The DNA fragment detection focusing technique has further enhanced the sensitivity and information of DNA targets. The DNA fragment detection method was established by capillary electrophoresis with laser‐induced fluorescence detection and restriction endonuclease chromatographic fingerprinting (CE‐LIF‐REF) in our experiment. The silica capillary column was coated with short linear polyarclarylamide (SLPA) using nongel sieving technology. The excision product of various restricted enzymes of DNA fragments was obtained by REF with the molecular biology software Primer Premier 5. The PBR322/BsuRI DNA marker was used to establish the optimization method. The markers were focused electrophoretically and detected by CE‐LIF. The results demonstrate that the CE‐LIF‐REF with SLPA can improve separation, sensitivity and speed of analysis. This technique may be applied to analysis of the excision product of various restricted enzymes of prokaryotic plasmid (pIRES2), eukaryote plasmid (pcDNA3.1) and the PCR product of codon 248 region of gastric cancer tissue. The results suggest that this method could very sensitively separate the excision products of various restricted enzymes at a much better resolution than the traditional agarose electrophoresis. The paper presents a new form of the electrothermal model of the ferromagnetic core for SPICE and the way, in which the parameters of the model are determined. The manner of determining magnetic, geometric and thermal parameters of this model is described. The correctness of the proposed model and the manner of determining the values of the parameters are verified by comparing the calculated and measured characteristics of the selected ferromagnetic cores.  相似文献

A new method using a column packed with graphene as adsorbent was developed for the preconcentration of trace amounts of cobalt (Co) and nickel (Ni) prior to their determinations by flame atomic absorption spectrometry. Several factors influencing the extraction efficiency of Co and Ni and their subsequent determinations, such as pH, amounts of the chelating agent, flow rates of sample and eluent solution, eluent type and its volume, breakthrough volume, and adsorption capacity were established. Under the optimum conditions, the calibration graphs were linear in the range of 4.0‐200.0 μg L?1 and 5.0‐200.0 μg L?1 with detection limits of 0.36 μg L?1 and 0.51 μg L?1 for Co and Ni, respectively. Good relative standard deviations for ten determinations of 100.0 μg L?1 of Co and Ni were 3.2 and 3.6%, respectively. The results for determination of Co and Ni in tap water, river water, sea water, vegetable and spiked samples have demonstrated the accuracy and applicability of the proposed method. To validate the proposed method, three certified reference materials of environment water (GSBZ 50030‐94 and GSB 07‐1186‐2000) and tomato leaf (GSBZ 51001‐94) were analyzed, and the determined values were in good agreement with the certified values.  相似文献

In 2006, P. J. Cameron and J. Ne?et?ril introduced the following variant of homogeneity: we say that a structure is homomorphism-homogeneous if every homomorphism between finite substructures of the structure extends to an endomorphism of the structure. In this paper we classify finite homomorphism-homogeneous point-line geometries up to a certain point. We classify all disconnected point-line geometries, and all connected point-line geometries that contain a pair of intersecting proper lines (we say that a line is proper if it contains at least three points). In a way, this is the best one can hope for, since a recent result by Rusinov and Schweitzer implies that there is no polynomially computable characterization of finite connected homomorphism-homogeneous point-line geometries that do not contain a pair of intersecting proper lines (unless P=coNP).  相似文献

A palladium-based catalyst (Fe3O4/SiO2/HPG–OPPh2–PNP) supported on chlorodiphenylphosphine-functionalized magnetic nanoparticles was successfully prepared from Fe3O4/SiO2 with sequential attachment of glycerol and chlorodiphenylphosphine, followed by treatment of an ethanolic solution of palladium chloride with hydrazine. The as-prepared catalyst was characterized by ICP-AES, FTIR, XRD, SEM, and TEM. The Fe3O4/SiO2/HPG–OPPh2–PNP was found as a magnetically separable and highly active catalyst for Suzuki coupling reactions of aryl iodides, bromides, and chlorides as well as Heck reactions of aryl iodides and bromides. Under appropriate conditions, all reactions afforded the desired products in moderate to excellent yields. Moreover, this catalyst can be easily recovered by using a magnetic field and directly reused for at least six cycles without significant loss of its activity.  相似文献
