A rapid, sensitive and selective ion-pair reversed-phase liquid chromatography-electrospray ionization isotope dilution tandem mass spectrometry (IP-LC-ESI-ID-MS/MS) was developed for quantitative analysis of free intracellular metabolites in cell cultures. As an application a group of compounds involved in penicillin biosynthesis pathway of Penicillium chrysogenum cells, such as penicillin G (PenG), 6-aminopenicillanic acid (6-APA), benzylpenicilloic acid (PIO), ortho-hydroxyphenyl acetic acid (o-OH-PAA), phenylacetic acid (PAA), 6-oxopipeidine-2-carboxylic acid (OPC), 8-hydroxypenicillic acid (8-HPA), L-alpha-(delta-aminoadipyl)-L-alpha-cystenyl-D-alpha-valine (ACV) and isopenicillin N (IPN) were chosen. (13)C-labeled analogs of the metabolites were added to the sample solutions as internal standards (I.S.). Sample mixtures were analyzed without any sample pretreatment. A new column association using comprehensive two-dimensional gas chromatography for the detailed molecular analysis of hydrocarbon mixtures is reported in this paper. In order to compare the impact of two different secondary columns, a novel column combination relying on a GC x 2GC system was used. This system is based on a non-polar first column (PONA) combined with both a permethylated beta-cyclodextrin (beta-Dex 120) stationary phase and a polysilphenylensiloxane (BPX 50) in the second dimension. Compared to BPX 50 stationary phase, the implementation of beta-cyclodextrin columns as the second dimension was found to improve the resolution between paraffins and naphthenes in the naphtha range but not in the middle distillate range. Attempts to improve the results and to understand the interaction mechanism remained unsuccessful. Therefore, the benefits of the beta-Dex 120-column are only demonstrated on heavy naphtha cut for the quantitation of hydrocarbons.  相似文献

We present an application of our recently proposed coupled reference interaction site model (RISM) molecular dynamics (MD) solvation free energy methodology [Freedman and Truong, Chem. Phys. Lett. 381, 362 (2003); J. Chem. Phys. 121, 2187 (2004)] to study the conformational stability of alanine dipeptide in aqueous solution. In this methodology, radial distribution functions obtained from a single MD simulation are substituted into a RISM expression for solvation free energy. Consequently, iterative solution of the RISM equation is not needed. The relative solvation free energies of seven different conformations of the alanine dipeptide in aqueous solution are calculated. Results from the coupled RISM/MD methodology are in good agreement with those from earlier simulations using the accurate free energy perturbation approach, showing that the alphaR conformation is most stabilized by solution. This study establishes a framework for applying this coupled RISM/MD method to larger biological systems.  相似文献

The stability constants of monoligand complexes of copper(II) ions with glycyl-glycyl-glycine zwitterions (triglycine, HL±) and triglycinate ions (L?) in a water–ethanol solvent with 0.0, 0.1, 0.3, and 0.5 mole fractions of ethanol at an ionic strength of 0.1 created by sodium perchlorate and temperature Т = 298.15 K are determined by means of potentiometric titration. It is found that an increase of ethanol content improves the stability of the investigated complexes, due mainly to the resolvation of ligands.  相似文献

PtSn/θ-Al2O3 catalysts with different amounts of K (0.14, 0.22, 0.49, 0.72, and 0.96 wt%) are prepared to investigate the K effects on the PtSn catalyst in propane dehydrogenation (PDH). KPtSn catalyst with 0.xx wt% K, 0.5 wt% Pt and 0.75 wt% Sn is designated as xx-KPtSn. PDH was performed at 873 K and a gas hourly space velocity (GHSV) of 53,000 mL/gcat h. The temperature-programmed desorption (NH3-TPD), temperature-programmed reduction (TPR) and CO chemisorption of the KPtSn catalysts with K added revealed the potassium addition blocked the acid sites, promoted the reduction of Sn oxide and decreased the Pt dispersion. The formations of cracking products and higher hydrocarbons on acid sites were suppressed by the K effect of blocking the acid sites. In contrast, K addition at more than 0.72 wt% rather increased cracking products and the amount of coke, resulting in the severe deactivation of catalysts. The high cracking products on the KPtSn catalysts with the high amount of K should not be related to the acid sites, because the acid sites were monotonously decreased with an increase in the amount of K. Instead, the potassium affected the characteristics of PtSn. The interaction between Pt and Sn could be weakened by enriching the reduced Sn, because the K component promoted the reduction of Sn oxide in the TPR experiments. Therefore, the 14-KPtSn catalyst with the low amount of K exhibits the highest stability and selectivity among the prepared KPtSn catalysts due to the compromise of the advantageous (blocking the acid sites) and bad (weakening the interaction between Pt and Sn) effects of the K addition in PDH.  相似文献

Rod-like cellulose nanowhiskers and spherical cellulose nanoparticles were prepared from wood-pulp-derived cellulose powder by mechanical refining processes such as high-pressure homogenization (HPH) and ball-milling (BM). The nanowhiskers obtained by the HPH method were found to be 200–500 nm long and 11–16 nm wide. The diameters of the nanoparticles were in the range 40–200 nm, depending on the BM time, and were reduced to 25–50 nm after extra HPH. By adjusting the BM time, cellulose nanoparticles having different polymorphs with similar morphologies were prepared. The X-ray diffraction patterns revealed the recrystallization of cellulose I (1 h of BM time) or cellulose II (4–8 h of BM time) in ball-milled nanoparticles after water washing and solvent exchange treatments. The nanowhisker widths derived from the specific surface areas (SSA) by adsorption methods such as Congo red dye, nitrogen, and water vapor, sorptions were in agreement with those obtained from transmission electron microscopy and atomic force microscopy images. Similar SSA values were obtained for micro- and nano-scale cellulose materials using water vapor adsorption methods, and the SSAs of nanoparticles obtained by different adsorption methods are also discussed.  相似文献

An ‘on-water’, efficient, high yielding, expeditious method has been developed for the synthesis of 1,4-dihydropyridine (1,4-DHP) derivatives via an one-pot multi-component condensation of dimedone or 4-hydroxycoumarine, aldehydes, and ammonium acetate using Fe3O4@SiO2 nanoparticles as a recyclable heterogeneous catalyst. This method takes advantage of the fact that water, a green solvent is used in combination with Fe3O4@SiO2 nanoparticles as catalyst which can be easily recovered magnetically and reused for further runs.  相似文献

Two methods for automated analysis of extracts from edible muscle tissue of Atlantic salmon are described. Oxolinic acid and flumequine are extracted with phosphate buffer pH 9, and the extracts are analysed by high-performance liquid chromatography using a column-switching system. One method applies on-line concentration and clean-up of the extracts on a precolumn packed with polystyrene-divinylbenzene. This method was useful for the analysis of oxolinic acid and flumequine in the microgram/g range. The other method was based on on-line dialysis and concentration of the dialysate on the polymeric precolumn. This method was shown to be a reliable method for residue analysis, and the limit of detection was 2 ng/g for oxolinic acid and 3 ng/g for flumequine with fluorescence detection.  相似文献

Amines are converted in good to excellent yields to phenylsulfides or phenyl-selenides via nucleophilic displacement on the corresponding ditosylamides. The conversion of (?)-(S)-1-phenylethylamine to the chiral phenylsulfide 6 was found to proceed with inversion of configuration. A simple one-pot procedure for the preparation of ditosylamides is reported.  相似文献

This article reports on the peak shape analysis of X‐ray photoelectron spectra of gold‐silver core‐shell (Au@Ag) nanoparticles (NPs) using an asymmetric Gaussian–Lorentzian mixed function. Unlike Ag NPs, Au@Ag NPs have no oxide peak and show asymmetric line shape with a high energy tail in Ag 3d core‐level spectra. A monotonic increase in the Ag 3d binding energy and a decrease in the degree of asymmetry with increasing the Ag shell thickness were observed supporting the occurrence of charge transfer from Au core to Ag shell.
