A simple method for the simultaneous, rapid and sensitive determination of N-acylhomoserine lactone signaling molecules in bacterial isolates, without prior sample preconcentration and with minimal sample cleanup, is presented. The analysis relies on the combination of analyte preconcentration and separation on a single device: a relatively large sample volume (1–5 L) is directly loaded onto a laboratory-made, miniaturized (75 m i. d.) reverse phase nano-liquid chromatography column, connected on-line to a microelectrospray-ionization ion trap mass spectrometer. In a first step the analyte is adsorbed (and so concentrated) at the beginning of the column, and is eluted and selectively separated in a second step by the organic mobile phase. Sample preconcentration follows the mechanisms of solid phase extraction on a nano-scale, while separation takes place according to classical liquid chromatography separation principles. The columns can be manufactured easily, are simply connected, and used with minimal solvent amounts; this makes this method extremely robust and cost-effective. The analytical setup was found to be routinely quantitative down to a concentration of 10 ng/mL (corresponding to a total analyte amount of 10 pg or ca. 50 fmol). The limit of detection was reached at 1 ng/mL (1 pg, ca. 5 fmol). Compared to the classical AHL analysis of bacterial cultures with biosensors, where selectivity and sensitivity is often limited, this rapid analytical technique is a substantial qualitative and quantitative improvement. Two unsubstituted N-acylhomoserine lactones could be identified and quantified from a Burkholderia cepacia culture supernatant in a chloroform extract. 相似文献

Annealed submonolayer CoAg/Ru(0001) films form an alloy with a structure that contains droplets of Ag surrounded by Co [G. E. Thayer, V. Ozolins, A. K. Schmid, N. C. Bartelt, M. Asta, J. J. Hoyt, S. Chiang, and R. Q. Hwang, Phys. Rev. Lett. 86, 660 (2001)]. To understand how surface stress contributes to the formation of this structure, we use scanning tunneling microscopy to extract atomic displacements at the boundaries between regions of Co and Ag. Comparing our measurements to Frenkel-Kontorova model calculations, we show how stress due to lattice mismatch contributes to the formation of the alloy droplet structure. In particular, we quantitatively evaluate how competing strain and chemical energy contributions determine surface structure. 相似文献

The oxidation of Pd(111) leads to an incommensurate surface oxide, which was studied by the use of scanning tunneling microscopy, surface x-ray diffraction, high resolution core level spectroscopy, and density functional calculations. A combination of these methods reveals a two-dimensional structure having no resemblance to bulk oxides of Pd. Our study also demonstrates how the atomic arrangement of a nontrivial incommensurate surface can be solved by molecular dynamics in a case where experimental techniques alone give no solution. 相似文献

The microbial flavoenzyme EpiD catalyzes the oxidative decarboxylation of peptidyl-cysteines to peptidyl-aminoenethiols. These unusual C-terminally modified peptides are intermediates in the biosynthesis of the tetracyclic peptide antibiotic epidermin, which belongs to the lantibiotics family. The peptide SFNSYCC represents the C-terminal partial sequence of the natural precursor peptide EpiA. EpiA is posttranslationally modified to form finally the lantibiotic epidermin. The substrate specificity of EpiD was investigated using high-resolution mass spectrometry and the heptapeptide library SFNSXCC. The enzymatic conversion of particular peptides can be observed by a mass loss of m/z 46. In contrast to the previously used triple quadrupole instrument, electrospray ionization Fourier transform ion cyclotron resonance mass spectrometry (ESI-FTICR-MS) was able to resolve and detect all precursor and converted peptides with identical nominal masses in a single measurement, avoiding the necessity to investigate single peptides. The structure of nanowires of different metals grown within nanoporous alumina membranes has been studied by EXAFS, WAXS and
high energy X-ray diffraction. Nanowires of gold, silver, copper and iron adopt the lattice structure and bond distances of
the bulk metals. Cobalt nanowires on the other hand were composed of a mixture of hcp phase, stable at room temperature, and
fcc phase, which in bulk cobalt is normally stable only at high temperatures, in a ratio depending on the pore size. The nanowires
are non-continuous but are made of nanocrystallites whose shape and size was found to depend strongly on the metal. All the
metals except gold showed the presence of a preferred orientation which was slight in the case of Ag and Cu but much stronger
in the case of iron and cobalt nanowires.

Variable displacement vane pumps are used in a variety of applications, most prominently in the context of modern automotive vehicles. Minimal models capturing the main dynamic effects of such pumps have remained scarce. In this contribution, a minimal model taking into account the force effects from line pressure exposure and dead volume compression and expansion is presented. In this article, we extend mean 3D projective shape change in matched pairs to independent samples. We provide a brief introduction of projective shapes of spatial configurations obtained from their digital camera images, building on previous results of Crane and Patrangenaru (J Multivar Anal 102:225–237, 2011). The manifold of projective shapes of k-ads in 3D containing a projective frame at five given landmark indices has a natural Lie group structure, which is inherited from the quaternion multiplication. Here, given the small sample size, one estimates the mean 3D projective shape change in two populations, based on independent random samples of possibly different sizes using Efron’s nonparametric bootstrap. This methodology is applied in three relevant applications of analysis of 3D scenes from digital images: visual quality control, face recognition, and scene recognition. 相似文献
