The limitation of current dissociative fluorescence enhancement techniques is that the lanthanide chelate structures used as molecular probes are not stable enough in one-step assays with high concentrations of complexones or metal ions in the reaction mixture since these substances interfere with lanthanide chelate conjugated to the detector molecule. Lanthanide chelates of diethylenetriaminepentaacetic acid (DTPA) are extremely stable, and we used EuDTPA derivatives conjugated to antibodies as tracers in one-step immunoassays containing high concentrations of complexones or metal ions. Enhancement solutions based on different β-diketones were developed and tested for their fluorescence-enhancing capability in immunoassays with EuDTPA-labelled antibodies. Characteristics tested were fluorescence intensity, analytical sensitivity, kinetics of complex formation and signal stability. Formation of fluorescent complexes is fast (5 min) in the presented enhancement solution with EuDTPA probes withstanding strong complexones (ethylenediaminetetra acetate (EDTA) up to 100 mM) or metal ions (up to 200 μM) in the reaction mixture, the signal is intensive, stable for 4 h and the analytical sensitivity with Eu is 40 fmol/L, Tb 130 fmol/L, Sm 2.1 pmol/L and Dy 8.5 pmol/L. With the improved fluorescence enhancement technique, EDTA and citrate plasma samples as well as samples containing relatively high concentrations of metal ions can be analysed using a one-step immunoassay format also at elevated temperatures. It facilitates four-plexing, is based on one chelate structure for detector molecule labelling and is suitable for immunoassays due to the wide dynamic range and the analytical sensitivity.  相似文献

We use the three-dimensional Mercedes-Benz model for water and Monte Carlo simulations to study the structure and thermodynamics of the hydrophobic interaction. Radial distribution functions are used to classify different cases of the interaction, namely, contact configurations, solvent separated configurations, and desolvation configurations. The temperature dependence of these cases is shown to be in qualitative agreement with atomistic models of water. In particular, while the energy for the formation of contact configurations is favored by entropy, its strengthening with increasing temperature is accounted for by enthalpy. This is consistent with our simulated heat capacity. An important feature of the model is that it can be used to account for well-converged thermodynamics quantities, e.g., the heat capacity of transfer. Microscopic mechanisms for the temperature dependence of the hydrophobic interaction are discussed at the molecular level based on the conceptual simplicity of the model.  相似文献

Titanium dioxide (TiO2) is a photoactive material with various interesting and useful properties. One of those is the perfect wettability of TiO2 surface after ultraviolet (UV) illumination. Wettability of a solid surface plays an important role in the field of printing, coating, and adhesion among others. Here we report on a superhydrophobic and photoactive liquid flame spray (LFS) generated TiO2 nanoparticle coating that can be applied on web-like materials such as paper and board in one-step roll-to-roll process. The LFS TiO2 nanoparticle coated paper and board were superhydrophobic instantly after the coating procedure because of spontaneously accumulated carbonaceous overlayer on TiO2, and thus there was no need for any type of separate hydrophobization treatment. The highly photoactive LFS TiO2 nanoparticle coating could be converted steplessly from superhydrophobic to superhydrophilic by UV-illumination, and the coating gave strong response to natural daylight illumination even in the shade. The superhydrophobic LFS TiO2 coated surface can be used as an intelligent substrate, where photo-generated hydrophilic patterns guide the fluid setting and figure formation. Our study reveals that the wettability changes on the LFS TiO2 surface were primarily caused by the photocatalytic removal of the carbonaceous material from TiO2 during the UV-illumination and spontaneous accumulation of the carbonaceous material on the surface of the metal oxide during storage in the dark. The latter mechanism was found to be a temperature activated process which could be significantly speeded up by heat treatment. If other mechanisms such as surface oxidization, increment of hydroxyl groups, or charge separation played a role in the wetting phenomena on TiO2, their effect was rather secondary as the removal and accumulation of the carbonaceous material dominated the wettability changes on the surface. Our study gives valuable information on the complex issue of photo-induced wettability changes on TiO2.  相似文献

To provide a solid chemical basis for the mechanistic interpretations of the thio effects observed for large ribozymes, the cleavage of triribonucleoside 3',3',5'-phosphoromonothioate triesters and diribonucleoside 3',3'-phosphorodithioate diesters has been studied. To elucidate the role of the neighboring hydroxy group of the departing 3'-linked nucleoside, hydrolysis of 2',3'-O-methyleneadenosin-5'-yl bis[5'-O-methyluridin-3'-yl] phosphoromonothioate (1 a) has been compared to the hydrolysis of 2',3'-O-methyleneadenosin-5'-yl 5'-O-methyluridin-3'-yl 2',5'-di-O-methyluridin-3'-yl phosphoromonothioate (1 b) and the hydrolysis of bis[uridin-3'-yl] phosphorodithioate (2 a) to the hydrolysis of uridin-3'-yl 2',5'-di-O-methyluridin-3'-yl phosphorodithioate (2 b). The reactions have been followed by RP HPLC over a wide pH range. The phosphoromonothioate triesters 1 a,b undergo two competing reactions: the starting material is cleaved to a mixture of 3',3'- and 3',5'-diesters, and isomerized to 2',3',5'- and 2',2',5'-triesters. With phosphorodithioate diesters 2 a,b, hydroxide-ion-catalyzed cleavage of the P--O3' bond is the only reaction detected at pH >6, but under more acidic conditions desulfurization starts to compete with the cleavage. The 3',3'-diesters do not undergo isomerization. The hydroxide-ion-catalyzed cleavage reaction with both 1 a and 2 a is 27 times as fast as that compared with their 2'-O-methylated counterparts 1 b and 2 b. The hydroxide-ion-catalyzed isomerization of the 3',3',5'-triester to 2',3',5'- and 2',2',5'-triesters with 1 a is 11 times as fast as that compared with 1 b. These accelerations have been accounted for by stabilization of the anionic phosphorane intermediate by hydrogen bonding with the 2'-hydroxy function. Thio substitution of the nonbridging oxygens has an almost negligible influence on the cleavage of 3',3'-diesters 2 a,b, but the hydrolysis of phosphoromonothioate triesters 1 a,b exhibits a sizable thio effect, k(PO)/k(PS)=19. The effects of metal ions on the rate of the cleavage of diesters and triesters have been studied and discussed in terms of the suggested hydrogen-bond stabilization of the thiophosphorane intermediates derived from 1 a and 2 a.  相似文献

The performance of quantum chemical methods for geometry prediction of hafnocenes was evaluated. HF, B3LYP and MP2 in combination with nonrelativistic (MHF) and relativistic (MWB and LANL2DZ) basis sets for hafnium together with standard basis sets 3-21G*, 6-31G* and 6-311G** for other elements were applied. Five basic structural parameters of the optimized structures of the hafnocenes were compared with experimental crystal structures obtained from the Cambridge structural database. Altogether 80 hafnocenes were included in the analysis. The results show that relativistic corrections are necessary for Hf atom. However, even the Hartree–Fock (HF) method, when combined with relativistic pseudopotentials, reproduces the experimental crystal structures with significant accuracy. The good performance of the HF method can be understood to originate from the absence of significant near-degeneracy correlations for hafnium. On average, the B3LYP and MP2 methods provide structural parameters somewhat closer to the experimental ones.  相似文献

Preparation and full characterization of the main-group diradical *NSNSC-CNSSN*, 8, the MF6- salt (As, Sb) of radical cation +NSNSC-CNSSN*, 8*+, and the AsF6- salt of the dication +NSNSC-CNSSN+, 82+, are presented. 8, a=6.717 (4), b=11.701(2), c=8.269(3) A, alpha=gamma=90, beta=106.69(3) degrees, monoclinic, space group P21/n, Z=4, T=203 K; 8SbF6, a=6.523(2), b=7.780(2), c=12.012(4) A, alpha=91.994(4), beta=96.716(4), gamma=09.177(4) degrees, triclinic, space group P, Z=2, T=198 K; 8[AsF6]2, a=12.7919(14), b=9.5760(11), c=18.532(2) A, alpha=gamma=90, beta=104.034(2) degrees, monoclinic, space group Pn, Z=6, T=198 K. Preparation of 8MF6 was carried out via a reduction of [CNSNS]2[MF6]2 (M=As, Sb) with either ferrocene or a SbPh3-NBu4Cl mixture. In the solid state, diamagnetic 8SbF6 contains centrosymmetric dimers [8*+]2 linked via two-electron four-centered pi*-pi* interactions with a thermally excited triplet state as detected by electron paramagnetic resonance (EPR). This is the first observation of a triplet excited state for a 7pi 1,2,3,5-dithiadiazolyl radical dimer. The singlet-triplet gap of the [-CNSSN*]2 radical pair was -1800+/-100 cm(-1) (-22+/-1 kJ/mol) with the ZFS components |D|=0.0267(6) cm(-1) and |E|=0.0012(1) cm(-1), corresponding to an in situ dimerization energy of ca. -11 kJ/mol. Cyclic voltammetry measurements of 8[AsF6]2 showed two reversible waves associated with a stepwise reduction of the two isomeric rings [E1/2 (+2/+1)=1.03 V; E1/2 (+1/0)=0.47 V, respectively]. 8MF6 (M=As, Sb) was further reduced to afford the mixed main-group diradical 8, containing two isomeric radical rings. In solution, 8 is thermodynamically unstable with respect to *NSSNC-CNSSN*, but is isolable in the solid state because of its low solubility in SO2. Likewise, 8SbF6, 8 is dimeric, with pi*-pi* interactions between different isomeric rings, and consequently diamagnetic; however, a slight increase in paramagnetism was observed upon grinding [from C=6.5(3)x10(-4) emu.K/mol and temperature-independent paramagnetism (TIP)=1.3(1)x10(-4) emu/mol to C=3.2(1)x10(-3) emu.K/mol and TIP=9.0(1)x10(-4) emu/mol], accompanied by an increase in the lattice-defect S=1/2 sites [from 0.087(1) to 0.43(1)%]. Computational analysis using the multiconfigurational approach [CASSCF(6,6)/6-31G*] indicated that the two-electron multicentered pi*-pi* bonds in [8*+]2 and [8]2 have substantial diradical characters, implying that their ground states are diradicaloid in nature. Our results suggest that the electronic structure of organic-radical ion pairs, for example, [TTF*+]2, [TCNE*-]2, [TCNQ*-]2, [DDQ*-]2, and related pi dimers, can be described in a similar way.  相似文献

Nanotubes with diameters in the magnitude of 10 nm can be treated by quantum chemical methods, made possible by utilization of line group symmetries. The methodology presented here is applied for magnesium dichloride nanotubes. The structural preferences of MgCl2 nanotubes are strongly dependent on the tube diameter. The rolled layer of a crystalline sheet becomes favorable above a diameter of 3.4 nm, below which alternative stable coordination modes dominate. Application of the methodology for other nanotubular compounds, including multiwalled nanotubes, is expected to be straightforward.  相似文献
