Treatment of heterocyclic β‐ketonitriles 1a,b with hydrazine hydrate and phenylhydrazine afforded the hydrazine derivatives 2a‐d which cyclized in PPA into pyrrolo[3,4‐c]pyrazoles 3a‐d. Reaction of 1a,b with cyanoacetohydrazide furnished the cyanoacetyl pyrrolo[3,4‐c]pyrazoles 4a,b. The hydrazine 2c reacted with β‐diketone and β‐ketoesters to afford pyrazolyl‐pyrrolines 5‐7. Also the later hydrazine reacted with some D‐aldoses and aceteophenone to give the corresponding hydrazones 10‐12 and hydrazine carboxamide derivatives 15a,b respectively.  相似文献

The 13C-NMR spectra of the synthetic membrane modifying nonadecapeptide Boc-(Aib-l-Ala)5-Gly-Ala-Aib-Pro-Ala- Aib-Aib-Glu(OBz)-Gln-OMe (Aib = α-aminoisobutyric acid), and of synthetic intermediates were used for conformational analysis in solution. The assignments of the 13C-NMR signals of Aib are based on the magnetic nonequivalence (MNE) of the geminal Cβ-signals in asymmetric environment resulting in a shift difference of 0.2–0.5 ppm due to neighbouring chiral residues. More than 4 ppm MNE are observed due to α-helical conformation and about 2.5 ppm for Aib situated in the corners of a rigid β-turn. The Ala-Cα signal is also sensitive to different secondary structures. The Cα signal for C-terminal alanine is found at 49–50 ppm, and for alanine within unordered oligopeptides it absorbs at 50–51 ppm. α-Helical environment shifts the Ala-Cα signal to lower field down to 54 ppm. In methanolic solution the nonadecapeptide shows a α-helical N-terminal region. For the C-terminus beginning with proline-14 no periodically ordered conformation is observed, and we suggest a sequence of β-turns. Furthermore the typical E/Z isomerism of the prolyl-peptide bond can be observed on proline itself and on its neighbour alanine.  相似文献

Ligand exchange reactions between phenanthrene or 9,10-dimethylphenanthrene with ferrocene effected in the presence of AlCl3-Al were carried out under a variety of conditions. With phenanthrene (I), hydrogenation at the C-9 and C-10 positions could take place during the reaction and the cationic products obtained were the η6-phenanthrene-η5-cyclopentadienyliron and η6-9,10-dihydro-phenanthrene-η5-cyclopentadienyliron moncations (II and III), and the η6-phenanthrene-trans-bis5-cyclopentadienyliron and η6-9,10-dihydrophenanthrene-trans-bis5-cyclopentadienyliron dications (IV and V). With 9,10-dimethylphenanthrene (VI), reactions carried out in refluxing cyclohexane gave the non-hydrogenated η6-9,10-dimethylphenanthrene-η5-cyclopentadienyliron monocation (VII) and η6-9,10-dimethylphenanthrene-trans-bis5-cyclopentadienyliron dication (VIII). When higher temperatures were used in an attempt to promote hydrogenation, decomposition predominated and no cationic product could be obtained. These finding are discussed and contrasted with previous results obtained from similar reactions using anthracene or 9,10-dimethylanthracene.  相似文献

2,5-Dihydro-5-oxo-1,2,4-triazine and some of its alkylated derivatives have been prepared. Nmr spectroscopic analysis has established that the 2,5-dihydro-5-oxo tautomers are preferred over the 4,5-dihydro-5-oxo ones. This preference, and the behavior of 1,2,4-triazines in some other chemical reactions has been interpreted in terms of electron-electron repulsions between the lone pairs of electrons of N1 and N2 in this ring system.  相似文献

A highly sensitive and selective catalytic adsorptive cathodic striping procedure for the determination of trace germanium is presented. The method is based on adsorptive accumulation of the Ge(IV)‐gallic acid (GA) complex onto a hanging mercury drop electrode, followed by reduction of the adsorbed species. The reduction current is enhanced catalytically by addition of vanadium(IV)‐EDTA. The optimal experimental conditions include the use of 0.03 mol/L HClO4 (pH1.6), 6.0×10?3 mol/L GA, 3.0×10?3 mol/L V(IV), 4.0×10?3 mol/L EDTA, an accumulation potential of ?0.10 V(vs. Ag/AgCl), an accumulation time of 120 s and a differential pulse potential scan mode. The peak current is proportional to the concentration of Ge(IV) over the range of 3.0×10?11 to 1.0×10?8 mol/L and the detection limit is 2×10?11 mol/L for a 120 s adsorption time. The relative standard deviation at 5.0×10?10 mol/L level is 3.1%. No serious interferences were found. The method was applied to the determination of germanium in ore, mineral water and vegetable samples with satisfactory results.  相似文献

The effect of conformational preference on the chiroptical properties of α-substituted succinic acids and esters 2 has been investigated using variable-temperature CD in solvents of different polarity. All available evidence indicates that when R' in 2 is alkyl or halogen, the preferred conformation is 1e with the -CH2COOR group eclipsed by the CO bond, while when R' in 2 is hydroxy, methoxy or amino, the major conformer is 1d in which the heteroatom is eclipsed by the CO bond. In both cases, conformation 1f appears to be the least favored, mainly on steric grounds. The results when R' = chloro or bromo do not support previously proposed conclusions and an alternative explanation is advanced. In the resultant octant projection (Fig, 2) it is shown that the sign of the Cotton effect for 1 will be determined by the position of the groups X and Y in the back octants, and since one of these is always H in the two favored conformations, the sign is actually determined by the position of the other group. This octant projection predicts successfully the sign of the ellipticity of the n→π* transition for any α-substituted succinic acid or ester with the appropriate substituent R', and appears to apply also to simple alkanoic acids and esters with the same substituents.  相似文献

A new diamine monomer containing flexible sulfone, sulfide, and amide units was prepared via three steps. Nucleophilic chloro displacement reaction of 4‐aminothiophenol with 4‐nitrobenzoyl chloride in the presence of propylene oxide afforded N‐(4‐mercapto‐phenyl)‐4‐nitrobenzamide and subsequent reduction of the nitro intermediate led to 4‐amino‐N‐(4‐mercapto‐phenyl)benzamide. Two moles of this amino thiophenol compound was reacted with bis‐(4‐chloro phenyl)sulfone to provide a novel diamine monomer. The diamine was reacted with aromatic dianhydrides to form polyimides via a two‐step polycondensation method, formation of poly(amic acid)s, followed by chemical imidization. The resulting polymers were characterized and their physical properties including thermal behavior, thermal stability, solubility and inherent viscosity were studied. Homologous recombination (HR) repair deficiency impairs the proper maintenance of genomic stability, thus rendering cancer cells vulnerable to loss or inhibition of DNA repair proteins, such as poly(ADP-ribose) polymerase-1 (PARP-1). Inhibitors of nuclear PARPs are effective therapeutics for a number of different types of cancers. Here we review key concepts and current progress on the therapeutic use of PARP inhibitors (PARPi). PARPi selectively induce synthetic lethality in cancer cells with homologous recombination deficiencies (HRDs), the most notable being cancer cells harboring mutations in the BRCA1 and BRCA2 genes. Recent clinical evidence, however, shows that PARPi can be effective as cancer therapeutics regardless of BRCA1/2 or HRD status, suggesting that a broader population of patients might benefit from PARPi therapy. Currently, four PARPi have been approved by the Food and Drug Administration (FDA) for the treatment of advanced ovarian and breast cancer with deleterious BRCA mutations. Although PARPi have been shown to improve progression-free survival, cancer cells inevitably develop resistance, which poses a significant obstacle to the prolonged use of PARP inhibitors. For example, somatic BRCA1/2 reversion mutations are often identified in patients with BRCA1/2-mutated cancers after treatment with platinum-based therapy, causing restoration of HR capacity and thus conferring PARPi resistance. Accordingly, PARPi have been studied in combination with other targeted therapies to overcome PARPi resistance, enhance PARPi efficacy, and sensitize tumors to PARP inhibition. Moreover, multiple clinical trials are now actively underway to evaluate novel combinations of PARPi with other anticancer therapies for the treatment of PARPi-resistant cancer. In this review, we highlight the mechanisms of action of PARP inhibitors with or without BRCA1/2 defects and provide an overview of the ongoing clinical trials of PARPi. A single-crystal neutron diffraction analysis of the cluster complex [H(4)Co(4)(C(5)Me(4)Et)(4)] was carried out on the new quasi-Laue diffractometer VIVALDI at the Institut Laue-Langevin. The structure consists of four face-bridging hydrides attached to a tetrahedral cobalt metal core. Average distances and angles in the core of the molecule are as follows: Co-Co = 2.571(8), Co-C = 2.158(6), Co-H = 1.749(7), H.H = 2.366(9) A; Co-H-Co = 94.6(3), H-Co-H = 85.1(3) degrees. The hydride ligands are located off the Co-Co-Co planes by an average distance of 0.923(8) A. It is suggested that the dimensions of the HCo(3) fragments found in this molecule provide reasonable estimates for analogous distances and angles associated with chemisorbed H atoms situated on the 3-fold hollows of a cobalt surface. Crystallographic details: space group P2(1)/a (monoclinic); a = 21.979(2), b = 10.924(1), c = 34.406(2) A; beta = 90.81(1) degrees; Z = 8. Final agreement factor: R(F) = 0.099 for 3779 reflections [I > 2sigma(I)] collected at 20 K.  相似文献

A theoretical study at the Hartree–Fock and density functional theory levels is performed on sulfonamide‐type bacteriostatic compounds with the aim to provide an insight into their structure–activity relationship. The basicity of the p‐amino group is analyzed by means of the proton affinities and the protonation energies, showing that molecules presenting bacteriostatic activity are less basic, i.e., they are characterized by larger protonation energies and smaller proton affinities. The acidity of the amide group is analyzed through the deprotonation energy. The results reveal that the more acidic molecules present a larger bacteriostatic activity. This result is also confirmed from a study of bond orders. A bond order analysis of the amide group suggests that the electron attracting group in these molecules is responsible for the increase in acidity. The charge of the SO2 group is also shown to be affected by the presence of the electron attracting group and consequently related to the acidity of the molecules. A geometric analysis shows that structures in which the amino group is more coplanar with respect to the benzenic ring possess larger bacteriostatic activity. A conformational analysis of these molecules illustrates that active molecules have relatively larger torsion energy barriers. © 2003 Wiley Periodicals, Inc.
