and sensitivity for bacteria.  相似文献

应用毛细管柱气相色谱法测定了食品中甜蜜素。选择弱极性的DB-5型毛细管色谱柱(0.53mm×30m,1.0μm)作为分离柱,采用氢火焰离子化检测器。对固体样品的处理方法改用粉碎机打碎,并按样品的类别分别用水或经石油醚提取去脂后,再用水超声提取,离心分离并过滤后取其滤液供色谱分析。甜蜜素的质量浓度在0.01~1.0g·L-1范围内与峰面积呈线性相关,检出限(3S/N)为1ng。在3个浓度水平进行加标回收试验,回收率在90.9%~99.5%之间,测定值的相对标准偏差(n=5)在2.9%~6.8%之间。按此法测定了30个食品样品中甜蜜素的含量,所得测定值与欧盟标准方法(EN 12857)测定值相符。  相似文献

采用1,2-双(三乙氧基硅基)乙烷(BTESE)和十三氟辛基三乙氧基硅烷(PFOTES)为前驱体,在酸性条件下通过溶胶-凝胶法制备了十三氟辛基修饰的有机-无机杂化SiO2膜材料。利用接触角测量、红外光谱、动态光散射和N2吸附等测试技术分别对膜材料的疏水性、溶胶粒径和孔结构进行表征,并深入研究有支撑膜材料的氢气渗透、分离性能以及长期水热稳定性。结果表明,十三氟辛基修饰后的膜材料由亲水性变成了疏水性,当nPFOTES/nBTESE=0.6时膜材料对水的接触角达到(110.4±0.4)°,膜材料还保持微孔结构,孔径分布在0.5~0.8 nm。氢气在修饰后的膜材料中的输运遵循微孔扩散机理,在300℃时,氢气的渗透率达到8.5×10-7mol·m-2·s-1·Pa-1,H2/CO2,H2/CO和H2/SF6的理想分离系数分别为5.49,5.90和18.36,均高于相应的Knudsen扩散分离因子。在250℃且水蒸气物质的量分数为5%水热环境下陈化250 h,氢气渗透率和H2/CO2的理想分离系数基本保持不变,膜材料具有良好的水热稳定性。  相似文献

Double‐shelled zirconia/titania (ZrO2/TiO2) hollow microspheres were prepared by the selective removal of the polymer components via the calcination of the corresponding tetra‐layer poly(N,N′‐methylenebisacryl amide‐co‐methacrylic acid) (P(MBA‐co‐MAA))/Zr(OH)4/poly(ethyleneglycol dimethacrylate‐co‐methacrylic acid) (P(EGDMA‐co‐MAA))/TiO2 hybrid microspheres. These tetra‐layer microspheres were synthesized by the combination of the distillation copolymerization of N,N(‐methylenebisacryl amide‐co‐methacrylic acid (MBA) or ethyleneglycol dimethacrylate (EGDMA) crosslinker and methacrylic acid (MAA) for the preparation of polymer core and third‐layer as well as the controlled sol‐gel hydrolysis of inorganic precursors for the construction of zirconium hydroxide (Zr(OH)4) and titania (TiO2) layers. The thicknesses of zirconia and titania shell‐layers were conveniently controlled via varying the feed of zirconium n‐butoxide (Zr(OBu)4) and titanium tetrabutoxide (TBOT) during the sol‐gel hydrolysis, while the sizes of polymer layers were tuned through a multi‐stage distillation precipitation copolymerization. The structure and morphology of the resultant microspheres were characterized by transmission electron microscopy (TEM), X‐ray diffractometer (XRD), X‐ray photoelectronic spectroscopy (XPS), and thermogrametric analysis (TGA).  相似文献

We present the design and synthesis of polyoxazole‐based macrocycles containing a phosphonate group. A reliable route was established that allows for convenient and versatile incorporation of various phosphonate functionalities such as phosphonate ester, acid, and salt at the macrocyclic ring periphery. 比较了D-101、D-160、AB-8、NKA-9和聚酰胺等5种吸附树脂对枇杷花总黄酮的吸附及解吸附性能。在静态吸附和动态吸附实验基础上,筛选出效果较好的AB-8树脂进行动态吸附参数的研究。考察了样品液pH值、样品液浓度、洗脱液浓度、上样速度、洗脱速度等对AB-8树脂吸附和解吸效果的影响,确定了AB-8树脂动态吸附枇杷花总黄酮的最佳条件。获得的最佳纯化条件如下,样品液pH值为5.5,样品液浓度为12mg/mL,洗脱液为30%的乙醇水溶液,上样速度为2BV/h,洗脱速度为1BV/h。纯化后样品总黄酮含量达86.7%,比纯化前总黄酮含量高5~6倍。实验结果表明,AB-8树脂可用于分离纯化枇杷花总黄酮。  相似文献

Ring opening reactions of N‐sulfonyl aziridines by primary and secondary amines in silica gel (SG)‐water system were achieved, which provided a mild, practical and environmentally benign method to synthesize mono‐ and bis‐sulfonyl substituted amines. When primary and secondary amines were used in excess, they reacted with N‐sulfonyl aziridines smoothly at room temperature, mainly affording 1:1 ring opening products. Reactions of primary amines with 2 equiv. of aziridines produced 2:1 ring opening products. Some 1:1 products can be cyclized with CS2 to synthesize N‐sulfonyl cyclothioureas also in water.  相似文献

A sensitive fluorescent probe, 2,2'-bisbenzimidazole (L), for CN has been developed. This structurally simple receptor displays great selectivity for the cyanide anion over other common inorganic anions in an aqueous environment. In addition, further study demonstrates the lower detection of the fluorescence response of the sensor to CN is in 10 9 mol/L range. Thus, the present probe should be applicable as a practical system for the monitoring of cyanide concentrations in aqueous samples.  相似文献
