Synthesis of NSC-341,964: An unexpected study on the stability of 2-aryl-4H-3,1-benzoxazin-4-ones

Resynthesis of NSC 341,964, which had been assigned structure 1 (1-[[3-(7-chloro-4-oxo-4H-3,1-benzoxazin-2-yl)phenyl]methyl]pyridinium chloride) was approached via 7-chloro-2-(3-methylphenyl)-4H-3,1-benzoxazin-4-one ( 5 ) obtained from 3-methylbenzoyl chloride ( 2 ) and 2-amino-4-chlorobenzoic acid ( 3 ) followed by dehydration in acetic anhydride. Radical bromination provided 6 which with pyridine afforded the bromide analog 7 of 1 . Ion exchange, however, gave ring-opened benzoic acid 8 rather than 1 . The original sample of NSC 341,964 also proved to be ring-opened material. However, 7 upon standing exhibited slow hydrolysis to 8 so that the structure of the original NSC 341,964 remains uncertain. A more direct route to compound 8 is also described.     

The isolation and structure determination of cryptomisrine,a novel indolo[3,2-b]quinoline dimeric alkaloid from cryptolepis sanguinolenta

The isolation and structure determination of cryptomisrine, a novel indolo[3,2-b]quinoline dimeric alkaloid obtained from extracts of the roots of the Ghanaian medicinal plant Cryptolepis sanguinolenta is reported. The structure determination was made via a consideration of the spectral data, including uv, ir, nmr, and mass spectra. In particular, one-dimensional proton/carbon nmr, one-dimensional nOe difference nmr, and a series of homonuclear (COSY) and inverse-detected heteronuclear two-dimensional (HMQC, HMBC) experiments were utilized, as well as high resolution FABMS. Cryptomisrine is most unusual in that its two monomeric parts apparently exist in such a C₂ symmetric environment that only one set of proton and carbon nmr resonances are observed. Cryptomisrine is the first example of a dimeric indolo-[3,2-b]quinoline alkaloid to have been isolated from nature.     

Synthesis and biological activity of novel epothilone aziridines

[reaction: see text]. A series of 12alpha,13alpha-aziridinyl epothilone derivatives were synthesized in an efficient manner from epothilone A. The final semisynthetic route involves a formal double-inversion of stereochemistry at both the C12 and C13 positions. All aziridine analogues were tested for effects on tubulin binding polymerization and cytotoxicity. The results indicate that the aziridine moiety is a viable isosteric replacement for the epoxide in the case of epothilones.     

Reactions of triosmium clusters with organic azides; x-ray crystal structures of [Os3(CO)10(NCMe)(N3COPh)], [Os3(μ-H)(CO)10(HN3Ph)], and [Os3(μ-H)2(CO)9(μ3-NPh)]

Organic azides [N₃R] react with [Os₃(CO)₁₁(NCMe)] and with [Os₃(μ-H)₂(CO)₁₀] to form [Os₃(CO)₁₀(NCMe)(N₃COR)] (R  Ph) and [Os₃(μ-H)(CO)₁₀(HN₃R)] (R  Ph, n-Bu, CH₂Ph, cyclo-C₆H₁₁), respectively; the latter may be converted to [Os₃(μ-H)₂(CO)₉(μ₃-NR)] by thermolysis; the molecular structure of the phenyl derivative of each class of compound has been confirmed by x-ray analysis.     

Molecular and crystal structures of ruthenium and osmium arene clusters

The molecular and crystal structures of a number of ruthenium and osmium clusters of nuclearity between three and six containing arene fragments such as C₆H₆, C₆H₃Me₃, C₆H₄Me₂ and C₆H₅Me have been investigated. Attention has been focused on the relationship between the terminal ( ⁶-coordination) and face-capping ( ₃: ²: ²: ²-coordination) bonding modes. Empirical packing potential energy calculations have been employed to investigate the intermolecular organization in the crystal. It has been shown that the arene fragments in mono-arene clusters form ribbons, while in bis-arene clusters graphitic-like interactions throughout the crystal are established. The factors controlling the ease of arene reorientational motion in the solid state has also been investigated in relation to the shape, size and geometry of the molecules and of their interlocking modes.     

Interactions of proteins with uniform colloidal hematite and chromium hydroxide particles. I. Adsorption

The adsorption of ovalbumin, -globulin, and lysozyme on uniform spherical hematite and chromium hydroxide particles in aqueous media has been studied as a function of the pH at a constant ionic strength. The uptake of ovalbumin and -globulin was greatest at their isoelectric points and differed little at 10^–2 and 10^–3 mol dm^–3 NaNO₃. The adsorption of lysozyme was strongly influenced by the ionic strength.The deposition of ovalbumin on hematite in the presence of Mg (NO₃)₂ was significantly greater than that with NaNO₃ under otherwise comparable conditions. Dialysis experiments with ovalbumin against magnesium nitrate solutions showed Mg²⁺ to be specifically bound to the protein.The shapes of isotherms indicated monolayer coverage for ovalbumin and multilayer coating for lysozyme for both adsorbents. The shapes of isotherms of -globulin on hematite point to a rearrangement of the protein on the particle surface, while a monolayer was found on chromium hydroxide particles.Supported by the NSF Grant CHE-9108420Part of a Ph.D. thesis

Symmetric nonnegative realizability via partitioned majorization

A sufficient condition for symmetric nonnegative realizability of a spectrum is given in terms of (weak) majorization of a partition of the negative eigenvalues by a selection of the positive eigenvalues. If there are more than two positive eigenvalues, an additional condition, besides majorization, is needed on the partition. This generalizes observations of Suleǐmanova and Loewy about the cases of one and two positive eigenvalues, respectively. It may be used to provide insight into realizability of 5-element spectra and beyond.
