桑蚕丝素蛋白初始结构对其矿化作用的影响

以碱金属离子诱导桑蚕丝素蛋白溶液发生构象转变, 研究了蛋白质初始结构对其矿化作用的影响. FT-IR, XRD和SEM等测试结果显示, 未经任何处理的桑蚕丝素蛋白溶液矿化后形成片状复合物, 其无机相以二水磷酸氢钙(DCPD)为主; 而经过K^＋和Na^＋金属离子处理后, 桑蚕丝素溶液的结构由无规线团/螺旋构象向β-折叠发生转变, 矿化后成纤维状, 并相互结合呈现纳米级的三维多孔结构, 其无机相以热力学稳定的羟基磷灰石(HA)为主. 可以认为, 丝素蛋白结构转化为较伸展的β-折叠后, 使得更多的亲水基团暴露在外面, 在丝素蛋白分子不断凝聚成纤过程中, HA结晶快速生长并附着在这些微纤上, 最终形成纤维状的丝素蛋白/HA复合物. 该结果为阐明蛋白质的生物矿化过程及其调控机理提供了理论依据, 同时可以从矿化复合物的形成来反映这些微量元素可能对骨组织形成的影响, 为临床骨组织的修复提供一定的参考.     

Zeb1 for RCP-induced oral cancer cell invasion and its suppression by resveratrol

Rab coupling protein (RCP) is upregulated in head and neck squamous cell carcinoma (HNSCC) and is correlated with the progression and survival of patients. However, the role of RCP in one of the aggressive types of HNSCC, oral squamous cell carcinoma (OSCC), remains elusive. In the present study, we identified the important role of Zeb1 in RCP-induced OSCC epithelial-to-mesenchymal transition (EMT) and invasion. RCP induces Zeb1 expression, and silencing Zeb1 expression significantly inhibits RCP-induced OSCC invasion. In addition, Zeb1 upregulates MT1-MMP expression to promote OSCC EMT and invasion. Furthermore, we observed that the β1 integrin/EGFR/β-catenin signaling cascade mediates RCP-induced Zeb1 expression to promote OSCC invasion. Notably, we provide evidence that resveratrol (REV) strongly inhibits RCP-induced Zeb1 expression through blocking β1 integrin endosome recycling and EGFR activation, leading to suppression of RCP-induced OSCC invasion, demonstrating the important role of RCP in OSCC invasion and its reversion by REV. Collectively, the present study provides evidence for the first time that RCP aggravates OSCC invasion through increasing Zeb1 expression and subsequently upregulating MT1-MMP expression and that this process is reversed by REV, providing novel biomarkers and indicating the therapeutic potential of REV in OSCC.Subject terms: Oral cancer, Cell invasion     

蒂巴茵衍生物的合成研究

The reaction of 7α-acetyl-6,14-endoethano-6,7,8,14-tetrahydrothebaine with 2-（thien-2-yl）ethylmagnesium bromide was investigated. The tertiary alcohol derivative 7α-[R-l-hydroxyl-l-methyl-3-（thien-2-yl）propyl]）-6,14- endoethano-6,7,8,14-tetrahydrothebaine （3） and a by-product 4 were isolated. The structure of 4 was elucidated by X-ray analysis. The Grignard reaction shows high degree of stereoselectivity according with Cram rule. The crystal structure of 4 indicates that dihydrofuran ring was opened to form a phenolic hydroxyl group and a three-membered ring structure. It maintains the main rigid structure of morphine and contains a C（6）-C（14） enthano bridge. The 1-hydroxyl-1-methyl-3-（thien-2-yl）propyl group at C（7） position adopted S-configuration.     

模糊正交法用于石墨炉探针原子化测定铋条件的研究

模糊正交法是通过把正交试验结果模糊化,然后用模糊数学的理论和方法来处理实验数据。与常规正交设计相比,能在同样实验工作量的情况下获得更多的信息。本文采用模糊正交法对元素Bi的测定条件进行优化,并将其同常规正交设计方差分析的结果进行了比较,并提出一个综合评价参数,对建立的分析方法进行了综合评价。     

Methodological modifications on quantification of phosphatidylethanol in blood from humans abusing alcohol,using high-performance liquid chromatography and evaporative light scattering detection

Background  

Phosphatidylethanol (PEth) is an abnormal phospholipid formed slowly in cell membranes by a transphosphatidylation reaction from phosphatidylcholine in the presence of ethanol and catalyzed by the enzyme phospholipase D. PEth in blood is a promising new marker of ethanol abuse depending on the high specificity and sensitivity of this marker. None of the biological markers used in clinical routine at the present time are sensitive and specific enough for the diagnosis of alcohol abuse.     

Direct separation of the enantiomers of cetirizine and related compounds by reversed-phase chiral HPLC

Summary Direct separations of the enantiomers of cetirizine and related compounds have been achieved by reversed-phase HPLC on the Chiralcel OD-R, a polysaccharide-derived chiral stationary phase; the mobile phase was usually perchlorate solution supplemented with acetonitrile. Resolution of the enantiomers of cetirizine and related compounds was good. The effect of the acetonitrile content of the mobile phase was investigated, and the effect of the structure of the chiral compounds on their behavior on the Chiralcel OD-R column is discussed.     

互通多孔碳/二氧化锰纳米复合材料的原位水热合成及电化学性能

以互通多孔碳（IPC）为载体，水热条件下在碳表面原位反应生成纳米结构的二氧化锰（MnO₂），制备互通多孔碳/二氧化锰纳米（IPC/MnO₂）复合电极材料. 采用扫描电镜（SEM），透射电镜（TEM），X射线衍射（XRD），热重分析（TGA）对其结构进行表征；采用循环伏安法、恒流充放电和交流阻抗对其电化学性能进行研究. 结果表明：生成的MnO₂均匀地负载在碳的表面，形成多层次结构，并且随着温度的升高IPC表面负载的MnO₂由纳米颗粒变为纳米片状结构；MnO₂纳米片具有典型的K-Birnessite 型晶体结构；复合物中MnO₂的含量约为34%（w）. 在100 ℃制备的IPC/MnO₂复合材料在三电极系统中最高比电容达到了411 F·g^-1；随着反应温度的升高,比容量先增长后基本保持不变. 以IPC/MnO₂为正极，活性炭（AC）为负极，1 mol·L^-1 Na₂SO₄溶液为电解液组装成IPC/MnO₂//AC 混合超级电容器，发现IPC/MnO₂电极的电容器其电位窗口从1 V扩展到1.8 V，容量可达86F·g^-1，且表现出良好的电容特性和大电流放电性能.     

Hydrothermal Synthesis and Crystal Structure of a Cadmium（Ⅱ） Polymer with One-dimensional Chain Structure： [Cd（MDPPz）（BDC）（H2O）]n

A metal-organic coordination compound formulated as [Cd(MDPPz)(BDC)(H2O)]n 1 (MDPPz=11-methyldipyrido[3,2-a:2',3'-c]phenazine,BDC=1,4-benzenedicarboxylate) has been hydrothermally synthesized and structurally characterized by elemental analysis,IR spectrum,fluorescence spectrum and single-crystal X-ray diffraction. The title compound crystallizes in the monoclinic system,space group C2/c,with a=30.673(8),b=9.623(3),c=20.784(6) ,β=98.646(3)o,V=6065(3)3,C27H17CdN4O5,Mr=589.85,Dc=1.292 g/cm3,μ(MoKα)=0.757 mm-1,F(000)=2360,Z=8,the final R=0.0381 and wR=0.0855 for 4191 observed reflections (I>2σ(I)). 1 exhibits blue fluorescence property at room temperature.     

Synthesis of Lanthanide Coordination Polymers with Benzophenone-4,4′-dicarboxylate： Effect of Lanthanide Contraction on Structures

Four lanthanide coordination polymers with benzophenone‐4,4′‐dicarboxylic acid (H₂bpndc) and 1,10‐phenanthroline (phen), [Ln₂(bpndc)₃(phen)] (Ln=La (1), Pr (2) and Tb (3)), [Yb(bpndc)₁₅(phen)].05H₂O (4) were obtained through solvothermal synthesis. The crystallographic data show that 1, 2, and 3 are isostructural, the Ln(III) ions in 1, 2 and 3 are all eight‐ and ten‐coordinated, respectively, and thus the Ln(III) ions are connected by bpndc ligands, resulting in an interpenetrating 3D structure. While in 4, the Yb(III) ions are eight‐coordinated and connected by bpndc ligands into a 3D structure with 1D rhombic channels, which result from the effect of lanthanide contraction from La(III) to Yb(III) ions, and the bpndc ligands in 1, 2, 3, and 4 display three types of coordination modes.
