A two-step, general synthesis of 1,4-benzodiazepine-2,5-diones (BZDs) is presented. This synthesis employs an Ugi four-component condensation using a convertible isocyanide (1-isocyanocyclohexene), followed by an acid-activated cyclization reaction. This synthesis represents a dramatically improved route to BZDs over those currently in the literature. In addition, since amino acids are not used as inputs, the potential for molecular diversity is much greater than that with existing syntheses. It was also found that BZDs substituted with methylenes at the C-3 and N-4 positions display conformational isomerism in the NMR spectra at room temperature. Variable-temperature NMR experiments support this observation and offer the interesting conclusion that the BZD core structure, in certain examples, might not be as rigid as previously supposed.  相似文献

The synthesis of 2-azathianthrene ([1,4]benzodithiino[2,3-c]pyridine), the only remaining monoazathianthrene yet to be reported, is described. Attempts at the direct condensation of disubstituted pyridines with the dianion of 1,2-dimercaptobenzene were generally unsuccessful requiring that the alternative condensation of the dianion with disubstituted pyridine 1-oxides be employed. The title compound was characterized by physical means including 13C-nmr spectroscopy. One analog, 4-nitro-2-azathianthrene was also studied by X-ray crystallographic means; the molecule crystallized with two molecules in the asymmetric unit P21/n, a = 20.712(3), b = 7.8109(13), c = 13.720(2)Å, β = 107.880(11)°, Z = 8, the data refined to a final R = 0.051 for 3061 reflections. Dihedral angles between the planes of the phenyl rings were 135.00(13) and 132.52(13)° for the two independent molecules contained in the crystal. Close non? bonded S ?O intramolecular contacts were observed in both molecules between the sulfur and nitro-group oxygens. Both nitro groups are twisted out of the plane of the pyridine ring and are oriented at angles of 28.75 and 38.82° respectively.  相似文献

A synthesis of a 1,3-dithiane corresponding to the C1-side chain of zaragozic acid D is described. An aldol reaction using an Evans oxazolidinone is the key step in controlling stereochemistry. Metallation of the derived dithiane monosulfoxide and coupling to an aldehyde effected construction of the C1-C7 bond. Subsequent steps are also reported, including acid-mediated ketalization resulting in formation of an advanced synthetic intermediate containing the bicyclic ketal core of the natural product.  相似文献

Chiral stationary phases (CSPs) based on polymeric (R,R)- or (S,S)-1,2-diaminocyclohexane (DACH) derivatives are synthesized. When bonded to 5 microm porous spherical silica gel, the poly (trans-1,2-cyclohexanediyl-bis acrylamide) based poly-cyclic amine polymer (P-CAP) stationary phases is proved to be effective chiral stationary phases that could be used in the normal-phase mode, polar organic mode and with halogenated solvents mobile phases, if desired. Since these are entirely synthetic CSPs, the elution order of all enantiomers can be reversed between the (R,R) P-CAP and (S,S) P-CAP columns. Because of the high loading of chiral selectors, the columns exhibit very high sample capacities. Thus, P-CAP columns are useful for preparative and semi-preparative enantiomeric separations. The application of these CSPs and optimization of their separations are discussed.  相似文献

A spectrum of oxidative lesions was observed in a bacteriophage-based model system that is very sensitive to the photodynamic activity of selected dyes. When suspensions of the intact bacteriophage Qβ were exposed to methylene blue plus light (MB+L), inactivating events, or "hits" occurred that were oxygen-dependent and that were associated with the formation of several specific lesions: (1) carbonyl moieties on proteins, (2) 8-oxo-7,8-dihydroguanine (8-oxoGua), and (3) single-strand breaks (ssb) in the RNA genome and (4) RNA-protein crosslinks. Formation of carbonyl groups associated with protein in the Qβ phage preparation correlated positively with photoinactivation of the phage with increasing doses of either of the sensitizers MB or rose bengal. Strand breaks in the Qβ genomic RNA were observable at high MB concentrations but appeared not to be significant at the lower concentrations of MB, as full-length Qβ RNA was observable well beyond the 99% inactivation point in MB dosage. It was shown that the number of 8-oxoGua lesions were unlikely to be sufficient to account for the number of lethal events. Following exposure to MB+L, crosslink formation between Qβ RNA and protein was observed by virtue of the location of RNA at the interface of phenol-aqueous extractions of phage suspensions. A significant increase over background of RNA-protein complexes (including full-length Qβ RNA) was observed at the lowest concentration of MB tested (0.5 μ M ), which corresponded roughly to an average of 2 lethal hits per phage or approximately 13% survival compared to the zero MB control (100% survival). Due to its close correlation with Qβ inactivation and its expected lethality, RNA-protein crosslink formation may be important as an inactivating lesion in bacteriophage Qβ following MB+L exposure.  相似文献

We show that the blue native gel polyacrylamide electrophoresis system (BN-PAGE) can be applied to pyruvate dehydrogenase complex (PDC). BN-PAGE has been used extensively to study the multisubunit enzymes of oxidative phosphorylation, as nondenaturing separation in the first dimension maintains holoenzyme integrity. However, the standard protocol was inappropriate for PDC as, at 10 MDa, it is approximately ten times larger than the largest respiratory chain enzyme complex. Therefore, agarose was substituted for polyacrylamide. Moreover, a substantial decrease in salt concentration was necessary to prevent dissociation of PDC. As with standard BN-PAGE, immunoblots of second-dimensional sodium dodecyl sulfate-PAGE (SDS-PAGE) provided more detailed information on specific subunits and subcomplexes. The method was applied to human heart mitochondrial fragments, control cultured human cells, rho0 cells that lack mitochondrial DNA, and two cell lines derived from patients with PDC deficiency. The PDC deficient cell lines showed a clear correlation between amount of PDC holoenzyme and disease severity. In cells lacking mitochondrial DNA, synthesis and assembly of all PDC subunits (all nuclearly encoded) appeared normal, suggesting that respiratory function has no regulatory role in PDC biogenesis. Blue native agarose gel electrophoresis coupled with standard second-dimensional SDS-PAGE provides a new tool to be used in conjunction with biochemical assays and immunoblots of one-dimensional SDS-PAGE to further elucidate the nature of PDC in normal and disease states. Furthermore, other cellular protein complexes of 1 MDa or more can be analysed by this method.  相似文献

Abstract— Ultraviolet B radiation (280-320 nm) can systemically suppress contact hypersensitivity (CHS), delayed type hypersensitivity (DTH) and tumor rejection responses in mice. Several models have been postulated for the initiation of this UVB-induced immune suppression and, although the complete mechanism is unclear, our early studies suggested that initiation is via the activation of a photoreceptor in the skin, identified as urocanic acid (UCA). Recent preliminary data from our laboratory and others indicated that UVA (320-400 nm)-emitting broadband sunlamps can also isomerize UCA but may not lead to immune suppression, in contrast to UVB-emitting sunlamps, which cause both effects. Although the reason for this inconsistency is unknown, the emission spectra of UVA lamps contain differing amounts of UVB, UVA-I (340-400 nm) and UVA-II (320-340 nm) from those of UVB sources. In this study we determined a detailed dose-response for the isomerization of UCA in mouse skin using the UVA-I, UVA-II and UVA-I+II wavelength ranges. The dose-response curves obtained were put on an equal energy basis by quantum correction and the possibility of wavelength interaction for this effect investigated. A simple additive wavelength interaction between UVA-I, UVA-II, and UVA-I+II was observed for trans-UCA photoisomerization. This result indicates that the failure of UVA-I, UVA-II or UVA-I+II radiation to induce immune suppression of the CHS response in an animal model is not due to complex wavelength interactions and/or the presence of an in vivo endogenous photosensitizer of UCA isomerization. Other factors, such as downstream blocking by UVA of the cis -UCA generated signal, may be involved.  相似文献

The reaction of ((t)BuNH)(3)PNSiMe(3) (1) with 1 equiv of (n)BuLi results in the formation of Li[P(NH(t)Bu)(2)(N(t)Bu)(NSiMe(3))] (2); treatment of 2 with a second equivalent of (n)BuLi produces the dilithium salt Li(2)[P(NH(t)Bu)(N(t)Bu)(2)(NSiMe(3))] (3). Similarly, the reaction of 1 and (n)BuLi in a 1:3 stoichiometry produces the trilithiated species Li(3)[P(N(t)Bu)(3)(NSiMe(3))] (4). These three complexes represent imido analogues of dihydrogen phosphate [H(2)PO(4)](-), hydrogen phosphate [HPO(4)](2)(-), and orthophosphate [PO(4)](3)(-), respectively. Reaction of 4 with alkali metal alkoxides MOR (M = Li, R = SiMe(3); M = K, R = (t)Bu) generates the imido-alkoxy complexes [Li(3)[P(N(t)Bu)(3)(NSiMe(3))](MOR)(3)] (8, M = Li; 9, M = K). These compounds were characterized by multinuclear ((1)H, (7)Li, (13)C, and (31)P) NMR spectroscopy and, in the cases of 2, 8, and 9.3THF, by X-ray crystallography. In the solid state, 2 exists as a dimer with Li-N contacts serving to link the two Li[P(NH(t)Bu)(2)(N(t)Bu)(NSiMe(3))] units. The monomeric compounds 8 and 9.3THF consist of a rare M(3)O(3) ring coordinated to the (LiN)(3) unit of 4. The unexpected formation of the stable radical [(Me(3)SiN)P(mu(3)-N(t)Bu)(3)[mu(3)-Li(THF)](3)(O(t)Bu)] (10) is also reported. X-ray crystallography indicated that 10 has a distorted cubic structure consisting of the radical dianion [P(N(t)Bu)(3)(NSiMe(3))](.2)(-), two lithium cations, and a molecule of LiO(t)Bu in the solid state. In dilute THF solution, the cube is disrupted to give the radical monoanion [(Me(3)SiN)((t)BuN)P(mu-N(t)Bu)(2)Li(THF)(2)](.-), which was identified by EPR spectroscopy.  相似文献

A three-dimensional finite element method (FEM) for the analysis of plane wave diffraction by a bi-periodic slab is described and implemented. A scattering matrix formalism based on the FEM allows the efficient treatment of light reflection and transmission by multilayer bi-periodic structures, and the computation of Bloch modes of three-dimensional arrays. Numerical simulations, which show the accuracy and flexibility of the FEM, are presented.  相似文献
